    Nutrient addition effects on tropical dry forests: a mini-review from microbial to ecosystem scales.

    Humans have more than doubled inputs of reactive nitrogen globally and greatly accelerated the biogeochemical cycles of phosphorus and metals. However, the impacts of increased element mobility on tropical ecosystems remain poorly quantified, particularly for the vast tropical dry forest biome. Tropical dry forests are characterized by marked seasonality, relatively little precipitation, and high heterogeneity in plant functional diversity and soil chemistry. For these reasons, increased nutrient deposition may affect tropical dry forests differently than wet tropical or temperate forests. Here, we review studies that investigated how nutrient availability affects ecosystem and community processes from the microsite to ecosystem scales in tropical dry forests. The effects of N and P addition on ecosystem carbon cycling and plant and microbial dynamics depend on forest successional stage, soil parent material, and rainfall regime. Responses may depend on whether overall productivity is N- vs. P-limited, although data to test this hypothesis are limited. These results highlight the many important gaps in our understanding of tropical dry forest responses to global change. Large-scale experiments are required to resolve these uncertainties

    Cancer and psychiatric diagnoses in the year preceding suicide

    BACKGROUND: Patients with cancer are known to be at increased risk for suicide but little is known about the interaction between cancer and psychiatric diagnoses, another well-documented risk factor. METHODS: Electronic medical records from nine healthcare systems participating in the Mental Health Research Network were aggregated to form a retrospective case-control study, with ICD-9 codes used to identify diagnoses in the 1 year prior to death by suicide for cases (N = 3330) or matching index date for controls (N = 297,034). Conditional logistic regression was used to assess differences in cancer and psychiatric diagnoses between cases and controls, controlling for sex and age. RESULTS: Among patients without concurrent psychiatric diagnoses, cancer at disease sites with lower average 5-year survival rates were associated with significantly greater relative risk, while cancer disease sites with survival rates of \u3e70% conferred no increased risk. Patients with most psychiatric diagnoses were at higher risk, however, there was no additional risk conferred to these patients by a concurrent cancer diagnosis. CONCLUSION: We found no evidence of a synergistic effect between cancer and psychiatric diagnoses. However, cancer patients with a concurrent psychiatric illness remain at the highest relative risk for suicide, regardless of cancer disease site, due to strong independent associations between psychiatric diagnoses and suicide. For patients without a concurrent psychiatric illness, cancer disease sites associated with worse prognoses appeared to confer greater suicide risk

    Photosynthesis-dependent Hâ‚‚Oâ‚‚ transfer from chloroplasts to nuclei provides a high-light signalling mechanism

    Chloroplasts communicate information by signalling to nuclei during acclimation to fluctuating light. Several potential operating signals originating from chloroplasts have been proposed, but none have been shown to move to nuclei to modulate gene expression. One proposed signal is hydrogen peroxide (H2O2) produced by chloroplasts in a light-dependent manner. Using HyPer2, a genetically encoded fluorescent H2O2 sensor, we show that in photosynthetic Nicotiana benthamiana epidermal cells, exposure to high light increases H2O2 production in chloroplast stroma, cytosol and nuclei. Critically, over-expression of stromal ascorbate peroxidase (H2O2 scavenger) or treatment with DCMU (photosynthesis inhibitor) attenuates nuclear H2O2 accumulation and high light-responsive gene expression. Cytosolic ascorbate peroxidase over-expression has little effect on nuclear H2O2 accumulation and high light-responsive gene expression. This is because the H2O2 derives from a sub-population of chloroplasts closely associated with nuclei. Therefore, direct H2O2 transfer from chloroplasts to nuclei, avoiding the cytosol, enables photosynthetic control over gene expression

    THE RADIAL AND ROTATIONAL VELOCITIES OF PSO J318.5338-22.8603, A NEWLY CONFIRMED PLANETARY-MASS MEMBER OF THE β PICTORIS MOVING GROUP

