    NOTCH blockade combined with radiation therapy and temozolomide prolongs survival of orthotopic glioblastoma.

    Glioblastoma multiforme (GBM) is the most common malignant brain tumor in adults. The current standard of care includes surgery followed by radiotherapy (RT) and chemotherapy with temozolomide (TMZ). Treatment often fails due to the radiation resistance and intrinsic or acquired TMZ resistance of a small percentage of cells with stem cell-like behavior (CSC). The NOTCH signaling pathway is expressed and active in human glioblastoma and NOTCH inhibitors attenuate tumor growth in vivo in xenograft models. Here we show using an image guided micro-CT and precision radiotherapy platform that a combination of the clinically approved NOTCH/Îł-secretase inhibitor (GSI) RO4929097 with standard of care (TMZ + RT) reduces tumor growth and prolongs survival compared to dual combinations. We show that GSI in combination with RT and TMZ attenuates proliferation, decreases 3D spheroid growth and results into a marked reduction in clonogenic survival in primary and established glioma cell lines. We found that the glioma stem cell marker CD133, SOX2 and Nestin were reduced following combination treatments and NOTCH inhibitors albeit in a different manner. These findings indicate that NOTCH inhibition combined with standard of care treatment has an anti-glioma stem cell effect which provides an improved survival benefit for GBM and encourages further translational and clinical studies

    The Association between Primary Endometrioid Carcinoma of the Ovary and Synchronous Malignancy of the Endometrium

    Objective. Ovarian and endometrial cancers coincide rather frequently in the same patient. Few data are available on the involvement of the specific morphological subtypes. To identify histological pathways in the synchronous occurrence, a population-based study was performed in The Netherlands. Methods. Using the national pathology database (PALGA) information of ovarian cancers and of earlier or later cancer in the endometrium was obtained. 5366 Patients were identified with primary malignant epithelial or borderline malignancy. Results. In 157 cases (2.9%) a new primary malignancy in the endometrium was diagnosed (146 within 1 year). The ratio of observed versus expected number of synchronous malignancy in the endometrium was estimated at 3.6 (95% CI: 2.7–4.7). Among 460 ovarian endometrioid carcinoma patients 53 cases showed a second primary endometrial cancer; 40 out of these 53 cases (75.5%) showed at both organ sites an endometrioid adenocarcinoma. Conclusion. These findings suggest an important role for the endometrioid subtype and prompt to mechanism-based studies incorporating molecular techniques

    Surface coating and particle size are main factors explaining the transcriptome-wide responses of the earthworm Lumbricus rubellus to silver nanoparticles

    Due to the unique properties of differently sized and coated silver nanoparticles (AgNPs), they are used in important industrial and biomedical applications. However, their environmental fate in soil ecosystems and potential mechanisms of toxicity remain elusive, especially at the level of transcriptional regulation. We investigated the transcriptome-wide responses of the earthworm Lumbricus rubellus exposed to nine AgNPs differing in surface coating/charge (bovine serum albumin/negative AgNP_BSA, chitosan/positive AgNP_Chit, and polyvinylpyrrolidone/neutral AgNP_PVP) and sizes (20, 35 and 50 nm) at concentrations close to the EC50 value related to reproduction. AgNO3 was used in two concentrations to benchmark the AgNP effects against those of the Ag salt. A correlation was observed between the number of differentially expressed genes (DEGs) and Ag internal body concentration. Only metallothionein was regulated by all treatments. Medium sized AgNPs caused the most pronounced transcriptional responses, while AgNO3 affected the transcriptome less. Medium sized AgNP_BSA exposure caused the most extensive transcriptional responses with 684 DEGs. Gene ontology enrichment analysis of medium sized AgNP_BSA affected DEGs revealed that mitochondrial electron transport, autophagy and phagocytosis, mesoderm and heart development and microtubule organisation were affected. This was also confirmed by gene set enrichment for KEGG pathway analysis, indicating that phagocytosis, autophagy and signalling pathways related to mesoderm formation were significantly up regulated. All AgNP_BSA and AgNP_PVP exposures caused severe down regulation of ribosomal translation, suggesting that the high energy-demanding protein synthesis process is inhibited. Our data confirm the mechanisms previously identified among other animal models and human cell lines. To conclude, coating formulation and particle size severely impact transcriptional responses at a particular nanoparticle size, suggesting diverse mechanistic responses depending on the coating type

    The influence of a PHI-5-loaded silicone membrane, on cutaneous wound healing in vivo

    This study investigated whether a novel ionogenic substance, containing amongst others zinc and rubidium (PHI-5; Dermagenics Inc, Memphis, TN, USA), could improve the healing of full-thickness skin wounds. Uniform wounds were created on the right flank of guinea pigs. Micro-grooved silicone rubber membranes, containing 0 (controls), 1.25, 5.00, or 10.00 μg PHI-5, were sutured onto this wound. Standardized digital wound photographs were made after 1, 3, and 6 weeks. Also, wound biopsies were taken after 3 and 6 weeks for histological and histomorphometrical evaluation. For all study groups, 6 animals were used. Analysis of the 1-week digital photographs showed that the surface area of the wounds decreased significantly, with an increasing PHI-5 concentration. No other differences were found in the wound photographs. Also, no differences were measured in histomorphometry at 3 and 6 weeks. Concluding, in our study model a single application of PHI-5 did have a significant positive influence on initial wound healing
