    Antibiotic Susceptibility and Detection of Resistance Genes of E. coli among Healthy Pregnant Women in Designated Hospitals around Osogbo, Osun State, Nigeria

    Treatment of E. coli is now a challenge globally, due to continuous increase in resistance towards commonly prescribed antibiotics, thus posing a threat to treatment. Hence, the aim of the study is to determine antibiotics sensitivity and resistance genes of E.coli from apparently healthy pregnant women in Osogbo, Osun State, Nigeria using standard methods. A cross-sectional study design was used to collect 150 mid-stream urine (MSU) samples from apparently healthy pregnant women. Standard inoculating loop technique was used by culturing 0.001 ml of MSU on Cysteine Lactose Electrolyte Deficient (CLED) agar, Blood agar and MacConkey agar and incubated at 37 °C for 24 h. A standard agar disc diffusion method was used to determine antimicrobial susceptibility pattern of the isolates. The molecular detection of resistance genes was done using PCR techniques.  The ages of women enrolled in this study ranged from 22 to 42 years (mean ± standard deviation = 31 ± 4.7 years). Eight isolates were positive for E. coli.  Escherichia coli showed high percentage of resistance to ampicillin and low resistance to ciprofloxacin and penicillin. All the E. coli isolates were sensitive to levofloxacin, and most were resistant to Meropenem. Multiple drug resistance was observed in all the isolates. Resistance genes in VIM 390bp, bla ctx-M 585bp and TEM 517bp were detected in some of the representative E. coli isolates profiled.     Evidence of Heavy Metals Distribution in Placenta in Association with Residual Levels in Some Dams’ Organs from Bodija abattoir, Oyo State, Nigeria

    Heavy metals have been known to be causing serious detrimental effect on the health of livestock and human populace in general. However, little study had been carried out on the ability of heavy metal to cross placental barrier, which has already been bio-accumulated in the dam. Therefore, this study aimed at showing the evidence of placental barrier crossing by selected heavy metals. The study was carried out in Bodija abattoir, Ibadan, Oyo-state, Nigeria, located on latitude 70020N, longitude 3005E. A cross-sectional study design was adopted and lasted for 6 weeks. Samples were collected from kidney cortices, apical lobe of dam’s liver and a portion of fetal placenta. 12 sample each from liver, kidney and the placenta which were analyzed using Atomic Absorption Spectrophotometer (AAS). Results were subjected to descriptive statistics, t-test and correlation using SPSS17.0 package.From this study cadmium (Cd) and lead (Pb) residues were not found in the samples, while chromium (Cr) was found in all the samples (100% prevalence) and the total prevalence for the heavy metals in the study was 33.4%. The mean chromium (Cr) residual values in placenta, kidney and liver were 0.89±0.66mg/kg, 1.32±0.94mg/kg and 1.00±0.87 mg/kg respectively. The correlation between chromium (Cr) in the dam’s kidney and placenta was 0.3, while for the dam’s liver and placenta was 0.6.  In comparison with permissible limit, the residual level in kidney 1.32±0.94, liver 1.00±0.87 and placenta 0.89±0.66, were within the permissible limit. The study revealed that chromium (Cr) residue was a major challenge in the organs sampled. Liver of the dam had the highest level of bioaccumulation and stronger correlation in the distribution to the placenta. It is therefore recommended that attention should be paid on the disposal of chromium (Cr) residues on the grazing floor where the bulk of the residues were from.     Amphibian host-defense peptides with potential for Type 2 diabetes therapy - an updated review

    Investigations conducted since 2018 have identified several host-defense peptides present in frog skin secretions whose properties suggest the possibility of their development into a new class of agent for Type 2 diabetes (T2D) therapy. Studies in vitro have described peptides that (a) stimulate insulin release from BRIN-BD11 clonal β-cells and isolated mouse islets, (b) display β-cell proliferative activity and protect against cytokine-mediated apoptosis and (c) stimulate production of the anti-inflammatory cytokine IL-10 and inhibit production of the pro-inflammatory cytokines TNF-α and IL-1β. Rhinophrynin-27, phylloseptin-3.2TR and temporin F are peptides with therapeutic potential. Studies in vivo carried out in db/db and high fat-fed mice have shown that twice-daily administration of [S4K]CPF-AM1 and [A14K]PGLa-AM1, analogs of peptides first isolated from the octoploid frog Xenopus amieti, over 28 days lowers circulating glucose and HbA1c concentrations, increases insulin sensitivity and improves glucose tolerance and lipid profile. Peptide treatment produced potentially beneficial changes in the expression of skeletal muscle genes involved in insulin signaling and islet genes involved in insulin secretion in these murine models of T2D. Lead compounds uncovered by the study of frog HDPs may provide a basis for the design of new types of agents that can be used, alone or in combination with existing therapies, for the treatment of T2D.

