    Reference Transcriptomes of Porcine Peripheral Immune Cells Created Through Bulk and Single-Cell RNA Sequencing

    Pigs are a valuable human biomedical model and an important protein source supporting global food security. The transcriptomes of peripheral blood immune cells in pigs were defined at the bulk cell-type and single cell levels. First, eight cell types were isolated in bulk from peripheral blood mononuclear cells (PBMCs) by cell sorting, representing Myeloid, NK cells and specific populations of T and B-cells. Transcriptomes for each bulk population of cells were generated by RNA-seq with 10,974 expressed genes detected. Pairwise comparisons between cell types revealed specific expression, while enrichment analysis identified 1,885 to 3,591 significantly enriched genes across all 8 cell types. Gene Ontology analysis for the top 25% of significantly enriched genes (SEG) showed high enrichment of biological processes related to the nature of each cell type. Comparison of gene expression indicated highly significant correlations between pig cells and corresponding human PBMC bulk RNA-seq data available in Haemopedia. Second, higher resolution of distinct cell populations was obtained by single-cell RNA-sequencing (scRNA-seq) of PBMC. Seven PBMC samples were partitioned and sequenced that produced 28,810 single cell transcriptomes distributed across 36 clusters and classified into 13 general cell types including plasmacytoid dendritic cells (DC), conventional DCs, monocytes, B-cell, conventional CD4 and CD8 αβ T-cells, NK cells, and γδ T-cells. Signature gene sets from the human Haemopedia data were assessed for relative enrichment in genes expressed in pig cells and integration of pig scRNA-seq with a public human scRNA-seq dataset provided further validation for similarity between human and pig data. The sorted porcine bulk RNAseq dataset informed classification of scRNA-seq PBMC populations; specifically, an integration of the datasets showed that the pig bulk RNAseq data helped define the CD4CD8 double-positive T-cell populations in the scRNA-seq data. Overall, the data provides deep and well-validated transcriptomic data from sorted PBMC populations and the first single-cell transcriptomic data for porcine PBMCs. This resource will be invaluable for annotation of pig genes controlling immunogenetic traits as part of the porcine Functional Annotation of Animal Genomes (FAANG) project, as well as further study of, and development of new reagents for, porcine immunology

    Non-Statistical Effects in Neutron Capture

    There have been many reports of non-statistical effects in neutron-capture measurements. However, reports of deviations of reduced-neutron-width distributions from the expected Porter-Thomas (PT) shape largely have been ignored. Most of these deviations have been reported for odd-A nuclides. Because reliable spin (J) assignments have been absent for most resonances for such nuclides, it is possible that reported deviations from PT might be due to incorrect J assignments. We recently developed a new method for measuring spins of neutron resonances by using the DANCE detector at LANSCE. Measurements made with a 147Sm sample allowed us to determine spins of almost all known resonances below 1 keV. Furthermore, analysis of these data revealed that the reduced-neutron-width distribution was in good agreement with PT for resonances below 350 eV, but in disagreement with PT for resonances between 350 and 700 eV. Our previous (n,alpha) measurements had revealed that the alpha strength function also changes abruptly at this energy. There currently is no known explanation for these two non-statistical effects. Recently, we have developed another new method for determining the spins of neutron resonances. To implement this technique required a small change (to record pulse-height information for coincidence events) to a much simpler apparatus: A pair of C6D6 gamma-ray detectors which we have employed for many years to measure neutron-capture cross sections at ORELA. Measurements with a 95Mo sample revealed that not only does the method work very well for determining spins, but it also makes possible parity assignments. Taken together, these new techniques at LANSCE and ORELA could be very useful for further elucidation of non-statistical effects.Comment: 8 pages, 3 figures, for proceedings of CGS1

    First-principles study of orthorhombic CdTiO3 perovskite

    In this work we perform an ab-initio study of CdTiO3 perovskite in its orthorhombic phase using FLAPW method. Our calculations help to decide between the different cristallographic structures proposed for this perovskite from X-Ray measurements. We compute the electric field gradient tensor (EFG) at Cd site and obtain excellent agreement with available experimental information from a perturbed angular correlation (PAC) experiment. We study EFG under an isotropic change of volume and show that in this case the widely used "point charge model approximation" to determine EFG works quite well.Comment: 4 pages, 1 figure.     Environmental Democratisation : Assessing the Impact of Democratisation on Environmental Capacity in South and Southeastern Europe

