    ‘And yet I’m an adult now’. The influence of parental criticism on women’s body satisfaction/dissatisfaction during emerging adulthood

    Body dissatisfaction is widely recognized as an important public health concern mainly for women because of its increasing incidence worldwide. The aim of this exploratory study was to test the predictive role of family influence on body satisfaction (BS) among young Italian women, during a decade conceptualized as \u2018emerging adulthood\u2019. Instruments designed to assess BS, appearance-focused parental comments, maternal and paternal criticism were administered to 688 women aged between 18 and 28 years (M age = 23.40; SD = 2.33). Two regression analyses were used with life satisfaction levels, parental comments, and maternal/paternal criticism as independent variables to explain variance in BS for younger (18\u201323 years) and older (24\u201328 years) women. The data reveal an interesting difference in the influence of parental criticism on BS depending on the age cohort: maternal criticism negatively predicts BS in younger women, while paternal criticism is a risk factor for BS in older women

    Effect of a submaxillary gland extract on Ehrlich tumor growth in mice

    Ablation of host submaxillary glands modifies Ehrlich tumor growth and tumor-infiltrating leukocytes, possibly by modifications in the serum level of growth factors produced by this gland. To extend this research, 7-month-old male EPM-1 mice (N = 30) were divided into two groups: 1) inoculated with tumor cells previously incubated with submaxillary salivary gland extract (SGE) in PBS for 30 min at 37%; 2) inoculated with tumor cells previously incubated with PBS, under the same conditions. Animals were inoculated into the footpad with 40 µl of a suspension containing 4.5 x 107 tumor cells/ml, and footpad thickness was measured daily for 10 days. Sections and smears of tumor cells were prepared from the tumor mass to determine mitosis frequency, percent of tumor cells immunopositive to nerve (NGF) and epidermal (EGF) growth factors and percent of tumor-infiltrating leukocytes. The incubation of tumor cells with SGE produced a tumor reduction of about 30% in size (P<0.01). This effect was not related to loss of cell viability during incubation, but a 33% increase (P<0.05) in the percentage of dead or dying tumor cells and a 15% increase in the percent of NGF/EGF-positive tumor cells (P<0.01) were observed in vivo at the end of experiment. Tumor-infiltrating lymphocytes and mitosis frequency did not differ between groups.     Simplified reactor design for mixed culture-based electrofermentation toward butyric acid production

    Mixed microbial culture (MMC) electrofermentation (EF) represents a promising tool to drive metabolic pathways toward the production of a specific compound. Here, the MMC-EF process has been exploited to obtain butyric acid in simplified membrane-less reactors operated by applying a difference of potential between two low-cost graphite electrodes. Ten values of voltage difference, from -0.60 V to -1.5 V, have been tested and compared with the experiment under open circuit potential (OCP). In all the tested conditions, an enhancement in the production rate of butyric acid (from a synthetic mixture of glucose, acetate, and ethanol) was observed, ranging from 1.3- to 2.7-fold relative to the OCP. Smaller enhancements in the production rate resulted in higher values of the calculated specific energy consumption. However, at all applied voltages, a low flow of current was detected in the one-chamber reactors, accounting for an average value of approximately -100 µA. These results hold a substantial potential with respect to the scalability of the electrofermentation technology, since they pinpoint the possibility to control MMC-based bioprocesses by simply inserting polarized electrodes into traditional fermenters

    Analysis of cytosine methylation in genomic dna of solanum × michoacanum (+) s. tuberosum somatic hybrids

    Interspecific somatic hybridization is a noteworthy breeding strategy that allows the production of novel genetic variability when crossing barriers exist between two parental species. Although the genetic consequences of somatic hybridization have been well documented, little is known on its impact at the epigenetic level. The objective of our research was to investigate the epigenetic changes, in particular DNA methylation, occurring in a population of potato somatic hybrids. The analysis of 96 Solanum × michoacanum (+) S. tuberosum somatic hybrids from five fusion combinations and their parents was carried out by methylation-sensitive amplified polymorphism (MSAP) and high-performance liquid chromatography (HPLC) methods. Six MSAP primer combinations generated 622 unique bands, of which 295 were fully methylated. HPLC analysis showed from 15.5% to 16.9% total cytosine methylation within the parental forms. Overall, the MSAP and HPLC methods indicated an increase in DNA methylation in the somatic hybrids in comparison to their parents. Among the latter, a lower degree of DNA methylation in the wild S. × michoacanum species than S. tuberosum was found. Our findings indicated that somatic hybridization changed the level of cytosine methylation in the studied potato somatic hybrids

    SerpinB3 as hepatic marker of post-resective shear stress

    Post-resective liver failure is a frequent complication of liver surgery and it is due to portal hyperperfusion of the remnant liver and to arterial vasoconstriction, as buffer response of the hepatic artery. In this context, splenectomy allows a reduction of portal flow and increases the survival chance in preclinical models. SerpinB3 is over-expressed in the liver in oxidative stress conditions, as a mechanism of cell defense to provide survival by apoptosis inhibition and cell proliferation. In this study, the expression of SerpinB3 was assessed as predictor of liver damage in in vivo models of major hepatic resection with or without splenectomy. Wistar male rats were divided into 4 groups: group A received 30% hepatic resection, group B > 60% resection, group C > 60% resection with splenectomy and group D sham-operated. Before and after surgery liver function tests, echo Doppler ultrasound and gene expression were assessed. Transaminase values and ammonium were significantly higher in groups that underwent major hepatic resection. Echo Doppler ultrasound showed the highest portal flow and resistance of the hepatic artery in the group with > 60% hepatectomy without splenectomy, while the association of splenectomy determined no increase in portal flow and hepatic artery resistance. Only the group of rats without splenectomy showed higher shear-stress conditions, reflected by higher levels of HO-1, Nox1 and of Serpinb3, the latter associated with an increase of IL-6. In conclusion, splenectomy controls inflammation and oxidative damage, preventing the expression of Serpinb3. Therefore, SerpinB3 can be considered as a marker of post-resective shear stress

