A temperatureâcontrolled, circular maintenance system for studying growth and development of pelagic tunicates (salps)
Salps have attracted attention as zooplankton organisms that may be able to expand their habitat range and increase their ecological importance in the face of ongoing global warming. Due to their gelatinous nature, unique feeding strategy, and reproductive ecology such changes could have profound impacts on regional marine ecosystems. While their role in the regional carbon cycle is receiving attention, our knowledge of their physiology and life cycle is still limited. This knowledge gap is mainly due to their fragile gelatinous nature, which makes it difficult to capture and maintain intact specimen in the laboratory. We present here a modified kreisel tank system that has been tested onboard a research vessel with the Southern Ocean salp Salpa thompsoni and at a research station with Salpa fusiformis and Thalia democratica from the Mediterranean Sea. Successful maintenance over days to weeks allowed us to obtain relative growth and developmental rates comparable to in situ field samples of S. thompsoni and S. fusiformis, and provided insights into previously unknown features of their life cycle (e.g., testes development). Our results show that traditional methods of estimating growth, such as cohort analysis, may lead to a general overestimation of growth rates and neglect individual strategies (e.g., shrinkage), which can affect the results and conclusions drawn from population dynamic models. By providing a starting point for the successful maintenance of different species, comparable experiments on the physiology of salps is made possible. This will contribute to refining model parameters and improving the reliability of the predictions
Seasonal and Form-SpeciïŹc Gene Expression Signatures Uncover Different Generational Strategies of the Pelagic Tunicate Salpa thompsoni During the Southern Ocean Winter
The pelagic tunicate Salpa thompsoni is recognized as a major metazoan grazer in the Southern Ocean. Long term observations show an increase in this speciesâ biomass and a southward shift in its distribution both of which are positively correlated with ocean warming and winter sea ice decline around the Antarctic Peninsula. However, our understanding on how salps adapt their life cycle to the extreme seasonality of the Southern Ocean and the putative differences between its two reproductive forms (aggregates, solitaries) is rudimentary. In particular, our current knowledge of whether and how S. thompsoni overwinter is limited, largely due to winter sampling constraints. In this study, we investigated the form-speciïŹc gene expression proïŹles of Salpa thompsoni during the austral autumn and winter. Between the seasons, genes related to translation showed the biggest difference in gene expression. We found more genes were upregulated in solitaries compared to aggregates, indicating a potentially form-speciïŹc overwintering strategy. Our data provide ïŹrst insights into the seasonal and form-speciïŹc physiology of salps by considering their complex life cycle, thereby contributing to a more comprehensive understanding of the response of salps to seasonal changes in their environment and to anthropogenic induced global climate change
A TNF-Regulated Recombinatorial Macrophage Immune Receptor Implicated in Granuloma Formation in Tuberculosis
Macrophages play a central role in host defense against mycobacterial infection and anti- TNF therapy is associated with granuloma disorganization and reactivation of tuberculosis in humans. Here, we provide evidence for the presence of a T cell receptor (TCR) αÎČ based recombinatorial immune receptor in subpopulations of human and mouse monocytes and macrophages. In vitro, we find that the macrophage-TCRαÎČ induces the release of CCL2 and modulates phagocytosis. TNF blockade suppresses macrophage-TCRαÎČ expression. Infection of macrophages from healthy individuals with mycobacteria triggers formation of clusters that express restricted TCR VÎČ repertoires. In vivo, TCRαÎČ bearing macrophages abundantly accumulate at the inner host-pathogen contact zone of caseous granulomas from patients with lung tuberculosis. In chimeric mouse models, deletion of the variable macrophage-TCRαÎČ or TNF is associated with structurally compromised granulomas of pulmonary tuberculosis even in the presence of intact T cells. These results uncover a TNF-regulated recombinatorial immune receptor in monocytes/macrophages and demonstrate its implication in granuloma formation in tuberculosis

Molecular bases of soft coral reproduction
The early development of the hard coral Acropora millepora has been described at both the morphological and molecular levels, and comparative studies are under way for other Scleractinia. Complementary data on Octocoral embryogenesis are, however, essential for a broader understanding of the different developmental strategies in Cnidaria and their evolution. Although the morphology of soft coral embryogenesis has been previously described, data are scarce and molecular data complementing the morphology are missing. This thesis involves the investigation of reproduction and early development in soft corals, using Lobophytum pauciflorum as model. As in the majority of soft corals, L. pauciflorum colonies are either female or male. The time of reproduction approximately coincides with the annual synchronised mass spawning of hard corals on the Great Barrier Reef (GBR), although in favourable years studied, it occurred a lunar month earlier. The process of gametogenesis is longer in the gonochoric soft coral than in the hermaphrodite hard coral A. millepora or the gonochoric hard coral Fungia concinna. However oogenesis is completed within a year, which is shorter than has been reported for a conspecific population in Taiwan. In addition, bidirectional sex change was observed in F. concinna, suggesting that this trait could also be a predominant reproductive feature in fungiid corals.
After studying the gametogenic cycle of L. pauciflorum on the GBR and identifying the time point of spawning as 4 â 5 nights after the austral spring full moon, investigation of early development was possible. Following mixing of eggs and sperm, the first obvious signs of successful fertilisation occurred at the 16 â 32 cell stages. Through a series of cell divisions leading to irregular shapes, embryos developed into a swimming planula within 48 hours of fertilisation. Planulae then stayed in the water column for up to a week before settling and developing into a primary polyp with 8 tentacular buds in the 24 hours after settlement. Within the first week of metamorphosis, sclerites were visible inside the polyp tissue. Early development in L. pauciflorum appears to be slower than in A. millepora, and it was not possible to identify developmental stages corresponding to those of hard corals on the basis of morphology. Some similarities with hard corals were observed in terms of timing of expression of genes involved in calcification.
