    Progress with variable cycle engines

    The evaluation of components of an advanced propulsion system for a future supersonic cruise vehicle is discussed. These components, a high performance duct burner for thrust augmentation and a low jet noise coannular exhaust nozzle, are part of the variable stream control engine. An experimental test program involving both isolated component and complete engine tests was conducted for the high performance, low emissions duct burner with excellent results. Nozzle model tests were completed which substantiate the inherent jet noise benefit associated with the unique velocity profile possible of a coannular exhaust nozzle system on a variable stream control engine. Additional nozzle model performance tests have established high thrust efficiency levels at takeoff and supersonic cruise for this nozzle system. Large scale testing of these two critical components is conducted using an F100 engine as the testbed for simulating the variable stream control engine

    Variable Cycle Engine Technology Program Planning and Definition Study

    The variable stream control engine, VSCE-502B, was selected as the base engine, with the inverted flow engine concept selected as a backup. Critical component technologies were identified, and technology programs were formulated. Several engine configurations were defined on a preliminary basis to serve as demonstration vehicles for the various technologies. The different configurations present compromises in cost, technical risk, and technology return. Plans for possible variably cycle engine technology programs were formulated by synthesizing the technology requirements with the different demonstrator configurations

    Novel pollen analogue technique shows bumblebees display low floral constancy and prefer sites with high floral diversity

    This project was funded by the Leverhulme Trust and the Eva Crane Trust.Context There have been dramatic global declines in pollinating insects. A common land management intervention to support wild pollinators is to plant non-crop flowering plants (β€˜pollinator planting’). However, there are limited data on which species or spatial arrangement of planting provide maximum benefit to wild pollinators. Objectives Here we investigate which flowering species and locations are visited by free-foraging Bombus terrestris (buff-tailed bumblebees) in species-rich semi-natural grassland and woodland. Methods Two study nests of buff-tailed bumblebees were established in Wytham Woods, UK. Pollen analogue pigments were sprayed on open flowers in the study area over a period of two months, with unique colours used to identify separate sections of the study area. Pollen load analysis was used to identify forage species and foraging locations. Results Bumblebees showed low flower constancy, visiting five flower species per trip on average, and as a group the sampled bumblebees visited 36 of the 49 plant species identified in study area surveys. Many individuals foraged in multiple, spatially-discrete locations during single trips. Conclusions The positive relationship between floral diversity and pollen load species diversity, and the positive relationship between site floral diversity and frequency of visitation, suggest behavioural strategies that maximize the diversity of flower species visited, in line with the energetic costs and benefits hypothesis. This supports recommendations for pollinator plantings with high species diversity, potentially spread across many small forage areas across the landscape.Peer reviewe

    Functional evolution of the trace amine associated receptors in mammals and the loss of TAAR1 in dogs

    <p>Abstract</p> <p>Background</p> <p>The trace amine associated receptor family is a diverse array of GPCRs that arose before the first vertebrates walked on land. Trace amine associated receptor 1 (TAAR1) is a wide spectrum aminergic receptor that acts as a modulator in brain monoaminergic systems. Other trace amine associated receptors appear to relate to environmental perception and show a birth-and-death pattern in mammals similar to olfactory receptors.</p> <p>Results</p> <p>Across mammals, avians, and amphibians, the TAAR1 gene is intact and appears to be under strong purifying selection based on rates of amino acid fixation compared to neutral mutations. We have found that in dogs it has become a pseudogene. Our analyses using a comparative genetics approach revealed that the pseudogenization event predated the emergence of the Canini tribe rather than being coincident with canine domestication. By assessing the effects of the TAAR1 agonist Ξ²-phenylethylamine on [<sup>3</sup>H]dopamine uptake in canine striatal synaptosomes and comparing the degree and pattern of uptake inhibition to that seen in other mammals, including TAAR1 knockout mice, wild type mice and rhesus monkey, we found that the TAAR1 pseudogenization event resulted in an uncompensated loss of function.</p> <p>Conclusion</p> <p>The gene family has seen expansions among certain mammals, notably rodents, and reductions in others, including primates. By placing the trace amine associated receptors in an evolutionary context we can better understand their function and their potential associations with behavior and neurological disease.</p