    PSO J318.5338−-22.8603 is an extremely-red planetary-mass object that has been identified as a candidate member of the β\beta Pictoris moving group based on its spatial position and tangential velocity. We present a high resolution KK-band spectrum of PSO J318.5338−-22.8603. Using a forward-modeling Markov Chain Monte Carlo approach, we report the first measurement of the radial velocity and vv sin(ii) of PSO J318.5−-22, −-6.0−1.1+0.8^{+0.8}_{-1.1} km s−1^{-1} and 17.5−2.8+2.3^{+2.3}_{-2.8} km s−1^{-1}, respectively. We calculate the space velocity and position of PSO J318.5−-22 and confirm that it is a member of the β\beta Pictoris moving group. Adopting an age of 23±\pm3 Myr for PSO J318.5−-22, we determine a mass of 8.3±0.58.3\pm0.5 MJupM_{\rm{Jup}} and effective temperature of 1127−26+241127^{+24}_{-26} K using evolutionary models. PSO J318.5338−-22.8603 is intermediate in mass and temperature to the directly-imaged planets β\beta Pictoris b and 51 Eridani b, making it an important benchmark object in the sequence of planetary-mass members of the β\beta Pictoris moving group. Combining our vv sin(ii) measurement with recent photometric variability data, we constrain the inclination of PSO J318.5−-22 to >29∘>29^{\circ} and its rotational period to 5-10.2 hours. The equatorial velocity of PSO J318.5−-22 indicates that its rotation is consistent with an extrapolation of the velocity-mass relationship for solar system planets.Comment: 8 pages, 5 figures, 2 tables, ApJ accepte

    CUBES: a UV spectrograph for the future

    In spite of the advent of extremely large telescopes in the UV/optical/NIR range, the current generation of 8-10m facilities is likely to remain competitive at ground-UV wavelengths for the foreseeable future. The Cassegrain U-Band Efficient Spectrograph (CUBES) has been designed to provide high-efficiency (>40%) observations in the near UV (305-400 nm requirement, 300-420 nm goal) at a spectral resolving power of R>20,000, although a lower-resolution, sky-limited mode of R ~ 7,000 is also planned. CUBES will offer new possibilities in many fields of astrophysics, providing access to key lines of stellar spectra: a tremendous diversity of iron-peak and heavy elements, lighter elements (in particular Beryllium) and light-element molecules (CO, CN, OH), as well as Balmer lines and the Balmer jump (particularly important for young stellar objects). The UV range is also critical in extragalactic studies: the circumgalactic medium of distant galaxies, the contribution of different types of sources to the cosmic UV background, the measurement of H2 and primordial Deuterium in a regime of relatively transparent intergalactic medium, and follow-up of explosive transients. The CUBES project completed a Phase A conceptual design in June 2021 and has now entered the Phase B dedicated to detailed design and construction. First science operations are planned for 2028.     High-throughput amplicon-based copy number detection of 11 genes in formalin-fixed paraffin-embedded ovarian tumour samples by MLPA-seq

    The Clinical and Scientific Collaborators of the AOCS group include Martin Oehler.Whilst next generation sequencing can report point mutations in fixed tissue tumour samples reliably, the accurate determination of copy number is more challenging. The conventional Multiplex Ligation-dependent Probe Amplification (MLPA) assay is an effective tool for measurement of gene dosage, but is restricted to around 50 targets due to size resolution of the MLPA probes. By switching from a size-resolved format, to a sequence-resolved format we developed a scalable, high-throughput, quantitative assay. MLPA-seq is capable of detecting deletions, duplications, and amplifications in as little as 5ng of genomic DNA, including from formalin-fixed paraffin-embedded (FFPE) tumour samples. We show that this method can detect BRCA1, BRCA2, ERBB2 and CCNE1 copy number changes in DNA extracted from snap-frozen and FFPE tumour tissue, with 100% sensitivity and >99.5% specificity.Olga Kondrashova, Clare J.     Early above- and below-ground responses of subboreal conifer seedlings to various levels of deciduous canopy removal

    We examined the growth of understory conifers, following partial or complete deciduous canopy removal, in a field study established in two regions in Canada. In central British Columbia, we studied the responses of three species (Pseudotsuga menziesii var. glauca (Beissn.) Franco, Picea glauca (Moench) Voss x Picea engelmannii Parry ex Engelm., and Abies lasiocarpa (Hook.) Nutt.), and in northwestern Quebec, we studied one species (Abies balsamea (L.) Mill.). Stem and root diameter and height growth were measured 5 years before and 3 years after harvesting. Both root and stem diameter growth increased sharply following release but seedlings showed greater root growth, suggesting that in the short term, improvement in soil resource capture and transport, and presumably stability, may be more important than an increase in stem diameter and height growth. Response was strongly size dependent, which appears to reflect greater demand for soil resources as well as higher light levels and greater tree vigour before release for taller individuals. Growth ratios could not explain the faster response generally attributed to true fir species or the unusual swift response of spruces. Good prerelease vigour of spruces, presumably favoured by deciduous canopies, could explain their rapid response to release
