    Effect of peri-implant mucosal thickness on esthetic outcomes and the efficacy of soft tissue augmentation procedures: Consensus report of group 2 of the SEPA/DGI/OF workshop

    OBJECTIVES: The aim of this study was to comprehensively assess the literature in terms of the effect of peri‐implant mucosal thickness on esthetic outcomes and the efficacy of soft tissue augmentation procedures to increase the mucosal thickness with autogenous grafts or soft tissue substitutes. MATERIAL AND METHODS: Two systematic reviews (SR) were performed prior to the consensus meeting to assess the following questions. Review 1, focused question: In systemically healthy patients with an implant‐supported fixed prosthesis, what is the influence of thin as compared to thick peri‐implant mucosa on esthetic outcomes? Review 2, focused question 1: In systemically healthy humans with at least one dental implant (immediate or staged implant), what is the efficacy of connective tissue graft (CTG), as compared to absence of a soft tissue grafting procedure, in terms of gain in peri‐implant soft tissue thickness (STT) reported by randomized controlled clinical trials (RCTs) or controlled clinical trials (CCTs)? Review 2, focused question 2: In systemically healthy humans with at least one dental implant (immediate or staged implant), what is the efficacy of CTG, as compared to soft tissue substitutes, in terms of gain in peri‐implant STT reported by RCTs or CCTs? The outcomes of the two SRs, the consensus statements, the clinical implications, and the research recommendations were discussed and subsequently approved at the consensus meeting during the group and plenary sessions. CONCLUSIONS: There was a tendency of superior esthetic outcomes in the presence of a thick mucosa. The connective tissue graft remains the standard of care in terms of increasing mucosa thickness

    Global guidance on environmental life cycle impact assessment indicators: impacts of climate change, fine particulate matter formation, water consumption and land use

    Purpose Guidance is needed on best-suited indicators to quantify and monitor the man-made impacts on human health, biodiversity and resources. Therefore, the UNEP-SETAC Life Cycle Initiative initiated a global consensus process to agree on an updated overall life cycle impact assessment (LCIA) framework and to recommend a non-comprehensive list of environmental indicators and LCIA characterization factors for (1) climate change, (2) fine particulate matter impacts on human health, (3) water consumption impacts (both scarcity and human health) and 4) land use impacts on biodiversity. Methods The consensus building process involved more than 100 world-leading scientists in task forces via multiple workshops. Results were consolidated during a 1-week Pellston Workshopℱ in January 2016 leading to the following recommendations. Results and discussion LCIA framework: The updated LCIA framework now distinguishes between intrinsic, instrumental and cultural values, with disability-adjusted life years (DALY) to characterize damages on human health and with measures of vulnerability included to assess biodiversity loss. Climate change impacts: Two complementary climate change impact categories are recommended: (a) The global warming potential 100 years (GWP 100) represents shorter term impacts associated with rate of change and adaptation capacity, and (b) the global temperature change potential 100 years (GTP 100) characterizes the century-scale long term impacts, both including climate-carbon cycle feedbacks for all climate forcers. Fine particulate matter (PM2.5) health impacts: Recommended characterization factors (CFs) for primary and secondary (interim) PM2.5 are established, distinguishing between indoor, urban and rural archetypes. Water consumption impacts: CFs are recommended, preferably on monthly and watershed levels, for two categories: (a) The water scarcity indicator “AWARE” characterizes the potential to deprive human and ecosystems users and quantifies the relative Available WAter REmaining per area once the demand of humans and aquatic ecosystems has been met, and (b) the impact of water consumption on human health assesses the DALYs from malnutrition caused by lack of water for irrigated food production. Land use impacts: CFs representing global potential species loss from land use are proposed as interim recommendation suitable to assess biodiversity loss due to land use and land use change in LCA hotspot analyses. Conclusions The recommended environmental indicators may be used to support the UN Sustainable Development Goals in order to quantify and monitor progress towards sustainable production and consumption. These indicators will be periodically updated, establishing a process for their stewardship

    Effects of Clinically Relevant MPL Mutations in the Transmembrane Domain Revealed at the Atomic Level through Computational Modeling

    BACKGROUND: Mutations in the thrombopoietin receptor (MPL) may activate relevant pathways and lead to chronic myeloproliferative neoplasms (MPNs). The mechanisms of MPL activation remain elusive because of a lack of experimental structures. Modern computational biology techniques were utilized to explore the mechanisms of MPL protein activation due to various mutations. RESULTS: Transmembrane (TM) domain predictions, homology modeling, ab initio protein structure prediction, and molecular dynamics (MD) simulations were used to build structural dynamic models of wild-type and four clinically observed mutants of MPL. The simulation results suggest that S505 and W515 are important in keeping the TM domain in its correct position within the membrane. Mutations at either of these two positions cause movement of the TM domain, altering the conformation of the nearby intracellular domain in unexpected ways, and may cause the unwanted constitutive activation of MPL's kinase partner, JAK2. CONCLUSIONS: Our findings represent the first full-scale molecular dynamics simulations of the wild-type and clinically observed mutants of the MPL protein, a critical element of the MPL-JAK2-STAT signaling pathway. In contrast to usual explanations for the activation mechanism that are based on the relative translational movement between rigid domains of MPL, our results suggest that mutations within the TM region could result in conformational changes including tilt and rotation (azimuthal) angles along the membrane axis. Such changes may significantly alter the conformation of the adjacent and intrinsically flexible intracellular domain. Hence, caution should be exercised when interpreting experimental evidence based on rigid models of cytokine receptors or similar systems

