    Lead shielding efficiency from the gamma background measurements in the salt cavern of the Polkowice–Sieroszowice copper mine

    The studies of lead shielding efficiency from the gamma background measurements were performed in the salt cavern of the copper mine - a site considered for an underground laboratory. Within the energy range of 50–2700 keV, the measured gamma-ray count rates normalized to the mass of the high-purity detectors germanium crystal are 5.93 and 6.32 s−1kg−1 for the used low-background and portable spectrometers, respectively. The gamma-ray flux of 0.124 (2) cm−2s−1 connected with the natural radioisotopes was observed by the portable HPGe, including 0.026 (1) cm−2s−1 contribution of radon decay products, whereas the photon flux at the spectrum continuum was 0.18 (5) cm−2s−1

    Objects of humour: the puppet as comic performer

    Many of the studies that explore the fascination audiences have with puppets have focused largely on the relationship between the operator and the object and the illusion engendered through performance. Those that attend to the issue of humour, such as Dina and Joel Sherzer’s Humour and Comedy in Puppetry in 1987, tend to address generic comic components of specific puppet practices, and only minimally engage with the more fundamental concerns about how the object may be viewed humorously by audiences. This article intends to bridge this gap in scholarship by exploring the similarities between spectatorship and humour in relation to puppet practices. Drawing links between the incongruities inherent within puppet forms, particularly those revealed through the juxtaposition of object and human operator, and theories of humour, I argue that there is amusement to be found in seeing the inanimate animated, which is similar to the pleasure found in incongruous humour. While not all puppets are used for comic purposes, my argument suggests that the fundamental collaboration required for an audience to appreciate a puppet performance lends the form a particular comic specialism which may help explain why, historically, puppets appear to thrive in comic contexts

    Regulation of DNA Methylation Patterns by CK2-Mediated Phosphorylation of Dnmt3a

    DNA methylation is a central epigenetic modification that is established by de novo DNA methyltransferases. The mechanisms underlying the generation of genomic methylation patterns are still poorly understood. Using mass spectrometry and a phosphospecific Dnmt3a antibody, we demonstrate that CK2 phosphorylates endogenous Dnmt3a at two key residues located near its PWWP domain, thereby downregulating the ability of Dnmt3a to methylate DNA. Genome-wide DNA methylation analysis shows that CK2 primarily modulates CpG methylation of several repeats, most notably of Alu SINEs. This modulation can be directly attributed to CK2-mediated phosphorylation of Dnmt3a. We also find that CK2-mediated phosphorylation is required for localization of Dnmt3a to heterochromatin. By revealing phosphorylation as a mode of regulation of de novo DNA methyltransferase function and by uncovering a mechanism for the regulation of methylation at repetitive elements, our results shed light on the origin of DNA methylation patterns

    Fuzzy oil drop model to interpret the structure of antifreeze proteins and their mutants

    Mutations in proteins introduce structural changes and influence biological activity: the specific effects depend on the location of the mutation. The simple method proposed in the present paper is based on a two-step model of in silico protein folding. The structure of the first intermediate is assumed to be determined solely by backbone conformation. The structure of the second one is assumed to be determined by the presence of a hydrophobic center. The comparable structural analysis of the set of mutants is performed to identify the mutant-induced structural changes. The changes of the hydrophobic core organization measured by the divergence entropy allows quantitative comparison estimating the relative structural changes upon mutation. The set of antifreeze proteins, which appeared to represent the hydrophobic core structure accordant with “fuzzy oil drop” model was selected for analysis
