No evidence for an effect of testosterone administration on delay discounting in male university students
SummaryIntertemporal choices between a smaller sooner and a larger delayed reward are one of the most important types of decisions humans face in their everyday life. The degree to which individuals discount delayed rewards correlates with impulsiveness. Steep delay discounting has been associated with negative outcomes over a wide range of behaviors such as addiction. However, little is known about the biological foundations of delay discounting. Here, we examine a potential causal link between delay discounting and testosterone, a hormone which has been associated with other types of impulsive behavior. In our double-blind placebo-controlled study 91 healthy young men either received a topical gel containing 50mg of testosterone (N=46) or a placebo (N=45) before participating in a delay discounting task with real incentives. Our main finding is that a single dose administration of testosterone did not lead to significant differences in discount rates between the placebo and the testosterone group. Within groups and in the pooled sample, no significant relationship between testosterone and discount rates was observed. At the same time, we do replicate standard findings from the delay discounting literature such as a magnitude-of-rewards effect on discount rates. In sum, our findings suggest that circulating testosterone does not have a significant effect on delay discounting in young men
Prognostic Significance of Erythropoietin in Pancreatic Adenocarcinoma
BACKGROUND: Erythropoietin (Epo) administration has been reported to have tumor-promoting effects in anemic cancer patients. We investigated the prognostic impact of endogenous Epo in patients with pancreatic ductal adenocarcinoma (PDAC). METHODOLOGY: The clinico-pathological relevance of hemoglobin (Hb, n = 150), serum Epo (sEpo, n = 87) and tissue expression of Epo/Epo receptor (EpoR, n = 104) was analyzed in patients with PDAC. Epo/EpoR expression, signaling, growth, invasion and chemoresistance were studied in Epo-exposed PDAC cell lines. RESULTS: Compared to donors, median preoperative Hb levels were reduced by 15% in both chronic pancreatitis (CP, p<0.05) and PDAC (p<0.001), reaching anemic grade in one third of patients. While inversely correlating to Hb (r = -0.46), 95% of sEPO values lay within the normal range. The individual levels of compensation were adequate in CP (observed to predicted ratio, O/P = 0.99) but not in PDAC (O/P = 0.85). Strikingly, lower sEPO values yielding inadequate Epo responses were prominent in non-metastatic M0-patients, whereas these parameters were restored in metastatic M1-group (8 vs. 13 mU/mL; O/P = 0.82 vs. 0.96; p<0.01)--although Hb levels and the prevalence of anemia were comparable. Higher sEpo values (upper quartile ≥ 16 mU/ml) were not significantly different in M0 (20%) and M1 (30%) groups, but were an independent prognostic factor for shorter survival (HR 2.20, 10 vs. 17 months, p<0.05). The pattern of Epo expression in pancreas and liver suggested ectopic release of Epo by capillaries/vasa vasorum and hepatocytes, regulated by but not emanating from tumor cells. Epo could initiate PI3K/Akt signaling via EpoR in PDAC cells but failed to alter their functions, probably due to co-expression of the soluble EpoR isoform, known to antagonize Epo. CONCLUSION/SIGNIFICANCE: Higher sEPO levels counteract anemia but worsen outcome in PDAC patients. Further trials are required to clarify how overcoming a sEPO threshold ≥16 mU/ml by endogenous or exogenous means may predispose to or promote metastatic progression
Thermal corrections to the Casimir effect
The Casimir effect, reflecting quantum vacuum fluctuations in the
electromagnetic field in a region with material boundaries, has been studied
both theoretically and experimentally since 1948. The forces between dielectric
and metallic surfaces both plane and curved have been measured at the 10 to 1
percent level in a variety of room-temperature experiments, and remarkable
agreement with the zero-temperature theory has been achieved. In fitting the
data various corrections due to surface roughness, patch potentials, curvature,
and temperature have been incorporated. It is the latter that is the subject of
the present article. We point out that, in fact, no temperature dependence has
yet been detected, and that the experimental situation is still too fluid to
permit conclusions about thermal corrections to the Casimir effect.
Theoretically, there are subtle issues concerning thermodynamics and
electrodynamics which have resulted in disparate predictions concerning the
nature of these corrections. However, a general consensus has seemed to emerge
that suggests that the temperature correction to the Casimir effect is
relatively large, and should be observable in future experiments involving

Casimir forces and non-Newtonian gravitation
The search for non-relativistic deviations from Newtonian gravitation can
lead to new phenomena signalling the unification of gravity with the other
fundamental interactions. Various recent theoretical frameworks indicate a
possible window for non-Newtonian forces with gravitational coupling strength
in the micrometre range. The major expected background in the same range is
attributable to the Casimir force or variants of it if dielectric materials,
rather than conducting ones, are considered. Here we review the measurements of
the Casimir force performed so far in the micrometre range and how they
determine constraints on non-Newtonian gravitation, also discussing the
dominant sources of false signals. We also propose a geometry-independent
parameterization of all data in terms of the measurement of the constant c. Any
Casimir force measurement should lead, once all corrections are taken into
account, to a determination of the constant c which, in order to assess the
accuracy of the measurement, can be compared with its more precise value known
through microscopic measurements. Although the last decade of experiments has
resulted in solid demonstrations of the Casimir force, the situation is not
conclusive with respect to being able to discover new physics. Future
experiments and novel phenomenological analysis will be necessary to discover
non-Newtonian forces or to push the window for their possible existence into

Knowledge-based matrix factorization temporally resolves the cellular responses to IL-6 stimulation
<p>Abstract</p> <p>Background</p> <p>External stimulations of cells by hormones, cytokines or growth factors activate signal transduction pathways that subsequently induce a re-arrangement of cellular gene expression. The analysis of such changes is complicated, as they consist of multi-layered temporal responses. While classical analyses based on clustering or gene set enrichment only partly reveal this information, matrix factorization techniques are well suited for a detailed temporal analysis. In signal processing, factorization techniques incorporating data properties like spatial and temporal correlation structure have shown to be robust and computationally efficient. However, such correlation-based methods have so far not be applied in bioinformatics, because large scale biological data rarely imply a natural order that allows the definition of a delayed correlation function.</p> <p>Results</p> <p>We therefore develop the concept of graph-decorrelation. We encode prior knowledge like transcriptional regulation, protein interactions or metabolic pathways in a weighted directed graph. By linking features along this underlying graph, we introduce a partial ordering of the features (e.g. genes) and are thus able to define a graph-delayed correlation function. Using this framework as constraint to the matrix factorization task allows us to set up the fast and robust graph-decorrelation algorithm (GraDe). To analyze alterations in the gene response in <it>IL-6 </it>stimulated primary mouse hepatocytes, we performed a time-course microarray experiment and applied GraDe. In contrast to standard techniques, the extracted time-resolved gene expression profiles showed that <it>IL-6 </it>activates genes involved in cell cycle progression and cell division. Genes linked to metabolic and apoptotic processes are down-regulated indicating that <it>IL-6 </it>mediated priming renders hepatocytes more responsive towards cell proliferation and reduces expenditures for the energy metabolism.</p> <p>Conclusions</p> <p>GraDe provides a novel framework for the decomposition of large-scale 'omics' data. We were able to show that including prior knowledge into the separation task leads to a much more structured and detailed separation of the time-dependent responses upon <it>IL-6 </it>stimulation compared to standard methods.
