BackgroundThe aim of this study is to develop a method for optimal in vitro proliferation and differentiation of rat adipocyte. MethodsPreadipocyte were isolated from omentum of Rattus norvegicus Wistar using Collagenase type I dan II (Sigma). Cells were cultured in M199 culture containing either 0%, 8% or 10% Fetal Bovine Serum (FBS). Insulin were added into the media once the cells attached to the culture plate Proliferating and differentiated cells were counted and analysed by using Oil Red O dan Hematoxylen (HE) staining. Results This study demonstrated that adipocyte could be isolated using Collagenase type I but not type II. The addition of 10% FBSsignificantly increased the number of preadipocyte and differentiation of adipocyte more than those of 8% FBS and without FBS. The timing of FBS addition was best performed on day 8 using 10% FBS. Specific adipocyte staining using Oil Red O revealed thatthere were core lipids in mature adipocyte. ConclusionsCollagenase tipe I could be used to isolate preadipocyte cells. Supplementation of culture media with 8-10% FBS could enhance the in vitro proliferation of preadipocyte. Standard media M199 containing 10% FBS and insulin may provide an environment to differentiate preadipocyte specifically into adipocyte in vitro

The effect of Ramadan fasting on some blood parameters i.e. serum glucose, urea, uric acid, lipids and proteins, were investigated on young men in Ar Rohmah Islamic dormitory. Nineteen normal and healthy students aging between 12-25 years, residing in the Islamic dormitory, voluntarily to participated in the study. Blood samples were obtained from the volunteers on the 1st and 26th day of Ramadan and analyzed for the aforementioned biochemical parameters. A non-significant effect of Ramadan fasting was observed on most of the parameters studied. However, serum urea, triglycerides, total cholesterol and LDL-cholesterol were reduced significantly (p0.05) but remained within the physiological limits. Decrease in blood urea has been attributed to the effect of at least protein and triglycerides intake to increase lipolytic effect. The reduction in serum cholesterol and LDL is a beneficial effect of Ramadan fasting. The results of the study indicated thatRamadan fasting is quite safe for normal healthy adults

Modifying Effect of Anthocyanin from Purple Sweet Potatoes on Visceral Fat Tissue Inflammation and Liver Oxidative Stress in Psychological Stress-Induced Mice

Psychological stress generates inflammation and oxidative stress in various tissues. Visceral fat as the target site of inflammation is then correlated with stress-induced obesity. Redox imbalance following inflammation also has prominent impacts on hepatic tissue. Therefore, the development of anti-inflammatory and antioxidant properties from plant bioactive compounds is necessary to be investigated. Anthocyanin (ANC) from various plants is previously described as a powerful anti-inflammatory and antioxidant even though its effect in psychological stress remains underexplored. Purple sweet potatoes (PSP) has ANC as its natural pigment. Previous studies revealed the potential effect of ANC from PSP on behavior and an antidepressant candidate. Thus, this study was purposed to determine the effect of ANC from PSP on inflammation and oxidative stress in visceral fat and hepatic tissue respectively. A total of 25 adult male BALB/c mice were assigned into groups of control, stress, stress+ANC 10 mg/kgBW, stress+ANC 20 mg/kgBW and stress+ANC 40 mg/kgBW. Restraint stress was applied 2 hours/day for 14 days. Enzyme-linked immunoassay (ELISA) was conducted to measure level of IL-6 and IL-10 in visceral fat as well as SOD and MDA from hepatic tissue. The results demonstrated that the supplementation of ANC reduces the level of IL-6 cytokine (p=0.005), tends to increase IL-10 (p=0.612), reduces hepatic SOD (p=0.03), and does not significantly affect the level of hepatic MDA (p=0.432). Both ratios of IL-6/IL-10 and SOD/MDA were reduced following ANC administration. Total ANC extracts are suggested to have a potential role of resisting inflammation and oxidative stress in the psychologically stressed model. Further studies are necessary to evaluate the benefits of ANC from PSP in other peripheral organs under psychological stress exposure

This study aimed to evaluate the effects of ciplukan (Physalis angulata L.) leaf ethanol extract on L-NG-nitroarginine methyl ester (L-NAME)-induced hypertensive rats. We randomly divided twenty-five Wistar rats into five groups. The sham group was given a PBS injection. The hypertensive group was injected with L-NAME on days 1 to 28. Three groups of hypertensive rats were given the extract on days 4 to 28. Blood pressure was measured using the tail-cuff method on days 0, 4, 10, and 27. The endothelial progenitor cells (EPCs) in the blood were measured by flow cytometry as a percentage of circulating angiogenic cells (CACs, CD34+/CD309+/CD45+) and endothelial colony-forming cells (ECFCs, CD34+/CD309+/CD45-). Serum NO and MDA levels, as well as serum SOD activity, were measured colorimetrically. Serum TNF-Ī± levels were measured by the ELISA method. The ciplukan leaf extract reduced systolic and diastolic blood pressure, reduced the percentage of EPCs in the blood, increased serum NO levels, reduced MDA levels, increased serum SOD activity, and reduced serum TNF-Ī± levels in L-NAME-induced hypertensive rats. It is concluded that ciplukan ethanol leaf extract exerts protective effects on L-NAME-induced hypertensive rats. These study results can strengthen the scientific basis of using ciplukan leaf ethanol extract to treat hypertension