    Cleft Palate Is Caused by CNS Dysfunction in Gad1 and Viaat Knockout Mice

    BACKGROUND: Previous studies have shown that disruption of GABA signaling in mice via mutations in the Gad1, Gabrb3 or Viaat genes leads to the development of non-neural developmental defects such as cleft palate. Studies of the Gabrb3 and Gad1 mutant mice have suggested that GABA function could be required either in the central nervous system or in the palate itself for normal palatogenesis. METHODOLOGY/PRINCIPAL FINDINGS: To further examine the role of GABA signaling in palatogenesis we used three independent experimental approaches to test whether Gad1 or Viaat function is required in the fetal CNS for normal palate development. We used oral explant cultures to demonstrate that the Gad1 and Viaat mutant palates were able to undergo palatogenesis in culture, suggesting that there is no defect in the palate tissue itself in these mice. In a second series of experiments we found that the GABA(A) receptor agonist muscimol could rescue the cleft palate phenotype in Gad1 and Viaat mutant embryos. This suggested that normal multimeric GABA(A) receptors in the CNS were necessary for normal palatogenesis. In addition, we showed that CNS-specific inactivation of Gad1 was sufficient to disrupt palate development. CONCLUSIONS/SIGNIFICANCE: Our results are consistent with a role for Gad1 and Viaat in the central nervous system for normal development of the palate. We suggest that the alterations in GABA signaling lead to non-neural defects such as cleft palate as a secondary effect due to alterations in or elimination of fetal movements

    Atomistic study on the pressure dependence of the melting point of NdFe12

    We investigated, using molecular dynamics, how pressure affects the melting point of the recently theorised and epitaxially grown structure NdFe12. We modified Morse potentials using experimental constants and a genetic algorithm code, before running two-phase solid-liquid coexistence simulations of NdFe12 at various temperatures and pressures. The refitting of the Morse potentials allowed us to significantly improve the accuracy in predicting the melting temperature of the constituent elements

    Quantitative molecular assessment of chimerism across tissues in marmosets and tamarins

    <p>Abstract</p> <p>Background</p> <p>Marmosets are playing an increasingly large and important role in biomedical research. They share genetic, anatomical, and physiological similarities with humans and other primate model species, but their smaller sizes, reproductive efficiency, and amenability to genetic manipulation offer an added practicality. While their unique biology can be exploited to provide insights into disease and function, it is also important that researchers are aware of the differences that exist between marmosets and other species. The New World monkey family Callitrichidae, containing both marmoset and tamarin species, typically produces dizygotic twins that show chimerism in the blood and other cells from the hematopoietic lineage. Recently, a study extended these findings to identify chimerism in many tissues, including somatic tissues from other lineages and germ cells. This has raised the intriguing possibility that chimerism may play an increasingly pervasive role in marmoset biology, ranging from natural behavioral implications to increased variability and complexity in biomedical studies.</p> <p>Results</p> <p>Using a quantitative PCR based methodology, Y-chromosomes can be reliably detected in the females with male fraternal twins allowing for a relative quantification of chimerism levels between individuals and tissues. With this approach in common marmosets (<it>Callithrix jacchus</it>) and cotton-top tamarins (<it>Saguinus oedipus</it>), chimerism was detected across a broad array of tissues. Chimerism levels were significantly higher in tissues primarily derived from the hematopoietic lineage, while they were lower, though still detectable, in tissues with other origins. Interestingly, animals with a characteristic marmoset wasting disease show higher levels of chimerism in those tissues affected. Fibroblast cell lines from chimeric individuals, however, are not found to be chimeric themselves.</p> <p>Conclusion</p> <p>Taken together, the levels of chimerism in tissues of different origins coupled with other lines of evidence suggest that indeed only hematopoietic cell lineages are chimeric in callitrichids. The chimerism detected in other tissues is likely the result of blood or lymphocytic infiltration. Using molecular methods to detect chimerism in a tissue sample seems to have allowed a substantial increase in the ability to detect these minor cell populations.</p
