    Introduction to the special issue on Antarctic oceanography in a changing world

The definitive version was published in Oceanography 25, no. 3 (2012): 14-17, doi:10.5670/oceanog.2012.68."Antarctic Oceanography in a Changing World" commemorates the twentieth anniversary of the commissioning of Research Vessel Icebreaker (RVIB) Nathaniel B. Palmer and the fifteenth anniversary of Antarctic Research and Supply Vessel (ARSV) Laurence M. Gould. The addition of these two Antarctic research vessels to the US fleet in the 1990s ushered in a new era of Antarctic oceanographic research for US scientists and their international collaborators. Although several US Coast Guard icebreakers in the Arctic and Antarctic waters conduct oceanographic research, their primary mission is icebreaking to facilitate access to land-based stations. The Palmer was, and remains to this day, the first and only purpose-built US research icebreaker in Antarctic service and has been serving sea-going scientists in all areas of Antarctica's seas for two decades. The Gould has afforded reliable year-round access to Palmer Station and has conducted oceanographic research in the Antarctic Peninsula area since 1997

    Metagenomics of pigmented and cholesterol gallstones: the putative role of bacteria

    There is growing evidence for bacteria playing a role in the pathogenesis and formation of pigmented gallstones from humans. These studies mainly involved cultivation of gallstone-associated bacteria and 16S rRNA profiling, providing an indirect link between processes involved in gallstone formation by the bacteria in-situ. Here, we provide functional metagenomic evidence of a range of genes involved in bile stress response, biofilm formation, and anaerobic energy metabolism by Gram-negative Klebsiella in pigmented gallstones from a 76-year-old male patient. Klebsiella was also present in one cholesterol-type stone in a 30-year-old female patient who had additional cholesterol gallstones characterised by Gram-positive bacteria. Pigmented stones further revealed a predominance of genes involved in carbohydrate metabolism, whilst cholesterol stones indicated a profile dominanted by protein metabolism possibly reflecting known chemical differences between Gram-negative and Gram-positive biofilm matrices. Archaeal genes were not detected. Complementary carbon and hydrogen isotopic analyses of cholesterol within the patients' stones revealed homogeneity, suggesting a common diet or cholesterol biosynthesis pathway that has little influence on microbial composition. This pilot study provides a framework to study microbial processes that play a potential role in gallstone formation across markedly different types of stones and patient backgrounds

    A method to localize gamma-ray bursts using POLAR

    The hard X-ray polarimeter POLAR aims to measure the linear polarization of the 50-500 keV photons arriving from the prompt emission of gamma-ray bursts (GRBs). The position in the sky of the detected GRBs is needed to determine their level of polarization. We present here a method by which, despite of the polarimeter incapability of taking images, GRBs can be roughly localized using POLAR alone. For this purpose scalers are attached to the output of the 25 multi-anode photomultipliers (MAPMs) that collect the light from the POLAR scintillator target. Each scaler measures how many GRB photons produce at least one energy deposition above 50 keV in the corresponding MAPM. Simulations show that the relative outputs of the 25 scalers depend on the GRB position. A database of very strong GRBs simulated at 10201 positions has been produced. When a GRB is detected, its location is calculated searching the minimum of the chi2 obtained in the comparison between the measured scaler pattern and the database. This GRB localization technique brings enough accuracy so that the error transmitted to the 100% modulation factor is kept below 10% for GRBs with fluence Ftot \geq 10^(-5) erg cm^(-2) . The POLAR localization capability will be useful for those cases where no other instruments are simultaneously observing the same field of view.Comment: 13 pages, 10 figure

