    Assay of Therapeutic Doses of Methotrexate in Body Fluids of Patients with Psoriasis

    A microbiologic technique for the assay of methotrexate (MTX) in urine, serum, erythrocytes, feces, and skin is described. The accuracy of the method equals that of routine microbiologic assays of folic acid. Important differences in serum MTX levels in psoriatic patients during the 24 hr after standardized intravenous and intramuscular administration were demonstrated. Repeated intravenous doses tended to be cleared from the blood uniformly.After oral doses many patients achieved peak serum levels within 2 hr, with fall of level by 4 hr. Others achieved lower levels and responded less well clinically. Persistence of high serum levels at 24 and 48 hr did not confer obvious clinical benefit or necessarily give rise to toxicity. The pattern of clearance from serum was not influenced by serum albumin levels or by renal function when the creatinine clearance was greater than 50 mI/min. However, impaired renal function was clearly correlated with slow clearance.Routine measurement of MTX blood levels is of value in patients with suspected malabsorption or partial renal failure

    Multivariate climate reconstruction for the last 14,000 years in southernmost South America

    Comparison between past changes in pollen assemblages and stable isotope ratios (deuterium and carbon) analyzed in the same peat core from Tierra del Fuego at latitude 55°S permitted identification of the relative contribution of precipitation versus temperature responsible for the respective change. Major steps in the sequence of paleoenvironmental changes, such as at 12700, 9000, 5000, and 4000 years ago are apparently related only to increase in precipitation, reflecting the latitudinal location and intensity of the westerly storm tracks. On the other hand, high paleoenvironmental variability, which is characteristic for the late-glacial and the latest Holocene, is related to temperature variability, which affects the relative moisture content. Comparison with other paleoenvironmental records suggests that the late-glacial temperature variability is probably related to variability in the extent of Antarctic sea-ice, which in turn appears to be related to the intensity of Atlantic deep-water circulation. Temperature variability during the latest Holocene, on the other hand, is probably related to the dynamics of the El Niño/Southern Oscillation

    A 700 year record of Southern Hemisphere extratropical climate variability

    Annually dated ice cores from West and East Antarctica provide proxies for past changes in atmospheric circulation over Antarctica and portions of the Southern Ocean, temperature in coastal West and East Antarctica, and the frequency of South Polar penetration of El Niño events. During the period AD 1700–1850, atmospheric circulation over the Antarctic and at least portions of the Southern Hemisphere underwent a mode switch departing from the out-of-phase alternation of multi-decadal long phases of EOF1 and EOF2 modes of the 850 hPa field over the Southern Hemisphere (as defined in the recent record by Thompson and Wallace, 2000; Thompson and Solomon, 2002) that characterizes the remainder of the 700 year long record. From AD 1700 to 1850, lower-tropospheric circulation was replaced by in-phase behavior of the Amundsen Sea Low component of EOF2 and the East Antarctic High component of EOF1. During the first phase of the mode switch, both West and East Antarctic temperatures declined, potentially in response to the increased extent of sea ice surrounding both regions. At the end of the mode switch, West Antarctic coastal temperatures rose and East Antarctic coastal temperatures fell, respectively, to their second highest and lowest of the record. Polar penetration of El Niño events increased during the mode switch. The onset of the AD 1700–1850 mode switch coincides with the extreme state of the Maunder Minimum in solar variability. Late 20th-century West Antarctic coastal temperatures are the highest in the record period, and East Antarctic coastal temperatures close to the lowest.     A Record of Atmospheric Co2 During the Last 40,000 Years from the Siple Dome, Antarctica Ice Core

    We have measured the CO2 concentration of air occluded during the last 40,000 years in the deep Siple Dome A ( hereafter Siple Dome) ice core, Antarctica. The general trend of CO2 concentration from Siple Dome ice follows the temperature inferred from the isotopic composition of the ice and is mostly in agreement with other Antarctic ice core CO2 records. CO2 rose initially at similar to 17.5 kyr B. P. ( thousand years before 1950), decreased slowly during the Antarctic Cold Reversal, rose during the Younger Dryas, fell to a local minimum at around 8 kyr B. P., and rose continuously since then. The CO2 concentration never reached steady state during the Holocene, as also found in the Taylor Dome and EPICA Dome C ( hereafter Dome C) records. During the last glacial termination, a lag of CO2 versus Siple Dome isotopic temperature is probable. The Siple Dome CO2 concentrations during the last glacial termination and in the Holocene are at certain times greater than in other Antarctic ice cores by up to 20 ppm (mumol CO2/mol air). While in situ production of CO2 is one possible cause of the sporadic elevated levels, the mechanism leading to the enrichment is not yet clear

