Background: Angiolipoma is a benign tumor composed of endothelial cells and mature adipocytes. Tumors reported in domestic species include two variants; infiltrative or non-infiltrative. Bitches and intact males seem predisposed. This mesenchyme tumor is commonly mistaken with lipoma due to its soft texture and subcutaneous site and often requires histopathology to confirm its diagnosis. Microscopic examination also enables the evaluation of surgical margins and rule out possible infiltrative sites. Complete surgical excision is usually curative. This study reports a case of non-infiltrating angiolipoma in a dog.Case: A 14-year-old mixed-breed dog was presented to a veterinary clinic in the city of Rio de Janeiro, RJ, Brazil. On palpation, a painless mass was noted, with high mobility and covered by intact hirsute skin in the right subcutaneous ventrolateral region. Computed tomography of the chest showed an expansive mass of uptake only from the edges of the soft tissues of the right subcutaneous ventrolateral region. The mass was homogeneous and well delimited, suggesting a neoplastic process. Subsequently, the mass was surgically removed, fixed in 10% buffered formalin, and sent for histopathological analysis. On macroscopic examination, the mass was well delimited, without skin coverage, and measured 2.3 × 1.9 × 0.6 cm. The consistency was smooth and unctuous in appearance with a compact cream-colored surface with blackish multifocal spots. Under microscopy, the histological sections showed neoplasm of mature adipocytes and of endothelial cells of blood vessels benign were filled with a marked amount of red blood cells. Multifocal fibrin thrombi and a mild inflammatory infiltrate composed of lymphocytes and rare mast cells were evident. There was no infiltration in the regional skeletal musculature. Thus, a diagnosis of non-infiltrative angiolipoma was established.Discussion: The diagnosis of non-infiltrating angiolipoma in this case was established through the results of histopathological examination. The occurrence of this neoplasm in dogs is uncommon, and the data reported in the veterinary medicine literature are scarce. However, in this study, it was found that the neoplasm on screening presented a behavior like that of lipomas, with noninvasive growth and the absence of local recurrence. The canine species does not commonly convey pain on palpation during a clinical examination, as observed in the present case. In humans, multiple angiolipoma nodules are common; this clinical presentation differs from that in animals, in which solitary nodules are generally observed. In dogs, as in the present case, they seem to have a predilection for the trunk. In animals, the pathogenesis of angiolipomas is not established, but in humans, it is based on theories that include the reaction to harmful stimuli and congenital malformation of adipose tissue. In humans, the presence of fibrin thrombi on the periphery of the region of cell proliferation are microscopic findings that can assist in the diagnosis of angiolipomas, an approach that was implemented in the present case. The occurrence of this neoplasm in dogs is uncommon, and the data reported in the veterinary medicine literature are scarce. The importance of an adequate description of angiolipomas is based on the need to provide information about its epidemiology, biological behavior, and prognosis

Scaffold repositioning of spiro-acridine derivatives as fungi chitinase inhibitor by target fishing and in vitro studies

Abstract The concept of “one target, one drug, one disease” is not always true, as compounds with previously described therapeutic applications can be useful to treat other maladies. For example, acridine derivatives have several potential therapeutic applications. In this way, identifying new potential targets for available drugs is crucial for the rational management of diseases. Computational methodologies are interesting tools in this field, as they use rational and direct methods. Thus, this study focused on identifying other rational targets for acridine derivatives by employing inverse virtual screening (IVS). This analysis revealed that chitinase enzymes can be potential targets for these compounds. Subsequently, we coupled molecular docking consensus analysis to screen the best chitinase inhibitor among acridine derivatives. We observed that 3 compounds displayed potential enhanced activity as fungal chitinase inhibitors, showing that compound 5 is the most active molecule, with an IC50 of 0.6 ng/µL. In addition, this compound demonstrated a good interaction with the active site of chitinases from Aspergillus fumigatus and Trichoderma harzianum. Additionally, molecular dynamics and free energy demonstrated complex stability for compound 5. Therefore, this study recommends IVS as a powerful tool for drug development. The potential applications are highlighted as this is the first report of spiro-acridine derivatives acting as chitinase inhibitors that can be potentially used as antifungal and antibacterial candidates

It is a descriptive study, with a qualitative approach, based on theoretical-methodological categories of the popular education and the symbolic market communication model. The data was collected in four teams of the Family Health Program. The results show the communication maps elaborated with the HCA which identifies the group of technical support, the technician team, the HCA themselves and the old inhabitants of the community as their main interlocutors. The communication with the other professionals of the team is acknowledged in a predominantly verticalized way with authoritarianism traces. The results highlight the importance of extending the dialogue and negotiation in the established communication within the team and with the community

The stepwise selection for ketoconazole resistance induces upregulation of C14-demethylase (CYP51) in Leishmania amazonensis

    No full text
Ketoconazole is a clinically safe antifungal agent that also inhibits the growth of Leishmania spp. A study was undertaken to determine whether Leishmania parasites are prone to becoming resistant to ketoconazole by upregulating C14-demethylase after stepwise pharmacological pressure. Leishmania amazonensis promastigotes [inhibitory concentration (IC)50 = 2 µM] were subjected to stepwise selection with ketoconazole and two resistant lines were obtained, La8 (IC50 = 8 µM) and La10 (IC50 = 10 µM). As a result, we found that the resistance level was directly proportional to the C14-demethylase mRNA expression level; we also observed that expression levels were six and 12 times higher in La8 and La10, respectively. This is the first demonstration that L. amazonensis can up-regulate C14-demethylase in response to drug pressure and this report contributes to the understanding of the mechanisms of parasite resistance
