Mass wasting at the base of the South central Chilean continental margin: the Reloca Slide
Offshore south central Chile (35° S–42° S), the morphology of the lowermost continental slope and trench floor witnesses a voluminous submarine mass-wasting event. The blocky slide body deposited in the Chile Trench at 73°46´ W 35°35´ S was targeted for study during RRS JAMES COOK Cruise JC23 and termed Reloca Slide. Its size of about 24 km3, its steep and high headscarp, the spatial distribution of slide deposits and the cohesive nature of major slide blocks make it interesting to address the issue of tsunami generation. We have obtained seismic reflection data that partly reveal the internal structure of the slide body. Gravity core samples were retrieved that will allow the slide to be dated and linked to the history of sedimentation and slope stability along this particular segment of the Chilean convergent margin. At present we assume a Holocene age for the sliding event

Die Ablaich- und Interstitialphase der Ă„sche (Thymallus thymallus L.) - Grundlagen und Auswirkungen anthropogener Belastungen -
Die vorliegende Arbeit beschäftigt sich mit den
Grundlagen der Ablaich- und Interstitialphase der Ă„sche (
Thymallus thymallus) und den Auswirkungen anthropogener
Gewässerveränderungen auf die frühen Entwicklungsstadien. In
einem deutschen Mittelgebiergsfluss, der Lahn, wurde das
Ablaichverhalten der Ă„sche beobachtet und die Anzahl und
Struktur verschiedener Äschenlaichplätze aufgenommen. Das
Laichsubstrat wurde erstmalig mit standardisierten Siebsätzen
(0,063-63mm) ausgesiebt und das Larvenaufkommen von 1995-1999
dokumentiert. Mithilfe von Schlauchsystemen mit verschiedenen
Tiefenhorizonten (10, 20 und 30cm Sedimentiefe)konnten die
Milieubedingungen (O2, CO2, NO3-, NO2-, NH4+, NH3, o-PO4-P,
pH-Wert, Temp.,Leitfähigkeit und SBV)im Interstitial an
natürlichen Laichplätzen erfasst werden. Zusätzlich wurden
stoffliche Belastungsszenarien durchgefĂĽhrt, bei denen
Kiesboxen mit Ă„scheneiern und Schlauchsysteme in
Gewässerabschnitten mit unterschiedlicher stofflicher Belastung
(vor und nach der Ausleitungsstrecke einer Kläranlage)
vergraben wurden. Die Boxen wurden zu drei verschiedenen
Entwicklungszeitpunkten (Augenpunktstadium, Schlupf und
Aufschwimmen) geborgen. Die Mortalität der Eier und Larven
wurde notiert und die verbliebenen Tiere unter dem Binokular
vermessen. Durch die zeitgleiche Onlinemessung der
chemisch-physikalischen Parameter in der freien Welle konnte
die Tagesperiodik der o.g. Parameter dokumentiert und ihr
Verlauf im Interstitial modelliert werden. In der Lahn wandern
die Äschen Anfang April an die Laichplätze, wo die Männchen
langgestreckte Reviere von 4,5 bis 8 m² verteidigen. Der
Ablaichvorgang begann nachmittags ab einer Wassertemp. von 8°C.
Zum Ablaichen wurden überströmte Flachwasserzonen (Mittelwerte:
Oberflächenströmung: 61 cm/s, Grundströmung 34 cm/s,
Wassertiefe 41 cm) genutzt. Zwischen den einzelnen
Ablaichvorgängen waren die Äschen in Ruhehabitaten (ufernahe
Kolke mit Ă„sten und Totholz bedeckt) zu finden. Das
Laichsubstrat war ĂĽberwiegend kiesig. Es bestand ĂĽberwiegend
aus Grobkies (Korngöße 20-63 mm: 30-50%), einigen Steinen
(>63 mm: 3-35%) und Mittelkies (6,3-20 mm: 14-30%). Der
Feinsedimentanteil (< 2 mm) war an allen drei untersuchten
Flussabschnitten gering (5,3-12,3%). Die Dauer der
Embryonalentwicklung war temperaturabhängig und betrug in der
Lahn durchschnittl. 156,3 Tagesgrade (16,9 Tage bei 9,25°C).
