    Navigating the International Legal Terrain for Animal Health and Protection: Specialist Agency or Framework Convention?

    This article identifies and analyses key themes in the history of efforts to make international law an effective instrument for protecting animals and their health, as well as touching upon the positive spillovers this can have for human and environmental health. The pursuit of fragmented and inconsistent approaches has made animal protection a secondary consideration, at best, in international relations. Non-governmental organisations (NGOs) and international non-government organisations (INGOs) have valiantly and persistently argued that there is a  legal ‘gap’ in the protection of animals at an international level, but they have never had a strong  institutional basis from which they could  engage collectively and effectively with state parties. We argue that the adoption of a binding international instrument focused on animal protection would fill this gap and we evaluate one particular recent proposal:  the draft United Nations Convention for Animal Health and Protection, sponsored by Global Animal Law

    Sub-2 cm/s passivation of silicon surfaces by aprotic solutions

    Minimizing recombination at semiconductor surfaces is required for the accurate determination of the bulk carrier lifetime. Proton donors, such as hydrofluoric acid and superacids, are well known to provide highly effective short-term surface passivation. We demonstrate here that aprotic solutions based on bis(trifluoromethanesulfonyl)methane (TFSM) in hexane or pentane can also result in excellent passivation of (100)-orientation silicon surfaces. We show that the optimized TFSM-pentane passivation scheme can measure effective lifetimes up to 20 ms, with a surface recombination velocity of 1.7 cm s1 at an excess carrier density of 1015 cm3 . Fitting injection-dependent lifetime curves requires chemical passivation and field effect passivation from a negatively charged layer with a charge density of 1010–1011 q cm2 . The slightly higher recombination velocity of 2.3 cm s1 measured with TFSM-hexane can be explained by a lower charge density in the passivating layer, suggesting that the steric hindrance associated with the solvent size could play a role in the passivation mechanism. Finally, phosphorus nuclear magnetic resonance experiments confirm that TFSM-based solutions have Lewis acidity without being superacids, which opens up opportunities for them to be used in materials systems sensitive to superacidic environments

    Opioid Use Disorder in the Active Service: Incidence Rates and Behavioral Health Considerations

    Military service members experience occupational specific injuries that often result in chronic pain, and comorbid behavioral health concerns that may be exacerbated by opioid use. Despite the vast amount of research examining substance use in the military, there is a dearth of literature examining the incidence of opioid use disorder (OUD). The present epidemiological study aims to examine the incident rate trends of OUD diagnoses among active duty service members between 2016 and 2021. The present study utilized data drawn from the Defense Medical Epidemiology Database (DMED) to conduct a retrospective cohort study and calculate the incidence rate of OUD (per 10,000) active duty United States service members. The average calculated incidence rate of OUD diagnoses from 2016 to 2021 was 6.71 (per 10,000) and declined by 34% each year on average. Opioid use disorder was most frequently diagnosed among service members who identified as male, married, white, between the ages of 30-39, junior enlisted, and serving in the Army. The incidence rate trends of OUD diagnoses among service members warrants further investigation into the mechanisms contributing to increased risk among identified groups and the development of mitigation strategies that can contribute to a reduction in opioid use and abuse

    Conservation conflict hotspots : mapping impacts, risk perception and tolerance for sustainable conservation management

Conservation conflicts are sometimes labelled human-wildlife conflict, focusing only on the direct negative impact of species (usually wildlife) on humans or vice versa. However, many authors now recognize that conservation conflicts arise between people with diverse views, when one party acts against the interests of another. They are thus human-human conflicts and not merely an impact on or from conservation. Conflict is not always directly correlated with impact because perceptions of risk, levels of tolerance and conservation values influence human responses. This review aims to define the concept of ‘conservation conflict hotspots’ and explore its practical applications in conservation. We propose that the interaction of impact, risk perception, level of tolerance in a context of conservation values can be mapped at a local scale, with spatial visualization assisting the prediction, understanding and management of such hotspots. The term conservation value incorporates measures of indigeneity, endemicity and demography along with emotional or cultural attachment to species or places. The umbrella terms of risk perception and tolerance capture many of the aspects of attitude, values and individual demographics that can influence people’s actions, enabling contextualization of relevant social factors at local scales. Spatially mapped layers enable us to plan and target conservation efforts towards human as well as ecological factors. The concept of ‘conservation conflict hotspot’ emphasizes the need for transdisciplinary research to understand underlying drivers of conflict and for dialogical and peace-building approaches to facilitate trust and cooperation amongst actors.     Response Type and Host Species may be Sufficient to Predict Dose-Response Curve Shape for Adenoviral Vector Vaccines.