    Although a relationship between established democratic political systems and environmental capacity has been described, the impact of the democratisation process on environmental capacity is not clear. The aim of this article is to determine the effect of both prior regime type and mode of transition on environmental capacity, through consideration of the cases of Bulgaria, Portugal, Romania and Spain. In addition, the importance of the European Union (EU) as an external actor shaping environmental capacity building is assessed. Findings indicate that variations in prior regime type and mode of transition had limited impact on environmental capacity development. Of greater importance were the persistent non-democratic legacies that influenced behaviours and actions during the democratisation period. The requirements of the EU were fundamental in ensuring that environmental issues remained on the domestic political agenda in such a context

    Frequency and Nature of Incidental Extra-Enteric Lesions Found on Magnetic Resonance Enterography (MR-E) in Patients with Inflammatory Bowel Diseases (IBD)

    The aim of this study was to determine the occurrence of extra-enteric findings in a large cohort of patients undergoing magnetic resonance enterography (MR-E) and to classify the clinical significance of these findings.We retrospectively analyzed 1154 MR-E performed in 1006 patients referred to our radiological department between 1999-2005. The reasons for referral were suspected or proven inflammatory bowel diseases (IBD) (n = 710), further diagnostic work-up for small bowel disease because of non-specific abdominal symptoms (SBD; n = 182) or suspected small bowel malignancies (SBM; n = 114). All extra-enteric findings were reviewed by a radiologist and a gastroenterologist and were classified as having high, moderate, or low significance for further diagnostic or therapeutic procedures.The average age of all patients was 40+/-16 (Mean+/-SD) years (y) (IBD 35+/-13 y; SBD 49+/-16 y; SBM 57+/-15 y). A total of 1113 extra-enteric findings were detected in 600 of 1006 patients (59.6%). Of these findings 180 (16.2%) were judged as having a high, 212 (19.0%) a moderate and 721 (64.8%) a low significance. On a per group basis in patients with IBD 12.0% of the findings were of major clinical significance compared to 13.7% and 33.3% in patients with SBD and SBM, respectively. The most common major findings were abscesses (69.9%) in the IBD group and extraintestinal tumors, metastases or masses in the SBD and SBM groups (41.9% and 74.2%, respectively).MR-E reveals a substantial number of extra-enteric findings, supporting the role of a cross-sectional imaging method for the evaluation of the small bowel

    Shape abnormalities of the caudate nucleus correlate with poorer gait and balance : results from a subset of the ladis study

    Objective Functional deficits seen in several neurodegenerative disorders have been linked with dysfunction in frontostriatal circuits and with associated shape alterations in striatal structures. The severity of visible white matter hyperintensities (WMHs) on magnetic resonance imaging has been found to correlate with poorer performance on measures of gait and balance. This study aimed to determine whether striatal volume and shape changes were correlated with gait dysfunction. Methods Magnetic resonance imaging scans and clinical gait/balance data (scores from the Short Physical Performance Battery [SPPB]) were sourced from 66 subjects in the previously published LADIS trial, performed in nondisabled individuals older than age 65 years with WMHs at study entry. Data were obtained at study entry and at 3-year follow-up. Caudate nuclei and putamina were manually traced using a previously published method and volumes calculated. The relationships between volume and physical performance on the SPPB were investigated with shape analysis using the spherical harmonic shape description toolkit. Results There was no correlation between the severity of WMHs and striatal volumes. Caudate nuclei volume correlated with performance on the SPPB at baseline but not at follow-up, with subsequent shape analysis showing left caudate changes occurred in areas corresponding to inputs of the dorsolateral prefrontal, premotor, and motor cortex. There was no correlation between putamen volumes and performance on the SPPB. Conclusion Disruption in frontostriatal circuits may play a role in mediating poorer physical performance in individuals with WMHs. Striatal volume and shape changes may be suitable biomarkers for functional changes in this population

    Inward-Looking Policies, Institutions, Autocrats, and Economic Growth in Latin America: An Empirical Exploration

    This paper explores the institutional determinants of economic growth in Latin America by taking advantage of recent empirical research that employs subjective and objective measures to test for a possible Northian explanation that links institutional quality and economic growth. I provide a framework that helps better understand the policymakers` choices and persistence regarding inward-looking policies that were pursued between the 1930s and the 1980s by arguing that in the Latin American case Olson`s (1982) idea of encompassing interest should be expanded to cover not only the economic stakes of power holders, but also, their political stakes, somewhat along the lines of work by Robinson (1997)