    Neoglacial climate anomalies and the Harappan metamorphosis

    Climate exerted constraints on the growth and decline of past human societies but our knowledge of temporal and spatial climatic patterns is often too restricted to address causal connections. At a global scale, the inter-hemispheric thermal balance provides an emergent framework for understanding regional Holocene climate variability. As the thermal balance adjusted to gradual changes in the seasonality of insolation, the Intertropical Convergence Zone migrated southward accompanied by a weakening of the Indian summer monsoon. Superimposed on this trend, anomalies such as the Little Ice Age point to asymmetric changes in the extratropics of either hemisphere. Here we present a reconstruction of the Indian winter monsoon in the Arabian Sea for the last 6000 years based on paleobiological records in sediments from the continental margin of Pakistan at two levels of ecological complexity: Sedimentary ancient DNA reflecting water column environmental states and planktonic foraminifers sensitive to winter conditions. We show that strong winter monsoons between ca. 4500 and 3000 years ago occurred during a period characterized by a series of weak interhemispheric temperature contrast intervals, which we identify as the early neoglacial anomalies (ENA). The strong winter monsoons during ENA were accompanied by changes in wind and precipitation patterns that are particularly evident across the eastern Northern Hemisphere and tropics. This coordinated climate reorganization may have helped trigger the metamorphosis of the urban Harappan civilization into a rural society through a push-pull migration from summer flood-deficient river valleys to the Himalayan piedmont plains with augmented winter rains. The decline in the winter monsoon between 3300 and 3000 years ago at the end of ENA could have played a role in the demise of the rural late Harappans during that time as the first Iron Age culture established itself on the Ghaggar-Hakra interfluve. Finally, we speculate that time-transgressive land cover changes due to aridification of the tropics may have led to a generalized instability of the global climate during ENA at the transition from the warmer Holocene thermal maximum to the cooler Neoglacial

    Biodegradation of textile waste by marine bacterial communities enhanced by light

    Knowledge of biofilm formation on pollutants in the marine realm is expanding, but how communities respond to substrates during colonization remains poorly understood. Here, we assess community assembly and respiration in response to two different micropollutants, virgin high‐density polyethylene (HDPE) microbeads and textile fibres under different light settings. Raman characterization, high‐throughput DNA sequencing data, quantitative PCR, and respiration measurements reveal how a stimulation of aerobic respiration by micropollutants is translated into selection for significantly different communities colonizing the substrates. Despite the lack of evidence for biodegradation of HDPE, an increased abundance and respiration of bacterial taxa closely related to hydrocarbonoclastic Kordiimonas spp. and Alteromonas spp. in the presence of textile waste highlights their biodegradation potential. Incubations with textile fibres exhibited significantly higher respiration rates in the presence of light, which could be partially explained by photochemical dissolution of the textile waste into smaller bioavailable compounds. Our results suggest that the development and increased respiration of these unique microbial communities may potentially play a role in the bioremediation of the relatively long‐lived textile pollutants in marine habitats, and that the respiration of heterotrophic hydrocarbon‐degrading bacteria colonizing marine pollutants can be stimulated by light

    Arroz: ensaio fatorial variedade x espaçamento x densidade

    In this experiment it was attempted to find better row spacing (0,20 m, 0,40 m and 0,60 m) and seed rate (3 and 6 grams of seeds/m) to be used in rice. The ordinary flooding was used as irrigation. Four varieties with different flowering periods were used: "Pratão" and "Iguape Agulha" are late varieties (150 days); "Batatais", "Dourado Precoce" early varieties (100 days). These two early varieties produce two harvests by ratooning. The data showed that the late varieties gave a better yeld on a single crop, but the greatest annual yeld by area was obtained when the ratooning was used. As far as amount of seed is concerned the data showed that the better yelds were obtained with 3 grams of seeds