    Vaccine dose-response curves can follow both saturating and peaking shapes. Dose-response curves for adenoviral vector vaccines have not been systematically described. In this paper, we explore the dose-response shape of published adenoviral animal and human studies. Where data were informative, dose-response was approximately five times more likely to be peaking than saturating. There was evidence that host species and response type may be sufficient for prediction of dose-response curve shape. Dose-response curve shape prediction could decrease clinical trial costs, accelerating the development of life-saving vaccines

    Immunologic Dose-Response to Adenovirus-Vectored Vaccines in Animals and Humans: A Systematic Review of Dose-Response Studies of Replication Incompetent Adenoviral Vaccine Vectors when Given via an Intramuscular or Subcutaneous Route.

    Optimal vaccine dosing is important to ensure the greatest protection and safety. Analysis of dose-response data, from previous studies, may inform future studies to determine the optimal dose. Implementing more quantitative modelling approaches in vaccine dose finding have been recently suggested to accelerate vaccine development. Adenoviral vectored vaccines are in advanced stage of development for a variety of prophylactic and therapeutic indications, however dose-response has not yet been systematically determined. To further inform adenoviral vectored vaccines dose identification, historical dose-response data should be systematically reviewed. A systematic literature review was conducted to collate and describe the available dose-response studies for adenovirus vectored vaccines. Of 2787 papers identified by Medline search strategy, 35 were found to conform to pre-defined criteria. The majority of studies were in mice or humans and studied adenovirus serotype 5. Dose-response data were available for 12 different immunological responses. The majority of papers evaluated three dose levels, only two evaluated more than five dose levels. The most common dosing range was 107-1010 viral particles in mouse studies and 108-1011 viral particles in human studies. Data were available on adenovirus vaccine dose-response, primarily on adenovirus serotype 5 backbones and in mice and humans. These data could be used for quantitative adenoviral vectored vaccine dose optimisation analysis

    Dynamics of muscle fibre growth during postnatal mouse development

    <p>Abstract</p> <p>Background</p> <p>Postnatal growth in mouse is rapid, with total skeletal muscle mass increasing several-fold in the first few weeks. Muscle growth can be achieved by either an increase in muscle fibre number or an increase in the size of individual myofibres, or a combination of both. Where myofibre hypertrophy during growth requires the addition of new myonuclei, these are supplied by muscle satellite cells, the resident stem cells of skeletal muscle.</p> <p>Results</p> <p>Here, we report on the dynamics of postnatal myofibre growth in the mouse extensor digitorum longus (EDL) muscle, which is essentially composed of fast type II fibres in adult. We found that there was no net gain in myofibre number in the EDL between P7 and P56 (adulthood). However, myofibre cross-sectional area increased by 7.6-fold, and length by 1.9-fold between these ages, resulting in an increase in total myofibre volume of 14.1-fold: showing the extent of myofibre hypertrophy during the postnatal period. To determine how the number of myonuclei changes during this period of intense muscle fibre hypertrophy, we used two complementary mouse models: <it>3F-nlacZ-E </it>mice express <it>nlacZ </it>only in myonuclei, while <it>Myf5</it><sup><it>nlacZ</it>/+ </sup>mice have β-galactosidase activity in satellite cells. There was a ~5-fold increase in myonuclear number per myofibre between P3 and P21. Thus myofibre hypertrophy is initially accompanied by a significant addition of myonuclei. Despite this, the estimated myonuclear domain still doubled between P7 and P21 to 9.2 × 10<sup>3 </sup>μm<sup>3</sup>. There was no further addition of myonuclei from P21, but myofibre volume continued to increase, resulting in an estimated ~3-fold expansion of the myonuclear domain to 26.5 × 10<sup>3 </sup>μm<sup>3 </sup>by P56. We also used our two mouse models to determine the number of satellite cells per myofibre during postnatal growth. Satellite cell number in EDL was initially ~14 satellite cells per myofibre at P7, but then fell to reach the adult level of ~5 by P21.</p> <p>Conclusions</p> <p>Postnatal fast muscle fibre type growth is divided into distinct phases in mouse EDL: myofibre hypertrophy is initially supported by a rapid increase in the number of myonuclei, but nuclear addition stops around P21. Since the significant myofibre hypertrophy from P21 to adulthood occurs without the net addition of new myonuclei, a considerable expansion of the myonuclear domain results. Satellite cell numbers are initially stable, but then decrease to reach the adult level by P21. Thus the adult number of both myonuclei and satellite cells is already established by three weeks of postnatal growth in mouse.</p