    A series of new radio-continuum (lambda=20 cm) mosaic images focused on the NGC 300 galactic system were produced using archived observational data from the VLA and/or ATCA. These new images are both very sensitive (rms=60 microJy) and feature high angular resolution (<10"). The most prominent new feature is the galaxy's extended radio-continuum emission, which does not match its optical appearance. Using these newly created images a number of previously unidentified discrete sources have been discovered. Furthermore, we demonstrate that a joint deconvolution approach to imaging this complete data-set is inferior when compared to an immerge approach.Comment: 13 pages, 12 figures, accepted to APSS, new version to correct the missing reference

    Seasonal Variations of Glaciochemical, Isotopic and Stratigraphic Properties in Siple Dome (Antarctica) Surface Snow

    Six snow-pit records recovered from Siple Dome, West Antarctica, during 1994 are used to study seasonal variations in chemical (major ion and H2O2), isotopic (deuterium) and physical stratigraphic properties during the 1988-94 period. Comparison of dD measurements and satellite-derived brightness temperature for the Siple Dome area suggests that most seasonal dD maxima occur within ±4 weeks of each 1 January. Several other chemical species (H2O2, non-sea-salt (nss) SO42-, methanesulfonic acid and NO3-) show coeval peaks with dD, together providing an accurate method for identifying summer accumulation. Sea-salt-derived species generally peak during winter/spring, but episodic input is noted throughout some years. No reliable seasonal signal is identified in species with continental sources (nssCa2+, nssMg2+), NH4+ or nssCl-. Visible strata such as large depth-hoar layers (\u3e5 cm) are associated with summer accumulation and its metamorphosis, but smaller hoar layers and crusts are more difficult to interpret. A multi-parameter approach is found to provide the most accurate dating of these snow-pit records, and is used to determine annual layer thicknesses at each site. Significant spatial accumulation variability exists on an annual basis, but mean accumulation in the sampled 10 km2 grid for the 1988-94 period is fairly uniform

    Residual cognitive deficits 50 years after lead poisoning during childhood

    The long term neurobehavioural consequences of childhood lead poisoning are not known. In this study adult subjects with a documented history of lead poisoning before age 4 and matched controls were examined with an abbreviated battery of neuropsychological tests including measures of attention, reasoning, memory, motor speed, and current mood. The subjects exposed to lead were inferior to controls on almost all of the cognitive tasks. This pattern of widespread deficits resembles that found in children evaluated at the time of acute exposure to lead rather than the more circumscribed pattern typically seen in adults exposed to lead. Despite having completed as many years of schooling as controls, the subjects exposed to lead were lower in lifetime occupational status. Within the exposed group, performance on the neuropsychological battery and occupational status were related, consistent with the presumed impact of limitations in neuropsychological functioning on everyday life. The results suggest that many subjects exposed to lead suffered acute encephalopathy in childhood which resolved into a chronic subclinical encephalopathy with associated cognitive dysfunction still evident in adulthood. These findings lend support to efforts to limit exposure to lead in childhood

    Farmland Prices: Is This Time Different?

    The historical behavior of farmland prices, rental rates, and rates of return are examined by treating farmland as an asset with an infinitely long life. It is found that high (low) farmland prices relative to rents have historically preceded extended periods of low (high) net rates of return, rather than greater (smaller) growth in rents. Our analysis shows that this attribute is shared with stocks and housing, and the financial literature provides ample evidence that other assets feature it as well. The long-run relationship linking farmland prices, rents, and rates of return is analyzed. Based on this relationship, we conclude that recent trends are unlikely to be sustainable. The study explores the expected paths that farmland prices and rates of return might follow if they were to eventually conform to the average values observed in the historical sample, and concludes with a discussion of the policy implications. Recommendations for policy makers include close monitoring of farmland lending practices and institutions to allow early identification of potential problems, and identifying in advance appropriate interventions in case recent farmland market trends were to suddenly change
