Tuning grid storage resources for LHC data analysis
Grid Storage Resource Management (SRM) and local file-system solutions are facing significant challenges to support efficient analysis of the data now being produced at the Large Hadron Collider (LHC). We compare the performance of different storage technologies at UK grid sites examining the effects of tuning and recent improvements in the I/O patterns of experiment software. Results are presented for both live production systems and technologies not currently in widespread use. Performance is studied using tests, including real LHC data analysis, which can be used to aid sites in deploying or optimising their storage configuration

Evaluation of a Brazilian fuel alcohol yeast strain for Scotch whisky fermentations
Traditionally, distilling companies in Scotland have employed a very limited number of yeast strains in the production of alcohol for Scotch whiskies. Recent changes such as the decline in availability of brewers’ yeast as a secondary yeast strain and the availability of yeast in different formats (e.g., dried and cream yeast as alternatives to compressed yeast) have promoted interest in alternative Scotch whisky distilling yeasts. In previous work, we investigated different strains of yeasts, specifically Brazilian yeasts which had been isolated from and used in fuel alcohol distilleries. One of the Brazilian yeasts (CAT 1) showed a comparable fermentation performance and superior stress tolerance compared with a standard commercial Scotch whisky distilling yeast (M Type). The Brazilian CAT 1 yeast isolate was further assessed in laboratory scale fermentations and subsequent new make spirit was subjected to sensory analyses. The spirits produced using the Brazilian strain had acceptable flavour profiles and exhibited no sensory characteristics that were atypical of Scotch whisky new make spirit. This study highlights the potential of exploiting yeast biodiversity in traditional Scotch whisky distillery fermentation processes
Thirty Meter Telescope Site Testing I: Overview
As part of the conceptual and preliminary design processes of the Thirty
Meter Telescope (TMT), the TMT site testing team has spent the last five years
measuring the atmospheric properties of five candidate mountains in North and
South America with an unprecedented array of instrumentation. The site testing
period was preceded by several years of analyses selecting the five candidates,
Cerros Tolar, Armazones and Tolonchar in northern Chile; San Pedro Martir in
Baja California, Mexico and the 13 North (13N) site on Mauna Kea, Hawaii. Site
testing was concluded by the selection of two remaining sites for further
consideration, Armazones and Mauna Kea 13N. It showed that all five candidates
are excellent sites for an extremely large astronomical observatory and that
none of the sites stands out as the obvious and only logical choice based on
its combined properties. This is the first article in a series discussing the

Materials for phantoms for terahertz pulsed imaging
Phantoms are commonly used in medical imaging for quality assurance, calibration, research and teaching. They may include test patterns or simulations of organs, but in either case a tissue substitute medium is an important component of the phantom. The aim of this work was to identify materials suitable for use as tissue substitutes for the relatively new medical imaging modality terahertz pulsed imaging. Samples of different concentrations of the candidate materials TX151 and napthol green dye were prepared, and measurements made of the frequency-dependent absorption coefficient (0.5 to 1.5 THz) and refractive index (0.5 to 1.0 THz). These results were compared qualitatively with measurements made in a similar way on samples of excised human tissue (skin, adipose tissue and striated muscle). Both materials would be suitable for phantoms where the dominant mechanism to be simulated is absorption (similar to ∼100 cm(-1) at 1 THz) and where simulation of the strength of reflections from boundaries is not important; for example, test patterns for spatial resolution measurements. Only TX151 had a frequency-dependent refractive index close to that of tissue, and could therefore be used to simulate the layered structure of skin, the complexity of microvasculature or to investigate frequency-dependent interference effects that have been noted in terahertz images
The night-sky at the Calar Alto Observatory
We present a characterization of the main properties of the night-sky at the
Calar Alto observatory for the time period between 2004 and 2007. We use
optical spectrophotometric data, photometric calibrated images taken in
moonless observing periods, together with the observing conditions regularly
monitored at the observatory, such as atmospheric extinction and seeing. We
derive, for the first time, the typical moonless night-sky optical spectrum for
the observatory. The spectrum shows a strong contamination by different
pollution lines, in particular from Mercury lines, which contribution to the
sky-brightness in the different bands is of the order of ~0.09 mag, ~0.16 mag
and ~0.10 mag in B, V and R respectively. The zenith-corrected values of the
moonless night-sky surface brightness are 22.39, 22.86, 22.01, 21.36 and 19.25
mag arcsec^-2 in U, B, V, R and I, which indicates that Calar Alto is a
particularly dark site for optical observations up to the I-band. The fraction
of astronomical useful nights at the observatory is ~70%, with a ~30% of
photometric nights. The typical extinction at the observatory is k_V~0.15 mag
in the Winter season, with little dispersion. In summer the extinction has a
wider range of values, although it does not reach the extreme peaks observed at
other sites. The median seeing for the last two years (2005-6) was ~0.90",
being smaller in the Summer (~0.87") than in the Winter (~0.96"). We conclude
in general that after 26 years of operations Calar Alto is still a good
astronomical site, being a natural candidate for future large aperture optical

Glass-Like Heat Conduction in High-Mobility Crystalline Semiconductors
The thermal conductivity of polycrystalline semiconductors with type-I
clathrate hydrate crystal structure is reported. Ge clathrates (doped with Sr
and/or Eu) exhibit lattice thermal conductivities typical of amorphous
materials. Remarkably, this behavior occurs in spite of the well-defined
crystalline structure and relatively high electron mobility (). The dynamics of dopant ions and their interaction with the
polyhedral cages of the structure are a likely source of the strong phonon

Cephalosporin-3’-diazeniumdiolate NO-donor prodrug PYRRO-C3D enhances azithromycin susceptibility of non-typeable Haemophilus influenzae biofilms

The Publisher's final version can be found by following the DOI link.Objectives: PYRRO-C3D is a cephalosporin-3-diazeniumdiolate nitric oxide (NO)-donor prodrug designed to selectively deliver NO to bacterial infection sites. The objective of this study was to assess the activity of PYRRO-C3D against non-typeable Haemophilus influenzae (NTHi) biofilms and examine the role of NO in reducing biofilm-associated antibiotic tolerance.
Methods: The activity of PYRRO-C3D on in vitro NTHi biofilms was assessed through CFU enumeration and confocal microscopy. NO release measurements were performed using an ISO-NO probe. NTHi biofilms grown on primary ciliated respiratory epithelia at an air-liquid interface were used to investigate the effects of PYRRO-C3D in the presence of host tissue. Label-free LC/MS proteomic analyses were performed to identify differentially expressed proteins following NO treatment.
Results: PYRRO-C3D specifically released NO in the presence of NTHi, while no evidence of spontaneous NO release was observed when the compound was exposed to primary epithelial cells. NTHi lacking β-lactamase activity failed to trigger NO release. Treatment significantly increased the susceptibility of in vitro NTHi biofilms to azithromycin, causing a log-fold reduction in viability (p<0.05) relative to azithromycin alone. The response was more pronounced for biofilms grown on primary respiratory epithelia, where a 2-log reduction was observed (p<0.01). Label-free proteomics showed that NO increased expression of sixteen proteins involved in metabolic and transcriptional/translational functions.
Conclusions: NO release from PYRRO-C3D enhances the efficacy of azithromycin against NTHi biofilms, putatively via modulation of NTHi metabolic activity. Adjunctive therapy with NO mediated through PYRRO-C3D represents a promising approach for reducing biofilm associated antibiotic tolerance
