    Maximum principle for a Markovian regime switching system under model uncertainty

    In this paper, we study a stochastic optimal control problem with a Markovian regime switching system, where the coefficients of the state equation and the cost functional are uncertain. First, we obtain the variational inequality by showing the continuity with respect to the uncertainty parameter of the variational equation, which is characterized as forward-backward stochastic differential equations. Second, using the linearization method and weak convergence technique, we prove the necessary stochastic maximum principle and show the sufficient condition of the stochastic optimal control. Finally, as an application, a risk-minimizing portfolio selection problem is studied. Meanwhile, the LβL^\beta-solution and LβL^\beta-estimate of stochastic differential equations with regime switching are given for \b=2k with kNk\in \mathbb{N}.Comment: 37 Page

    Buccal Transmucosal Delivery System of Enalapril for Improved Cardiac Drug Delivery: Preparation and Characterization

    Purpose: To prepare and characterize buccal transmucosal delivery system of enalapril maleate for overcoming its low bioavailability, and hence provide improved therapeutic efficacy and patient compliance.Methods: Transmucosal drug delivery systems of enalapril maleate were formulated as buccal films by solvent casting technique using polyvinylpyrrolidone K90, hydroxypropyl methylcellulose, sodium carboxymethylcellulose (high viscosity). The films were evaluated for film weight, thickness, folding endurance, drug content uniformity, surface pH, in vitro residence time, in vitro drug release and ex-vivo permeation.Results: All the formulations showed high drug content (96.45 to 98.49 %). Those with good swelling showed good residence time. In vitro drug release was highest for films prepared with high viscosity grade sodium carboxymethylcellulose (SCMC- HV,F2), releasing 92.24 % of drug in 1.5 h) followed by F4 (containing polyvinyl pyrrolidone K-90 1 % w/v and SCMC (HV) 1 % w/v). Ex-vivo drug permeation at the end of 10 h was 82.24 and 89.9 % for F2 and F4, respectively.Conclusion: Prompt drug release was obtained from the formulation (F2) containing SCMC 2 % w/v with 10 mg enalapril. However, on the basis of the highest swelling and residence time, and controlled drug release, formulation F4 (containing PVP K-90 and SCMC HV) would be suitable for the development of buccal film for effective therapy of cardiac diseases.Keywords: Cardiac disease, Transmucosal, Buccal films, Enalapril maleate, Drug release, Ex-vivo permeatio

    Increased brain iron in patients with thyroid-associated ophthalmopathy: a whole-brain analysis

    BackgroundTo investigate the whole-brain iron deposition alternations in patients with thyroid-associated ophthalmopathy (TAO) using quantitative susceptibility mapping (QSM).MethodsForty-eight patients with TAO and 33 healthy controls (HCs) were enrolled. All participants underwent brain magnetic resonance imaging scans and clinical scale assessments. QSM values were calculated and compared between TAO and HCs groups using a voxel-based analysis. A support vector machine (SVM) analysis was performed to evaluate the performance of QSM values in differentiating patients with TAO from HCs.ResultsCompared with HCs, patients with TAO showed significantly increased QSM values in the bilateral caudate nucleus (CN), left thalamus (TH), left cuneus, left precuneus, right insula and right middle frontal gyrus. In TAO group, QSM values in left TH were positively correlated with Hamilton Depression Rating Scale (HDRS) scores (r = 0.414, p = 0.005). The QSM values in right CN were negatively correlated with Montreal Cognitive Assessment (MoCA) scores (r = -0.342, p = 0.021). Besides that, a nearly negative correlation was found between QSM values in left CN and MoCA scores (r = -0.286, p = 0.057). The SVM model showed a good performance in distinguishing patients with TAO from the HCs (area under the curve, 0.958; average accuracy, 90.1%).ConclusionPatients with TAO had significantly increased iron deposition in brain regions corresponding to known visual, emotional and cognitive deficits. QSM values could serve as potential neuroimaging markers of TAO

    Study on risk control of water inrush in tunnel construction period considering uncertainty

    Water inrush risk is a bottleneck problem affecting the safety and smooth construction of tunnel engineering works, so the risk control of water inrush is important, however, geological uncertainty and artificial uncertainty always accompany tunnel construction. Uncertainty will not only affect the accuracy of water inrush risk assessment results, but also affect the reliability of water inrush risk decision-making results. How to control the influence of uncertainty on water inrush risk is key to solving the problem of water inrush risk control. Based on the definition of improved risk, a risk analysis model of water inrush based on a fuzzy Bayesian network is constructed. The main factors affecting the risk of water inrush are determined by sensitivity analysis, and possible schemes in risk control of water inrush are proposed. Based on the characteristics of risk control of water inrush in a tunnel, a multi-attribute group decision-making model is constructed to determine the optimal water inrush risk control scheme, so that the optimal scheme for reducing uncertainty in risk control of water inrush is determined. Finally, this system is applied to Shiziyuan Tunnel. The results show that the proposed risk control system for reducing uncertainty of water inrush is efficacious.