    Global guidance on environmental life cycle impact assessment indicators: Progress and case study

    International audiencePurpose: The life cycle impact assessment (LCIA) guidance flagship project of the United Nations Environment Programme (UNEP)/Society of Environmental Toxicology and Chemistry (SETAC) Life Cycle Initiative aims at providing global guidance and building scientific consensus on environmental LCIA indicators. This paper presents the progress made since 2013, preliminary results obtained for each impact category and the description of a rice life cycle assessment (LCA) case study designed to test and compare LCIA indicators. Methods: The effort has been focused in a first stage on impacts of global warming, fine particulate matter emissions, water use and land use, plus cross-cutting issues and LCA-based footprints. The paper reports the process and progress and specific results obtained in the different task forces (TFs). Additionally, a rice LCA case study common to all TF has been developed. Three distinctly different scenarios of producing and cooking rice have been defined and underlined with life cycle inventory data. These LCAs help testing impact category indicators which are being developed and/or selected in the harmonisation process. The rice LCA case study further helps to ensure the practicality of the finally recommended impact category indicators. Results and discussion: The global warming TF concludes that analysts should explore the sensitivity of LCA results to metrics other than GWP. The particulate matter TF attained initial guidance of how to include health effects from PM2.5 exposures consistently into LCIA. The biodiversity impacts of land use TF suggests to consider complementary metrics besides species richness for assessing biodiversity loss. The water use TF is evaluating two stress-based metrics, AWaRe and an alternative indicator by a stakeholder consultation. The cross-cutting issues TF agreed upon maintaining disability-adjusted life years (DALY) as endpoint unit for the safeguard subject 'human health'. The footprint TF defined main attributes that should characterise all footprint indicators. 'Rice cultivation' and 'cooking' stages of the rice LCA case study contribute most to the environmental impacts assessed. Conclusions: The results of the TF will be documented in white papers and some published in scientific journals. These white papers represent the input for the Pellston workshop', taking place in Valencia, Spain, from 24 to 29 January 2016, where best practice, harmonised LCIA indicators and an update on the general LCIA framework will be discussed and agreed on. With the diversity in results and the multi-tier supply chains, the rice LCA case study is well suited to test candidate recommended indicators and to ensure their applicability in common LCA case studies

    Domains belonging to the immunoglobulin-like fold are responsible for a wide variety of molecular recognition processes. Here we describe a new family of domains, the HYR family, which is predicted to belong to this fold, and which appears to be involved in cellular adhesion. HYR domains were identified in several eukaryotic proteins, often associated with Complement Control Protein (CCP) modules or arranged in multiple copies. Our analysis provides a sequence and structural basis for understanding the role of these domains in interaction mechanisms and leads to further characterization of heretofore undescribed repeated domains with similar folds found in several bacterial proteins involved in enzymatic activities (some chitinases) or in cell surface adhesion (streptococcal C-alpha antigen)

    International audiencev-mpl is a constitutively activated, truncated form of a cytokine receptor that has been transduced in a murine retrovirus, the myeloproliferative leukemia virus (MPLV). Expression of this oncogene results in the factor-independent proliferation of myeloid, erythroid, megakaryocytic, and mast precursor cells, which retain the ability to differentiate. However, no lymphoid disease was ever reported. To determine whether MPLV could infect and transform very early B cells and their precursors (BCPs), lymphoid long-term bone marrow cultures were infected with a helper-free MPLV. Within 3 wk after infection, highly proliferating BCPs could be isolated. These cells were able to clone spontaneously in semi-solid cultures, grown in the absence of feeder cell layer or exogenous growth factor and rapidly produced tumors after s.c. injection into synegic irradiated mice. In addition, MPLV transformation of pre-B cells led to the induction of an autocrine activity. Immunophenotypic and molecular analysis indicated that MPLV transformed early pro-B, pro-B, and pre-B cells, according to the expression of HSA, CD43, B220, Thy1, s-IgM and BP1 Ags, and to the rearrangements of Ig genes. Interestingly, MPLV-transformed BCPs expressed Mac1 Ag without acquiring further characteristics of macrophagic differentiation. Although the v-mpl cytoplasmic domain is devoid of tyrosine kinase consensus sequence, MPLV-transformed pre-B cells contained a major approximately 105-kDa tyrosine-phosphorylated protein that was not detected in uninfected cells or in cells transformed by the Abelson viral oncogene (v-abl). These results demonstrate that, like v-abl, the truncated cytokine receptor v-mpl is able to transform BCPs in vitro and suggest that the oncogenic transformation of BCPs by either v-mpl or v-abl use different pathways
