    Dynamo in the Desert: Energy Development and Environmental Contestation in the San Juan Basin, 1960-1985

    This thesis examines coal-energy development in the San Juan Basin, which is located in the culturally rich and environmentally diverse Four Corners region. Between 1960 and 1985 intensive coal-energy development—in the form of strip mines and power plants—took place in the western portion of the San Juan Basin. This period saw the enactment of major environmental legislation and the rise of the modern environmental movement. My thesis specifically focuses on how this development unleashed environmental damage upon the regions land, water, and air. Beyond examining environmental destruction, my thesis explores how different groups came into conflict over coal-energy development. The groups that most actively contested the development of coal-energy were Navajo tribal leaders, local Navajo communities, Navajo activists, environmental organizations, and energy and mining corporations. I make the argument that each group built a discourse around energy development, environmental legislation, and the natural environment in ways that conformed to each group\u27s particular interests. The mere specter of energy development unleashed intergroup conflict even when plans for surface mines, coal gasification, and power plant projects did not materialize. Failed energy projects still resulted in social and landscape changes, such as intergroup conflict, factionalizing of Navajo political and social structures, changes in legal control over land, and the designation of wilderness areas within coal regions

    The responses of Tribolium castaneum to wheat germ oil and fungal produced volatiles: Presentation

    The red flour beetle Tribolium castaneum Herbst (Coleoptera: Tenebrionidae) is a significant pest affecting a wide variety of different stored products around the Globe. Despite its economic impact, there is evidence that the lures currently used in traps to monitor for this species are largely ineffective. Based on the evolutionary history of T. castaneum, and the ecological niche it occupies, the volatiles of wheat germ oil and volatiles produced by grain-associated fungi have the potential to act as attractants for this species. We used electroantennography (EAG) to measure the electrophysiological response elicited by sixty-eight volatile compounds found in wheat germ oil and/or grain-associated fungi in two T. castaneum strains; an established lab population (CTC12 strain) and a recently caught wild population. Many volatile compounds from both sources elicited strong antennal depolarisations, and the responses of both strains were highly correlated. We then tested whether the compounds that triggered the strongest antennal depolarisations also elicited behavioural responses by using Y-tube olfactometer bioassays and identified several compounds attractive to both strains. The discovery of novel compounds that elicit strong EAG signals and behavioural responses could prove useful in the design of improved lures for T. castaneum and other stored product pests.     Decreased Serum Hepatocyte Growth Factor (HGF) in Autistic Children with Severe Gastrointestinal Disease

    Aim To assess serum Hepatocyte Growth Factor (HGF) levels in autistic children with severe gastrointestinal (GI) disease and to test the hypothesis that there is a relationship between GI pathology and HGF concentration. Subjects and Methods Serum from 29 autistic children with chronic digestive disease (symptoms for a minimum of 6–12 months), most with ileo-colonic lymphoid nodular hyperplasia (LNH—markedly enlarged lymphoid nodules) and inflammation of the colorectum, small bowel and/or stomach), and 31 controls (11 age matched autistic children with no GI disease, 11 age matched non autistic children without GI disease and 9 age matched non autistic children with GI disease) were tested for HGF using ELISAs. HGF concentration of autistic children with GI disease was compared to GI disease severity. Results Autistic children with GI disease had significantly lower serum levels of HGF compared to controls (autistic without GI disease; p = 0.0005, non autistic with no GI disease; p = 0.0001, and non autistic with GI disease; p = 0.001). Collectively, all autistic children had significantly lower HGF levels when compared to non autistic children (p < 0.0001). We did not find any relationship between severity of GI disease and HGF concentration in autistic children with GI disease. Discussion These results suggest an association between HGF serum levels and the presence of GI disease in autistic children and explain a potential functional connection between the Met gene and autism. The concentration of serum HGF may be a useful biomarker for autistic children, especially those with severe GI disease

    Light-emitting diode street lights reduce last-ditch evasive manoeuvres by moths to bat echolocation calls

    © 2015 The Authors. The light-emitting diode (LED) street light market is expanding globally, and it is important to understand how LED lights affect wildlife populations. We compared evasive flight responses of moths to bat echolocation calls experimentally under LED-lit and -unlit conditions. Significantly, fewer moths performed ‘powerdive’ flight manoeuvres in response to bat calls (feeding buzz sequences from Nyctalus spp.) under an LED street light than in the dark.     Communities as conduits of harm : A social identity analysis of appraisal, coping and justice-seeking in response to historic collective victimization