Die limitiernden Faktoren der Ei- und Embryonalentwicklung der
Ă„sche an der Lahn waren die Ammoniakkonzentrationen und der
Feinsedimentanteil im Interstitial. Im stofflich stark
belasteten Versuchsfeld traten Ammoniakkonzentrationen bis 108
µg/l NH3-N vor dem Schlupf auf. In der sensiblen Phase nach dem
Schlupf wurden NH3-N Werte bis 52 µg/l gemessen, die zum
Absterben aller Ă„scheneier fĂĽhrte. Als Grenzwert fĂĽr die
Larvalentwicklung wird eine NH3-N Konzentration von 25 µg/l
diskutiert. An den natürlichen Laichplätzen korrelierte die
Anzahl aufschwimmender Ă„schenlarven signifikant negativ mit dem
Feinsedimentanteil (Korngröße < 2 mm). Der höchste
Feinsedimentanteil von 23,7% trat nach Ausbelieben des
Winterhochwassers im Jahr 1996 auf und fĂĽhrte zu einer
Halbierung des Ă„schenlarvenaufkommens. Der pH-Wert der freien
Welle zeigte ausgeprägte tagesperiodische Schwankungen
(7,3-9,9) mit Maximalwerten in den Nachmittagsstunden. An den
natürlichen Laichplätzen lag der pH-Wert mit 8-8,9 in 10 cm
Sedimenttiefe im alkalischen Bereich. Die
Sauerstoffkonzentration lag tagsüber immer im übersättigten
Bereich (11,7 - 15 mg/l in 10 cm Sedimenttiefe, 11,2 - 16,5
mg/l in der freien Welle). Nachts sanken die O2-Werte in der
freien Welle bis 5,9 mg/l. Sowohl die hohen pH-Werte als auch
die hohen Sauerstoffkonzentrationen am Tage warenb auf die
erhöhte Photosyntheserate der Algen im eutrophierten Gewässer
zurĂĽckzufĂĽhren
Human endothelial-like differentiated precursor cells maintain their endothelial characteristics when cocultured with mesenchymal stem cell and seeded onto human cancellous bone
Introduction. Cancellous bone is frequently used for filling bone defects in a clinical setting. It provides favourable conditions for regenerative cells such as MSC and early EPC. The combination of MSC and EPC results in superior bone healing in experimental bone healing models. Materials and Methods. We investigated the influence of osteogenic culture conditions on the endothelial properties of early EPC and the osteogenic properties of MSC when cocultured on cancellous bone. Additionally, cell adhesion, metabolic activity, and differentiation were assessed 2, 6, and 10 days after seeding.
Results. The number of adhering EPC and MSC decreased over time; however the cells remained metabolically active over the 10-day measurement period. In spite of a decline of lineage specific markers, cells maintained their differentiation to a reduced level. Osteogenic stimulation of EPC caused a decline but not abolishment of endothelial characteristics and did not induce osteogenic gene expression. Osteogenic stimulation of MSC significantly increased their metabolic activity whereas collagen-1α and alkaline phosphatase gene expressions declined. When cocultured with EPC, MSC’s collagen-1α gene expression increased significantly. Conclusion. EPC and MSC can be cocultured in vitro on cancellous bone under osteogenic conditions, and coculturing EPC with MSC stabilizes the latter’s collagen-1α gene expression
Constitutive upregulation of the transforming growth factor-β pathway in rheumatoid arthritis synovial fibroblasts
Genome-wide gene expression was comparatively investigated in early-passage rheumatoid arthritis (RA) and osteoarthritis (OA) synovial fibroblasts (SFBs; n = 6 each) using oligonucleotide microarrays; mRNA/protein data were validated by quantitative PCR (qPCR) and western blotting and immunohistochemistry, respectively. Gene set enrichment analysis (GSEA) of the microarray data suggested constitutive upregulation of components of the transforming growth factor (TGF)-β pathway in RA SFBs, with 2 hits in the top 30 regulated pathways. The growth factor TGF-β1, its receptor TGFBR1, the TGF-β binding proteins LTBP1/2, the TGF-β-releasing thrombospondin 1 (THBS1), the negative effector SkiL, and the smad-associated molecule SARA were upregulated in RA SFBs compared to OA SFBs, whereas TGF-β2 was downregulated. Upregulation of TGF-β1 and THBS1 mRNA (both positively correlated with clinical markers of disease activity/severity) and downregulation of TGF-β2 mRNA in RA SFBs were confirmed by qPCR. TGFBR1 mRNA (only numerically upregulated in RA SFBs) and SkiL mRNA were not differentially expressed. At the protein level, TGF-β1 showed a slightly higher expression, and the signal-transducing TGFBR1 and the TGF-β-activating THBS1 a significantly higher expression in RA SFBs than in OA SFBs. Consistent with the upregulated TGF-β pathway in RA SFBs, stimulation with TGF-β1 resulted in a significantly enhanced expression of matrix-metalloproteinase (MMP)-11 mRNA and protein in RA SFBs, but not in OA SFBs. In conclusion, RA SFBs show broad, constitutive alterations of the TGF-β pathway. The abundance of TGF-β, in conjunction with an augmented mRNA and/or protein expression of TGF-β-releasing THBS1 and TGFBR1, suggests a pathogenetic role of TGF-β-induced effects on SFBs in RA, for example, the augmentation of MMP-mediated matrix degradation/remodeling

Beschleunigte Knochenheilung durch frĂĽhe Vaskularisierung mittels Implantation von EPC und MSC auf beta-TCP im Knochendefektmodell der Ratte
Fragestellung: In einem ausgedehnten Knochendefekt kann das Einwachsen von knochenbildenden Zellen limitiert sein, da ohne Gefässe die Ernährung der regenerativen Zellen im Knochenkonstrukt insuffizient ist. Endotheliale Progenitorzellen (EPC) sind wichtig bei der Neovaskularisierung. Die frühe Vaskularisierung von grossen Knochendefekten kann für das Überleben und die Funktion von mesenchymalen Stammzellen (MSC) und knochenbildenden Zellen entscheidend sein. Kann die Implantation von EPC und MSC auf osteokonduktiven beta-Tricalciumphosphat (beta-TCP) in einem "critical-size" Knochendefekt des Femur von athymischen Ratten die frühe Vaskularisierung und die Knochenheilung in vivo verbessern?