    Influência do peso da semente de arroz (Oryza sativa L.) sobre a germinação

    The main purpose of this work was to evaluate, through germination tests, the importance of the weight on rice seeds (Orza sativa L.). Batatais, Pratão Precoce. IAC-120, IAC-435 and IAC-1246 varieties were tested. The trial was conducted in the Seed Laboratory of the "Departamento de Agricultura e Horticultura da Escola Superior de Aricultura Luiz de Queiroz", from November, 1973. to December, 1974. Seeds of each variety was submited to a controled ventilation action through a blower, allowing the separation of each variety in to three fractions, with different weights, that is, light, median and heavy. The light, median and heavy seeds obtained were stored indry chamber and in laboratory. The germination tost were done in three periods (March/April, July and December, 1974), using as experimental design a factorial 3x2 whose variables were three weights and two environments of seeds conservation with four replications. Data analyses and the interpretation of the obtained results allowed us to draw the following conclusions: a) the heavy seeds and the median weight ones presented a higher percentage of germination than the light seeds in all studied varieties; b) this effect was less intense in the case of the Batatais and IAC-1246 varieties; c) the dry chamber environment showed superior to the laboratory for he conservation of the seeds.O objetivo principal do presente trabalho foi determinar através do teste de germinação, a importância do peso das sementes de arroz (Oryza sativa L.). Utilizaram-se sementes das variedades Batatais, Pratão Precoce, IAC-120, IAC-435 e IAC-1246. O trabalho foi conduzido no Laboratório de Sementes do D.A.H. da ESALQ no período de novembro de 1973 a dezembro de 1974. As sementes das cinco variedades foram submetidas à ação de ventilação controlada através de um assoprador, permitindo a separação de cada variedade em três frações com pesos diferentes, isto é, leve, média e pesada. As sementes leves, médias e pesadas, assim obtidas, foram armazenadas em câmara seca e laboratório. Os testes de germinação foram efetuados em três épocas (março/abril, julho e dezembro de 1974), utilizando-se como delineamento experimental um fatorial 3x2, cujas variáveis foram três pesos e dois ambientes de conservação das sementes, com quatro repetições. As análises dos dados e a interpretação dos resultados obtidos permitiram que se chegassem às seguintes conclusões: a) as sementes pesadas e médias apresentaram uma maior porcentagem de germinação que as leves em todas as variedades estudadas; b) este efeito foi menos intenso no caso das sementes das variedades Batatais e IAC-1246; c) o ambiente de câmara seca mostrou-se superior ao de laboratório para a conservação das sementes

    Atribacteria reproducing over millions of years in the Atlantic abyssal subseafloor

Mbio, 11(5), (2020): e01937-20, doi:10.1128/mBio.01937-20.How microbial metabolism is translated into cellular reproduction under energy-limited settings below the seafloor over long timescales is poorly understood. Here, we show that microbial abundance increases an order of magnitude over a 5 million-year-long sequence in anoxic subseafloor clay of the abyssal North Atlantic Ocean. This increase in biomass correlated with an increased number of transcribed protein-encoding genes that included those involved in cytokinesis, demonstrating that active microbial reproduction outpaces cell death in these ancient sediments. Metagenomes, metatranscriptomes, and 16S rRNA gene sequencing all show that the actively reproducing community was dominated by the candidate phylum “Candidatus Atribacteria,” which exhibited patterns of gene expression consistent with fermentative, and potentially acetogenic, metabolism. “Ca. Atribacteria” dominated throughout the 8 million-year-old cored sequence, despite the detection limit for gene expression being reached in 5 million-year-old sediments. The subseafloor reproducing “Ca. Atribacteria” also expressed genes encoding a bacterial microcompartment that has potential to assist in secondary fermentation by recycling aldehydes and, thereby, harness additional power to reduce ferredoxin and NAD+. Expression of genes encoding the Rnf complex for generation of chemiosmotic ATP synthesis were also detected from the subseafloor “Ca. Atribacteria,” as well as the Wood-Ljungdahl pathway that could potentially have an anabolic or catabolic function. The correlation of this metabolism with cytokinesis gene expression and a net increase in biomass over the million-year-old sampled interval indicates that the “Ca. Atribacteria” can perform the necessary catabolic and anabolic functions necessary for cellular reproduction, even under energy limitation in millions-of-years-old anoxic sediments.This work was supported primarily by the Deutsche Forschungsgemeinschaft (DFG) project OR 417/1-1 granted to W.D.O.