In order to extend the knowledge on soft coral microbial communities, microbes associated with the entire colony were also investigated in Lobophytum pauciflorum. In a first approach the possibility of sexâspecific differences in the core microbiome of L. pauciflorum was investigated. The profile of ribotypes present in at least 51 % of the samples was similar to the general profile when all ribotypes are considered. The microbial community in L. pauciflorum was dominated by Spirochaetesârelated ribotypes, though aâProteobacteria were the second most abundant class. Further, microbial profiles were very similar between male and female colonies in terms of highly abundant ribotypes, although sequences that cannot be assigned to any known bacteria were significantly more abundant among female than male colonies. Some sexâspecific differences were detectable in ribotypes of low abundance. However, at class and family level, these differences do not persist, suggesting that the different ribotypes exhibit similar functions. The high abundance of unknown ribotypes provides the potential for discovery of novel bacterial strains with potentially different functions. In a second study, the effect of environmental stresses on the microbial community of L. pauciflorum was investigated. Microbial profiles of Lobophytum fragments exposed to 1) elevated temperatures, 2) reduced pH and 3) a combination of elevated temperatures and reduced pH for 12 days did not vary from control conditions. However, a clear pattern of colony specific microbial profiles at ribotype level emerged. A Spirochaeteâdominated microbial community has only recently been identified in a gorgonian species in the Mediterranean Sea and deserves further characterisation. Combining the results of both studies showed that the microbial community of L. pauciflorum is conserved, with limited colony specific variability. Further, the microbial community appears robust to short term stresses, however the effect of longerâterm stress deserves further investigation

Damit steht Ăffentlichen Bibliotheken in Deutschland ein neuer Distributions- und Kommunikationskanal offen, den sie durch die Bereitstellung von mobilen Bibliotheksanwendungen sinnvoll nutzen könnten.
Ziel der vorliegenden Arbeit war es, sowohl aus Sicht der Ăffentlichen Bibliotheken als auch aus der Perspektive der potentiellen Nutzer, zu erforschen, ob und in welcher Form (Funktionen und Gestaltung) mobile Anwendungen fĂŒr Ăffentliche Bibliotheken in Deutschland heute und in Zukunft relevant und realisierbar sein könnten. Hauptgegenstand der AusfĂŒhrungen sind die DurchfĂŒhrung und Auswertung einer deutschlandweiten Online-Umfrage unter Ăffentlichen Bibliotheken sowie eines Workshops. Zudem wird ein auf Basis der Studienergebnisse erstellter Dummy einer mobilen Bibliotheks-Applikation vorgestellt.
Die vorliegende Arbeit wurde als eine WeiterfĂŒhrung der bereits bestehenden For-schung zu mobilen Anwendungen im Bibliotheksbereich verstanden. In diesem Sinne wurde der Fokus der AusfĂŒhrungen auf der Vermittlung der durch die umgesetzten Studien neu gewonnen Erkenntnisse gelegt
Facing Southern Ocean warming: Temperature effects on whole animal performance of Antarctic krill (Euphausia superba)
The ongoing environmental changes in the Southern Ocean may cause a dramatic decrease in habitat quality. Due to its central position in the food web, Antarctic krill (Euphausia superba) is a key species of the marine Antarctic ecosystem. It is therefore crucial to understand how increasing water temperatures affect important krill life-cycle processes. Here, a long-term (August â March) laboratory acclimation experiment at different temperature scenarios (0.5 âŠC, 1.5 âŠC, 2.5 âŠC, 3.5 âŠC, 5 âŠC, 7 âŠC) was performed and the effects of elevated temperatures on whole animal parameters (O2 consumption, body length, length of the digestive gland) were analyzed. The response of krill oxygen consumption to different experimental temperatures differed between acute/short-term and long-term acclimation. After 8 months, krill oxygen consumption remained unchanged up to temperatures of 3.5 âŠC and was significantly higher at temperatures > 3.5 âŠC. Krill acclimated to temperatures â„ 3.5 âŠC were significantly smaller at the end of the experiment. Limited food intake and/or conversion may
have contributed to this effect, especially pronounced after the onset of the reproductive period. In addition, the seasonal growth pattern in males differed from that of females. Together, our findings indicate that warming Southern Ocean waters are likely to increase metabolic rate in krill, possibly altering the amount of energy
available for other important life-cycle processes, a finding directly related to future population dynamics and
fisheries management
Seawater carbonate chemistry and the microbiome of the octocoral Lobophytum pauciflorum
Bacteria associated with marine invertebrates are thought to have a range of important roles that benefit the host including production of compounds that may exclude pathogenic microorganisms and recycling of essential nutrients. This study characterised the microbiome of a gonochoric octocoral, Lobophytum pauciflorum, and investigated whether either sex or environmental stresses influenced the diversity of the associated microbiome through amplicon profiling of the bacterial 16S rRNA gene. Sequences affiliated to Spirochaetaceae and Endozoicimonaceae dominated the microbiome of L. pauciflorum, representing 43% and 21% of the community, respectively. Among the dominant class affiliations, no sex-specific differences were detected, though unassigned sequences were at a 2-fold higher relative abundance in samples from female individuals than from males. These potentially novel sequences contributed to observed differences between sexes as detected by a multivariate analysis at the OTU level. Exposing L. pauciflorum fragments to increased temperature (31°C), decreased pH (7.9) or both stressors simultaneously for 12 days did not significantly alter the microbial community, indicating that the soft coral microbiome is relatively resilient to short-term environmental stress