Methodik: Humane EPC wurden aus Buffy-Coat und humane MSC aus Knochenmarkaspirat durch Dichtezentrifugation isoliert. 2.5 x 105 kultivierte und differenzierte EPC und MSC wurden in vitro auf beta-TCP geladen. In 145 athymischen, männlichen Ratten wurde das Femur osteotomiert, ein 5 mm Knochendefekt erzeugt und mit Fixateur externe stabilisiert. Die Knochendefekte wurden mit beta-TCP (Gruppe 1), beta-TCP und MSC (Gruppe 2), beta-TCP und EPC (Gruppe 3), beta-TCP und EPC und MSC (Gruppe 4) oder autologem Knochen (Gruppe 5) gefüllt. Nach 1 Woche (n=40), 4 Wochen (n=40), 8 Wochen (n=40) und 12 Wochen (n=25) wurden die Ratten getötet. Bei Pinlockerung wurde die Ratte ausgeschlossen. Die (immun)histologische Analyse (Färbung mit HE, VEGF-R2, vWF) der Vaskularisierung und Knochenneubildung erfolgte mit Image-Analysis-System. Nach 8 und 12 Wochen erfolgte ein µCT und ein 4-Punkte-Biegungstest. Für die statistische Analyse wurde der Kruskal-Wallis-Test verwendet.
Ergebnisse und Schlussfolgerungen: Nach 1 Woche zeigte sich bei der Implantation von EPC/MSC und EPC allein signifikant mehr primitive vaskuläre Plexus (p=0.01;p=0.048) als in Vergleichsgruppen. Im Vergleich zur TCP Gruppe war in allen anderen Versuchsgruppen signifikant mehr Knochenneubildung zu sehen (p<0.01). Ausserdem war in der EPC/MSC-Gruppe signifikant mehr Knochenbildung zu erkennen als in der MSC-Gruppe (p=0.03). Nach 12 Wochen zeigten alle Gruppen eine knöcherne Durchbauung des Defektes, jedoch zeigten bereits 8 Wochen nach Implantation von MSC/EPC 83% der Defekte eine stabile, knöcherne Durchbauung. Bei der Implantation von MSC kam es in 18% der Knochendefekte zum knöchernen Durchbau. Alle anderen experimentellen Gruppen zeigten nach 8 Wochen keine knöcherne Durchbauung. Diese Resultate konnten im µCT, biomechanischen Test und in der Histologie quantifiziert werden. EPC scheinen die frühe Vaskularisierung innerhalb eines Knochenkonstrukt in vivo zu stimulieren und das Einwachsverhalten von MSC zu verbessern, was zu einer beschleunigten Knochenheilung im Knochendefektmodell der Ratte führt
Measures against heat stress in the city of Gelsenkirchen, Germany
In the near-surface atmosphere, heat waves during the summer cause situations that may lead to human-biometeo-rological impairment. Because of their high population density, overheated cities are particularly strongly affected by heat stress. In the future, due to the effects of climate change, heat stress will increase in terms of its intensity and spatial expansion in the areas of concern. Taking the example of the city of Gelsenkirchen, Germany, this article presents a method for the identification of areas requiring adaptation or protection. A scenario of the future increase of heat stress events is presented, based on data of the German climate change model STAR II. For the identification of areas requiring adaptation and protection, spatial analyses of the urban heat island, land use and demographic aspects were performed using GIS tools. The application and assessment of adaptation measures is investigated for an urban quarter using the microscale numerical model ENVI-met. Finally adaptation measures in urban planning against heat stress are discussed. The relevant urban planning adaptation measures, which are also important in view of climate change, not only involve heat stress reduction in the residential areas already affected, but also in-volve the protection and optimisation of existing favourable and compensation areas
Nano-casted N-Doped Carbon Created From a Task-Specific Protic Salt and Controlled Porous Glass
3-dimensionally interconnected macroporous carbons are versatile materials that can be used in catalysis, electrochemical devices, and separation technology. Herein, the synthesis of a nitrogen doped carbonaceous material with a well-defined nanoarchitecture via nano-casting is demonstrated. A novel carbon source, a task-specific protic salt, has been proposed to create nitrogen doped carbon by direct carbonization within the pores of controlled macroporous glass. After the removal of macroporous glass from the composite using an aqueous sodium hydroxide solution and upon further heat treatment, an oxidation resistant doped carbon with high nitrogen content (6 mass %) is obtained. The materials formed during the different stages of the nano-casting process exhibit interesting properties such as hierarchical porosity, very high nitrogen content (15 mass %), and increased oxidational stability. A combination of different properties to create tailor-made materials for different applications using this technique is possible
