    A tutorial on regression-based norming of psychological tests with GAMLSS

    A norm-referenced score expresses the position of an individual test taker in the reference population, thereby enabling a proper interpretation of the test score. Such normed scores are derived from test scores obtained from a sample of the reference population. Typically, multiple reference populations exist for a test, namely when the norm-referenced scores depend on individual characteristic(s), as age (and sex). To derive normed scores, regression-based norming has gained large popularity. The advantages of this method over traditional norming are its flexible nature, yielding potentially more realistic norms, and its efficiency, requiring potentially smaller sample sizes to achieve the same precision. In this tutorial, we introduce the reader to regression-based norming, using the generalized additive models for location, scale, and shape (GAMLSS). This approach has been useful in norm estimation of various psychological tests. We discuss the rationale of regression-based norming, theoretical properties of GAMLSS and their relationships to other regression-based norming models. Based on 6 steps, we describe how to: (a) design a normative study to gather proper normative sample data; (b) select a proper GAMLSS model for an empirical scale; (c) derive the desired normed scores for the scale from the fitted model, including those for a composite scale; and (d) visualize the results to achieve insight into the properties of the scale. Following these steps yields regression-based norms with GAMLSS for a psychological test, as we illustrate with normative data of the intelligence test IDS-2.     Bayesian Gaussian distributional regression models for more efficient norm estimation

    A test score on a psychological test is usually expressed as a normed score, representing its position relative to test scores in a reference population. These typically depend on predictor(s) such as age. The test score distribution conditional on predictors is estimated using regression, which may need large normative samples to estimate the relationships between the predictor(s) and the distribution characteristics properly. In this study, we examine to what extent this burden can be alleviated by using prior information in the estimation of new norms with Bayesian Gaussian distributional regression. In a simulation study, we investigate to what extent this norm estimation is more efficient and how robust it is to prior model deviations. We varied the prior type, prior misspecification and sample size. In our simulated conditions, using a fixed effects prior resulted in more efficient norm estimation than a weakly informative prior as long as the prior misspecification was not age dependent. With the proposed method and reasonable prior information, the same norm precision can be achieved with a smaller normative sample, at least in empirical problems similar to our simulated conditions. This may help test developers to achieve cost‐efficient high‐quality norms. The method is illustrated using empirical normative data from the IDS‐2 intelligence test

    Improving confidence intervals for normed test scores:Include uncertainty due to sampling variability

    Test publishers usually provide confidence intervals (CIs) for normed test scores that reflect the uncertainty due to the unreliability of the tests. The uncertainty due to sampling variability in the norming phase is ignored. To express uncertainty due to norming, we propose a flexible method that is applicable in continuous norming and allows for a variety of score distributions, using Generalized Additive Models for Location, Scale, and Shape (GAMLSS; Rigby & Stasinopoulos, 2005). We assessed the performance of this method in a simulation study, by examining the quality of the resulting CIs. We varied the population model, procedure of estimating the CI, confidence level, sample size, value of the predictor, extremity of the test score, and type of variance-covariance matrix. The results showed that good quality of the CIs could be achieved in most conditions. The method is illustrated using normative data of the SON-R 6-40 test. We recommend test developers to use this approach to arrive at CIs, and thus properly express the uncertainty due to norm sampling fluctuations, in the context of continuous norming. Adopting this approach will help (e.g., clinical) practitioners to obtain a fair picture of the person assessed

    Rolled-Up Nanotech: Illumination-Controlled Hydrofluoric Acid Etching of AlAs Sacrificial Layers

    <p>Abstract</p> <p>The effect of illumination on the hydrofluoric acid etching of AlAs sacrificial layers with systematically varied thicknesses in order to release and roll up InGaAs/GaAs bilayers was studied. For thicknesses of AlAs below 10 nm, there were two etching regimes for the area under illumination: one at low illumination intensities, in which the etching and releasing proceeds as expected and one at higher intensities in which the etching and any releasing are completely suppressed. The &#8220;etch suppression&#8221; area is well defined by the illumination spot, a feature that can be used to create heterogeneously etched regions with a high degree of control, shown here on patterned samples. Together with the studied self-limitation effect, the technique offers a way to determine the position of rolled-up micro- and nanotubes independently from the predefined lithographic pattern.</p

