    Proctolin and an Endogenous Proctolin-Like Peptide Enhance the Contractility of the Limulus Heart

    Synthetic proctolin increases the force but not the rate of heart contractions of Limulus in a time- and dose-dependent manner. The threshold of this effect is 3 × 10−10M and the ED50 is approximately 10−8M. At concentrations up to 10−7 M, proctolin has no effect on the rhythmic electrical activity of the isolated cardiac ganglion, and it does not change the simple and compound postsynaptic potentials recorded at the cardiac neuromuscular junction. Proctolin acts directly on the cardiac muscle fibres. Electrically stimulated myocardia show a proctolin-induced increase in contraction amplitude with the same concentration dependence as the intact heart. A compound with an apparent molecular weight of 700–800 occurs in the Limulus nervous system, particularly in the cardiac ganglion. This compound resembles proctolin in being heat-stable, resistant to trypsin and chymotrypsin cleavage, and losing activity in a time-dependent manner in response to treatment with leucine aminopeptidase or pronase. This peptide induces spontaneous contractions and a contracture of the cockroach hindgut in a manner similar to proctolin. Moreover, the Limulus inotropic peptide, like proctolin, increases the force of contraction of the Limulus heart without affecting beat frequency. It is concluded that an endogenous, proctolin-like peptide is an inotropic modulator of the Limulus heart, acting directly on the muscle fibres and not affecting cardiac ganglion activity

    Using nestedness and species-accumulation analyses to strengthen a conservation plan for littoral forest birds in south-eastern Madagascar

    The littoral forests of south-eastern Madagascar are among the most threatened ecosystems on the island. A conservation plan has been developed for the region due to a proposed mining venture. Here, we provide a novel methodology to assess if the planned conservation measures would effectively conserve the bird diversity inhabiting these forests. Bird community composition within 30 littoral forest fragments was quantified with each fragment characterized by measures of fragment area, isolation, and internal habitat complexity. A nestedness and cumulative species–area analysis was conducted to ascertain the contribution of forest fragments of different sizes in capturing the overall bird species richness. Datasets representing the overall and forest-dependent bird assemblages were found to be significantly nested. The pattern of nestedness appeared to be driven by fragment size. However, cumulative species–area analyses showed that the assemblages were imperfectly nested with ten species displaying idiosyncratic distribution patterns. When a modest conservation target was set (the occurrence of a bird species in three or more fragments), the proposed conservation plan would only protect approximately half the species found in the littoral forests. We show that protecting an additional four large patches would mean that the proportion of forest-birds captured in three or more patches would increase to 70%

    Assembly of hundreds of novel bacterial genomes from the chicken caecum

    BACKGROUND: Chickens are a highly important source of protein for a large proportion of the human population. The caecal microbiota plays a crucial role in chicken nutrition through the production of short-chain fatty acids, nitrogen recycling, and amino acid production. In this study, we sequence DNA from caecal content samples taken from 24 chickens belonging to either a fast or a slower growing breed consuming either a vegetable-only diet or a diet containing fish meal. RESULTS: We utilise 1.6 T of Illumina data to construct 469 draft metagenome-assembled bacterial genomes, including 460 novel strains, 283 novel species, and 42 novel genera. We compare our genomes to data from 9 European Union countries and show that these genomes are abundant within European chicken flocks. We also compare the abundance of our genomes, and the carbohydrate active enzymes they produce, between our chicken groups and demonstrate that there are both breed- and diet-specific microbiomes, as well as an overlapping core microbiome. CONCLUSIONS: This data will form the basis for future studies examining the composition and function of the chicken caecal microbiota

    Contrasting alterations to synaptic and intrinsic properties in upper-cervical superficial dorsal horn neurons following acute neck muscle inflammation