    Coupled Biological and Thermochemical Process for Plastic Waste Conversion into Biopolymers

    The aqueous phase produced from the hydrothermal liquefaction (HTL) of three matrices (Plasmix treated with different operative conditions and polystyrene) was subjected to acidogenic fermentation (AF) batch tests to obtain organic acids, which are the ideal substrates for biopolymers (e.g., polyhydroxyalkanoates, PHA) production from mixed microbial cultures (MMC). Parallel tests in the presence of only HTL water fractions or only glucose (an easily biodegradable compound), or in presence of both, were conducted and compared to assess any possible recalcitrant or inhibitory effect of plastic waste from the HTL treatment during the AF process. These tests resulted, within approximately 30 days of operation, in a conversion of 96 ± 21% (COD/COD) of the Plasmix by-products after a 2h thermochemical treatment into organic acids, a 54 ± 7% (COD/COD) of conversion for Plasmix by-products treated 4h, and 29 ± 1% (COD/COD) of conversion in the presence of polystyrene residual water

    Hypoxia up-regulates SERPINB3 through HIF-2\u3b1 in human liver cancer cells.

    SERPINB3 is a cysteine-proteases inhibitor up-regulated in a significant number of cirrhotic patients carrying hepatocellular carcinoma (HCC) and recently proposed as a prognostic marker for HCC early recurrence. SERPINB3 has been reported to stimulate proliferation, inhibit apoptosis and, similar to what reported for hypoxia, to trigger epithelial-to-mesenchymal transition (EMT) and increased invasiveness in liver cancer cells. This study has investigated whether SERPINB3 expression is regulated by hypoxia-related mechanisms in liver cancer cells. Exposure of HepG2 and Huh7 cells to hypoxia up-regulated SERPINB3 transcription, protein synthesis and release in the extracellular medium. Hypoxia-dependent SERPINB3 up-regulation was selective (no change detected for SERPINB4) and operated through hypoxia inducible factor (HIF)-2\u3b1 (not HIF-1\u3b1) binding to SERPINB3 promoter, as confirmed by chromatin immuno-precipitation assay and silencing experiments employing specific siRNAs. HIF-2\u3b1-mediated SERPINB3 up-regulation under hypoxic conditions required intracellular generation of ROS. Immuno-histochemistry (IHC) and transcript analysis, performed in human HCC specimens, revealed co-localization of the two proteins in liver cancer cells and the existence of a positive correlation between HIF-2\u3b1 and SERPINB3 transcript levels, respectively. Hypoxia, through HIF-2\u3b1-dependent and redox-sensitive mechanisms, up-regulates the transcription, synthesis and release of SERPINB3, a molecule with a high oncogenic potential

    Memory for spatial locations in a patient with near space neglect and optic ataxia: Involvement of the occipitotemporal stream

    Previous studies suggested that the occipitoparietal stream orients attention toward the near/lower space and is involved in immediate reaching, whereas the occipitotemporal stream orients attention toward the far/upper space and is involved in delayed reaching. In the present study, we investigated the role of the occipitotemporal stream in attention orienting and delayed reaching in a patient (GP) with bilateral damage to the occipitoparietal areas and optic ataxia. GP and healthy controls took part in three experiments. In the experiment 1, the participants bisected lines oriented along radial, vertical, and horizontal axes. GP bisected radial lines farther, and vertical lines more above, than the controls, consistent with an attentional bias toward the far/upper space and near/lower space neglect. The experiment 2 consisted of two tasks: (1) an immediate reaching task, in which GP reached target locations under visual control and (2) a delayed visual reaching task, in which GP and controls were asked to reach remembered target locations visually presented. We measured constant and variable distance and direction errors. In immediate reaching task, GP accurately reached target locations. In delayed reaching task, GP overshot remembered target locations, whereas the controls undershot them. Furthermore, variable errors were greater in GP than in the controls. In the experiment 3, GP and controls performed a delayed proprioceptive reaching task. Constant reaching errors did not differ between GP and the controls. However, variable direction errors were greater in GP than in the controls. We suggest that the occipitoparietal damage, and the relatively intact occipitotemporal region, produced in GP an attentional orienting bias toward the far/upper space (experiment 1). In turns, the attentional bias selectively shifted toward the far space remembered visual (experiment 2), but not proprioceptive (experiment 3), target locations. As a whole, these findings further support the hypothesis of an involvement of the occipitotemporal stream in delayed reaching. Furthermore, the observation that in both delayed reaching tasks the variable errors were greater in GP than in the controls suggested that in optic ataxia is present not only a visuo- but also a proprioceptivo-motor integration deficit

Motor activity plays a pivotal role in psychological, educational and social terms: sport practice induces harmonious physical development with common important benefits independently from sport type and each sport imposes rules respect that children learns to know and respect step by step improving the social skills and cognitive abilities. Sport has a very important role in the growth of children and adolescents. Sport and physical activity work as a moral laboratory to practice decisionmaking and problem-solving skills, as well as teamwork and cooperation. In many pathological conditions, the sport practice is strongly discouraged, as in epileptic patients for the negative consequences on their physical condition and psychic. In general, several studies reported that physical activity has positive influence on seizure frequency and severity. As a result, attitudes regarding sports and epilepsy have changed considerably in the last decades and presently, the risk of convulsive seizures during sports practice is minimal in case of well-managed epilepsy. Evaluating the control of convulsive disease is therefore a key point to allow sports in the children and adolescents