    U radu je opisana izolacija pet sastojaka petroleterske i diklormetanske frakcije metanolnog ekstrakta kore biljke Delonix regia: lupeol (1), epilupeol (2), β-sitosterol (3), stigmasterol (4) i p-metoksibenzaldehid (5). Nadalje, testirano je antimikrobno djelovanje različitih ekstrakata difuzijskom metodom na disku (15 μg mm2). Zone inhibicije za sastojke topljive u petroleteru, tetraklormetanu i diklormetanu bile su 914 mm, 1113 mm, odnosno 920 mm, dok je zona inhibicije standarda kanamicina bila 2025 mm. U biološkom pokusu smrtnosti morskih kozica najveću toksičnost pokazali su spojevi topljivi u tetraklormetanu (LC50 = 0,83 μg mL1), dok je topljivost sastojaka topljivih u petroleteru i diklormetanu bila LC50 14,94, odnosno 3,29 μg mL1, a standarda vinkristin sulfata 0,812 μg mL1. Ovo je prvo izvješće o izolaciji sastojaka, antimikrobnom djelovanju i citotoksičnosti biljke D. regia.In this study five compounds, lupeol (1), epilupeol (2), β-sitosterol (3), stigmasterol (4) and p-methoxybenzaldehyde (5) were isolated from the petroleum ether and dichloromethane fractions of a methanolic extract of the stem bark of Delonix regia. Antimicrobial screening of the different extracts (15 μg mm2) was conducted by disc diffusion method. The zones of inhibition demonstrated by the petroleum ether, carbon tetrachloride and dichloromethane fractions ranged from 914 mm, 1113 mm and 920 mm, respectively, compared to kanamycin standard with the zone of inhibition of 2025 mm. In brine shrimp lethality bioassay, the carbon tetrachloride soluble materials demonstrated the highest toxicity with LC50 of 0.83 μg mL1, while petroleum ether and dichloromethane soluble partitionates of the methanolic extract revealed LC50 of 14.94 and 3.29 μg mL1, respectively, in comparison with standard vincristine sulphate with LC50 of 0.812 μg mL1. This is the first report on compounds separation from D. regia, their antimicrobial activity and cytotoxicity

    HUWE1 cooperates with RAS activation to control leukemia cell proliferation and human hematopoietic stem cells differentiation fate

    Acute myeloid leukemia (AML) is a poor prognosis hematopoietic malignance characterized by abnormal proliferation and differentiation of hematopoietic stem cells (HSCs). Although advances in treatment have greatly improved survival rates in young patients, in the elderly population, ~70% of patients present poor prognosis. A pan-cancer analysis on the TCGA cohort showed that AML has the second higher HUWE1 expression in tumor samples among all cancer types. In addition, pathway enrichment analysis pointed to RAS signaling cascade as one of the most important pathways associated to HUWE1 expression in this particular AML cohort. In silico analysis for biological processes enrichment also revealed that HUWE1 expression is correlated with 13 genes involved in myeloid differentiation. Therefore, to understand the role of HUWE1 in human hematopoietic stem and progenitor cells (HSPC) we constitutively expressed KRASG12V oncogene concomitantly to HUWE1 knockdown in stromal co-cultures. The results showed that, in the context of KRASG12V, HUWE1 significantly reduces cell cumulative growth and changes myeloid differentiation profile of HSPCs. Overall, these observations suggest that HUWE1 might contribute to leukemic cell proliferation and impact myeloid differentiation of human HSCs, thus providing new venues for RAS-driven leukemia targeted therapy approach

Unlike other types of energy recovery exchangers, the air temperature at the outlet of FBRs varies with time, which creates challenges when measuring the outlet temperature and effectiveness of FBRs since the actual outlet air temperature will include the transient response of the FBR and the temperature sensor. In this article, a validated numerical model of FBRs that takes into account the sensor response is used to quantify the temperature and effectiveness errors that result due to sensors’ response characteristics over a wide range of design parameters. The main contributions of this article are the practical recommendations for the temperature measurement for different types of FBRs developed for HVAC applications. The recommendations presented in this article could be implemented in future versions of the current standards (ASHRAE 84 and CSA C439-18 standards) for performance testing of air-to-air energy exchangers

    Single neuron perceptron is designed as a classifier of two different classes using the hardlimiter activation function (i.e. in the absence of light, and presence of light). An example is designed and tested so that the proposed circuit learned different categories and then used as a classifier for two different classes because of the use of single neuron. Additional electronic circuits were used for computation processes. The Computer simulation results indicate stable solution that compares with theoretical results. Single layer perceptron M-category classifier is designed as a classifier for more than two classes. An example is designed and tested for the verification. The example learns after (5) iterations. Computer simulation results indicate stable solution that compares favorably with theoretical results

    Mobile ad hoc network testbed using mobile robot technology

    MANET (Mobile Ad Hoc Network) researchers have shown increased interest in using mobile robot technology for their testbed platforms. Thus, the main motivation of this paper is to review various robot-based MANET testbeds that have been developed in previously reported research. Additionally, suggestions to heighten mobility mechanisms by using mobile robots to be more practical, easy and inexpensive are also included in this paper, as we unveils ToMRobot, a low-cost MANET robot created from an ordinary remote control car that is capable of performing a real system MANET testbed with the addition of only a few low-cost electronic components. Despite greatly reduced costs, the ToMRobot does not sacrifice any of the necessary MANET basic structures and will still be easily customizable and upgradeable through the use of open hardware technology like Cubieboard2 and Arduino, as its robot controller. This paper will also include guidelines to enable technically limited MANET researchers to design and develop the ToMRobot. It is hoped that this paper achieves its two pronged objectives namely (i) to facilitate other MANET researchers by providing them with a source of reference that eases their decision making for selecting the best and most suitable MANET mobile robots for real mobility in their MANET testbeds (ii) to provide MANET researchers with a prospect of building their own MANET robots that can be applied in their own MANET testbed in the future