    Background: Acute and chronic pain in axial structures, like the back and neck, are difficult to treat, and have incidence as high as 15%. Surprisingly, most preclinical work on pain mechanisms focuses on cutaneous structures in the limbs and animal models of axial pain are not widely available. Accordingly, we developed a mouse model of acute cervical muscle inflammation and assessed the functional properties of superficial dorsal horn (SDH) neurons.<p></p> Results: Male C57/Bl6 mice (P24-P40) were deeply anaesthetised (urethane 2.2?g/kg i.p) and the rectus capitis major muscle (RCM) injected with 40??l of 2% carrageenan. Sham animals received vehicle injection and controls remained anaesthetised for 2?hrs. Mice in each group were sacrificed at 2?hrs for analysis. c-Fos staining was used to determine the location of activated neurons. c-Fos labelling in carrageenan-injected mice was concentrated within ipsilateral (87% and 63% of labelled neurons in C1 and C2 segments, respectively) and contralateral laminae I - II with some expression in lateral lamina V. c-Fos expression remained below detectable levels in control and sham animals. In additional experiments, whole cell recordings were obtained from visualised SDH neurons in transverse slices in the ipsilateral C1 and C2 spinal segments. Resting membrane potential and input resistance were not altered. Mean spontaneous EPSC amplitude was reduced by ~20% in neurons from carrageenan-injected mice versus control and sham animals (20.63???1.05 vs. 24.64???0.91 and 25.87???1.32 pA, respectively). The amplitude (238???33 vs. 494???96 and 593???167 pA) and inactivation time constant (12.9???1.5 vs. 22.1???3.6 and 15.3???1.4?ms) of the rapid A type potassium current (IAr), the dominant subthreshold current in SDH neurons, were reduced in carrageenan-injected mice.<p></p> Conclusions: Excitatory synaptic drive onto, and important intrinsic properties (i.e., IAr) within SDH neurons are reduced two hours after acute muscle inflammation. We propose this time point represents an important transition period between peripheral and central sensitisation with reduced excitatory drive providing an initial neuroprotective mechanism during the early stages of the progression towards central sensitisation

    Extreme asteroids in the Pan-STARRS 1 Survey

    Using the first 18 months of the Pan-STARRS 1 survey we have identified 33 candidate high-amplitude objects for follow-up observations and carried out observations of 22 asteroids. 4 of the observed objects were found to have observed amplitude Aobs≥1.0A_{obs}\geq 1.0 mag. We find that these high amplitude objects are most simply explained by single rubble pile objects with some density-dependent internal strength, allowing them to resist mass shedding even at their highly elongated shapes. 3 further objects although below the cut-off for 'high-amplitude' had a combination of elongation and rotation period which also may require internal cohesive strength, depending on the density of the body. We find that none of the 'high-amplitude asteroids' identified here require any unusual cohesive strengths to resist rotational fission. 3 asteroids were sufficiently observed to allow for shape and spin pole models to be determined through light curve inversion. 45864 was determined to have retrograde rotation with spin pole axes λ=218±10∘,β=−82±5∘\lambda=218\pm 10^{\circ}, \beta=-82\pm 5^{\circ} and asteroid 206167 was found to have best fit spin pole axes λ=57±5∘\lambda= 57 \pm 5^{\circ}, β=−67±5∘\beta=-67 \pm 5^{\circ}. An additional object not initially measured with Aobs>1.0A_{obs}>1.0 mag, 49257, was determined to have a shape model which does suggest a high-amplitude object. Its spin pole axes were best fit for values λ=112±6∘,β=6±5∘\lambda=112\pm 6^{\circ}, \beta=6\pm 5^{\circ}.     PCR-Directed Formation of Viral Hybridsin Vitro

    AbstractWhen constructing viruses that have desired hybrid phenotypes, anticipated difficulties include the nonviability of many, possibly most, of the hybrid genomes that can be constructed by incorporation of DNA fragments. Therefore, many different hybrid genomes may have to be constructed in order to find one that is viable. To perform this combinatorial work in a single experiment, we have used bacteriophage T7-infected cell extracts to transfer DNAin vitro.In an extract, we have incubated T7 DNA, together with DNA obtained by polymerase chain reaction (PCR) amplification of the gene (gene 17) for the tail fiber of the T7-related bacteriophage, T3. Afterin vitropackaging of DNA in the extract, hybrid progeny bacteriophage were detected by probing with a T3-specific oligonucleotide; hybrids are found at a frequency of 0.1%. By determination of the nucleotide sequence of the entire gene 17 of 14 independently isolated hybrids, both right and left ends of the PCR fragment are found to be truncated in all hybrids. For all 14 hybrids, the right end is in the same location; the left end is found at 3 different locations. The nonrandom location of the ends is explained by selection among different inserts for viability; that is, most of the hybrid genomes are nonviable. Some hybrids acquire from T3 the desirable phenotype of nonadherence to agarose gels during agarose gel electrophoresis