    Social identity approach (SIA) research shows that commu- nity members often work together to support survivors of collective victimization and rectify social injustices. How- ever, complexities arise when community members have been involved in perpetrating these injustices. While many communities are unaware of their role in fostering victimi- zation, others actively deny their role and responsibility to restore justice. We explore these processes by investigating experiences of community violence and collective justice- seeking among Albanian survivors of dictatorial crimes. Sur- vivors (N = 27) were interviewed, and data were analysed using theoretical thematic analysis guided by the SIA. The analysis reveals the diverse ways communities can become harmful ‘Social Curses’. First, communities in their various forms became effective perpetrators of fear and control (e.g., exclusion and/or withholding ingroup privileges) dur- ing the dictatorship because of the close relationship between communities and their members. Second, commu- nities caused harm by refusing to accept responsibility for the crimes, and by undermining attempts at collective action to address injustices. This lack of collective accountability also fosters survivors' feelings of exclusion and undermines their hope for systematic change. Implica- tions for SIA processes relating to health/wellbeing (both Social Cure and Curse) are discussed. We also discuss impli- cations for understanding collective action and victimhood.Peer reviewe

    Peripheral blood responses to specific antigens and CD28 in sarcoidosis

    SummaryBackgroundPotential antigens inducing sarcoid inflammation include mycobacterial and auto-antigens. Paradoxically, peripheral anergy to common recall antigens also occurs, possibly due to impaired dendritic cell or regulatory T-cell responses, or impaired T-cell co-stimulation. The purpose of this study was to compare peripheral blood responses of patients with sarcoidosis to candidate antigens, and examine CD28 T-cell co-stimulation.MethodsPeripheral blood mononuclear cell (PBMC) responses were examined from patients with sarcoidosis (n=16) and healthy control subjects (n=22) following PBMC stimulation with: anti-CD3/CD28 coated beads; Mycobacterium tuberculosis ESAT-6 and KatG peptides; vimentin and lysyl tRNA peptides; and common recall antigens, including cytomegalovirus (CMV) cell lysate as well as CMV, Epstein-Barr virus, influenza virus (CEF) peptides.ResultsESAT-6/KatG peptide stimulation induced greater numbers of IFN-γ producing T-cells, and elevated IL-2, IL-6 and TNF-α production in sarcoidosis compared to purified protein derivative (PPD)-negative healthy control subjects. PBMCs from patients with sarcoidosis showed reduced IFN-γ producing T-cells following stimulation with CMV lysate, CEF peptides and CD3/CD28 beads; and reduced IL-4 and TNF-α production following CD3/CD28 activation.ConclusionsPatients with sarcoidosis exhibit greater PBMC responses to M. tuberculosis antigens compared to PPD-negative controls, but reduced T-cell responses to common recall antigens. One contributing mechanism may be impairment of T-cell CD28 co-stimulation

    PyEvolve: a toolkit for statistical modelling of molecular evolution

    BACKGROUND: Examining the distribution of variation has proven an extremely profitable technique in the effort to identify sequences of biological significance. Most approaches in the field, however, evaluate only the conserved portions of sequences – ignoring the biological significance of sequence differences. A suite of sophisticated likelihood based statistical models from the field of molecular evolution provides the basis for extracting the information from the full distribution of sequence variation. The number of different problems to which phylogeny-based maximum likelihood calculations can be applied is extensive. Available software packages that can perform likelihood calculations suffer from a lack of flexibility and scalability, or employ error-prone approaches to model parameterisation. RESULTS: Here we describe the implementation of PyEvolve, a toolkit for the application of existing, and development of new, statistical methods for molecular evolution. We present the object architecture and design schema of PyEvolve, which includes an adaptable multi-level parallelisation schema. The approach for defining new methods is illustrated by implementing a novel dinucleotide model of substitution that includes a parameter for mutation of methylated CpG's, which required 8 lines of standard Python code to define. Benchmarking was performed using either a dinucleotide or codon substitution model applied to an alignment of BRCA1 sequences from 20 mammals, or a 10 species subset. Up to five-fold parallel performance gains over serial were recorded. Compared to leading alternative software, PyEvolve exhibited significantly better real world performance for parameter rich models with a large data set, reducing the time required for optimisation from ~10 days to ~6 hours. CONCLUSION: PyEvolve provides flexible functionality that can be used either for statistical modelling of molecular evolution, or the development of new methods in the field. The toolkit can be used interactively or by writing and executing scripts. The toolkit uses efficient processes for specifying the parameterisation of statistical models, and implements numerous optimisations that make highly parameter rich likelihood functions solvable within hours on multi-cpu hardware. PyEvolve can be readily adapted in response to changing computational demands and hardware configurations to maximise performance.