    The interaction between AMPK beta 2 and the PP1-targeting subunit R6 is dynamically regulated by intracellular glycogen content

    11 påginas, 7 figuras.AMP-activated protein kinase (AMPK) is a metabolic stress-sensing kinase. We previously showed that glucose deprivation induces autophosphorylation of AMPKβ at threonine-148 (Thr-148), which prevents the binding of AMPK to glycogen. Furthermore, in MIN6 cells, AMPKβ1 binds to R6 (PPP1R3D), a glycogen-targeting subunit of protein phosphatase 1 (PP1), thereby regulating the glucose-induced inactivation of AMPK. Here, we further investigated the interaction of R6 with AMPKβ and the possible dependency on Thr-148 phosphorylation status. Yeast two-hybrid analyses and co-immunoprecipitation of the overexpressed proteins in HEK293T cells revealed that both AMPKβ1 and β2 wild-type (WT) isoforms bind to R6. The AMPKβ/R6 interaction was stronger with the muscle-specific β2-WT and required association with the substrate-binding motif of R6. When HEK293T cells or C2C12 myotubes were cultured in high-glucose medium, AMPKβ2-WT and R6 weakly interacted. In contrast, glycogen depletion significantly enhanced this protein interaction. Mutation of AMPKβ2 Thr-148 prevented the interaction with R6 irrespective of the intracellular glycogen content. Treatment with the AMPK activator oligomycin enhanced AMPKβ2/R6 interaction in conjunction with increased Thr-148 phosphorylation in cells grown in low glucose medium. These data are in accordance with R6 binding directly to AMPKβ2 when both proteins detach from the diminishing glycogen particle, which is simultaneous to increased AMPKβ2 Thr-148 autophosphorylation.     Patterns of recurrence following definitive chemoradiation for patients with proximal esophageal cancer

    Introduction: The aim of this retrospective study was to determine the patterns of recurrence and overall survival (OS) in patients achieving clinical complete response after treatment with definitive chemoradiation (CRT) for proximal esophageal cancer. Materials and methods: Patients with proximal esophageal cancer treated with CRT between 2004 and 2014 in 11 centers in the Netherlands were included. OS and progression-free survival (PFS) were calculated using the Kaplan-Meier method. Cumulative incidence of first recurrence (locoregional or distant) and locoregional recurrence (LRR) were assessed using competing risk analyses. Results: In 197 of the 200 identified patients, response was evaluated, 133 (68%) showed a complete response. In complete responders, median OS, three-year OS, and PFS were 45.0 months (95% CI 34.8-61.5 months), 58% (95% CI 48-66), and 49% (95% CI 40-57), respectively. Three- and five-year risk of recurrence were respectively 40% (95% CI 31-48), and 45% (95% CI 36-54). Three- and five-year risk of LRR were 26% (95% CI 19-33), and 30% (95% CI 22-38). Eight of 32 patients with an isolated LRR underwent salvage surgery, with a median OS of 32.0 months (95% CI 6.8-not reached). Conclusion: In patients with a complete response after definitive CRT for proximal esophageal cancer, most recurrences were locoregional and developed within the first three years after CRT.     Modifying Threat-related Interpretive Bias in Adolescents

    Socially anxious feelings sharply increase during adolescence and such feelings have been associated with interpretive biases. Studies in adults have shown that interpretive biases can be modified using Cognitive Bias Modification procedures (CBM-I) and subsequent effects on anxiety have been observed. The current study was designed to examine whether the CBM-I procedure has similar effects in adolescents. Unselected adolescents were randomly allocated to either a positive interpretation training (n = 88) or a placebo-control condition (n = 82). Results revealed that the training was successful in modifying interpretations and effects generalized to a new task. The interpretive bias effects were most pronounced in individuals with a threat-related interpretive bias at pre-test. No effects on state anxiety were observed. The current findings are promising with regard to applying bias modification procedures to adolescents, while further research is warranted regarding emotional effects
