    English abstract: Employment and crime are commonly assumed to be negatively correlated. Those employed are less likely to commit crimes, and conversely, those who have a criminal record are less likely to become employed. Criminological research has provided strong empirical and theoretical support for the link between employment and crime, but also suggests that a complex set of mechanisms may be at play. Additionally, studies show that employment can also increase the risk of criminal behaviour. In the introduction of this special issue, three causal relationships in the work-crime nexus will be discussed: employment causing crime, employment preventing crime, and crime blocking future employment. Dutch abstract: Woning, werk en wijf. In het reclasseringsjargon zijn dit al heel lang de drie voorwaarden voor succesvolle resocialisatie van delinquenten na detentie en voor preventie van recidive. Vooral werk wordt vaak gezien als een mogelijke oplossing voor problemen met delinquentie en criminaliteit. Het maatschappelijke belang en de – vaak ook complexe – rol van werk in de theorievorming hebben ertoe geleid dat het onderwerp ‘criminaliteit en werk’ in toenemende mate kan rekenen op de belangstelling van criminologen. In criminologisch onderzoek wordt de negatieve relatie tussen werk en criminaliteit zowel theoretisch als empirisch ondersteund (Fagan & Freeman, 1999; Staff e.a., 2010). Door het gebruik van sterke methodologische onderzoeksdesigns is de wetenschappelijke basis van deze algemene vooronderstelling sterker geworden. Door de invloed van het levensloopperspectief is binnen de criminologie sprake van een zekere revival van thema’s die enkele decennia terug centraal stonden in de criminologie, bijvoorbeeld in de Utrechtse school, die in werk vooral een kans op resocialisatie zag, en in de kritische criminologie, die vooral wees op het negatieve stigma van een strafblad voor het vinden van werk

    PROTEOFORMER 2.0 : further developments in the ribosome profiling-assisted proteogenomic hunt for new proteoforms

    PROTEOFORMER is a pipeline that enables the automated processing of data derived from ribosome profiling (RIBO-seq, i. e. the sequencing of ribosome-protected mRNA fragments). As such, genome-wide ribosome occupancies lead to the delineation of data-specific translation product candidates and these can improve the mass spectrometry-based identification. Since its first publication, different upgrades, new features and extensions have been added to the PROTEOFORMER pipeline. Some of the most important upgrades include P-site offset calculation during mapping, comprehensive data preexploration, the introduction of two alternative proteoform calling strategies and extended pipeline output features. These novelties are illustrated by analyzing ribosome profiling data of human HCT116 and Jurkat data. The different proteoform calling strategies are used alongside one another and in the end combined together with reference sequences from UniProt. Matching mass spectrometry data are searched against this extended search space with MaxQuant. Overall, besides annotated proteoforms, this pipeline leads to the identification and validation of different categories of new proteoforms, including translation products of up-and downstream open reading frames, 5 and 3 extended and truncated proteoforms, single amino acid variants, splice variants and translation products of so-called noncoding regions. Further, proof-of-concept is reported for the improvement of spectrum matching by including Prosit, a deep neural network strategy that adds extra fragmentation spectrum intensity features to the analysis. In the light of ribosome profiling-driven proteogenomics, it is shown that this allows validating the spectrum matches of newly identified proteoforms with elevated stringency. These updates and novel conclusions provide new insights and lessons for the ribosome profiling-based proteogenomic research field.     Scattered tree death contributes to substantial forest loss in California

    In recent years, large-scale tree mortality events linked to global change have occurred around the world. Current forest monitoring methods are crucial for identifying mortality hotspots, but systematic assessments of isolated or scattered dead trees over large areas are needed to reduce uncertainty on the actual extent of tree mortality. Here, we mapped individual dead trees in California using sub-meter resolution aerial photographs from 2020 and deep learning-based dead tree detection. We identified 91.4 million dead trees over 27.8 million hectares of vegetated areas (16.7-24.7% underestimation bias when compared to field data). Among these, a total of 19.5 million dead trees appeared isolated, and 60% of all dead trees occurred in small groups ( ≤ 3 dead trees within a 30 × 30 m grid), which is largely undetected by other state-level monitoring methods. The widespread mortality of individual trees impacts the carbon budget and sequestration capacity of California forests and can be considered a threat to forest health and a fuel source for future wildfires

    Bioleaching of metals from secondary materials using glycolipid biosurfactants

    With the global demand for economically important metals increasing, compounded by the depletion of readily accessible ores, secondary resources and low-grade ores are being targeted to meet growing demands. Novel technologies developed within biobased industries, such as microbial biosurfactants, could be implemented to improve the sustainability of traditional hydrometallurgy techniques. This study investigates newly developed microbial biosurfactants (acidic- and bolaform glycolipids) for the leaching of metals (particularly Cu and Zn) from a suite of mine tailings, metallurgical sludges and automotive shredder residues. Generally, acidic sophorolipids were the most performant, and optimal Cu leaching was observed from a fayalite slag (27%) and a copper sulfide mine tailing (53%). Further investigation of the leached fayalite material showed that leaching was occurring from small metallic Cu droplets in this material via a corrosion-based mechanism, and/or from Cu-Pb sulfides, selective against dominant Fe-silicate matrices. This study highlights that acidic sophorolipid microbial biosurfactants have the potential to leach Cu and Zn from low-grade secondary materials. It also provides important fundamental insights into biosurfactant-metal and mineral interactions that are currently unexplored. Together, the convergence of leaching and mining industries with bio-industries can improve material recovery and will positively impact the bio- and circular economies and the environment.The authors thank Bio Base Europe Pilot plant for supplying the biosurfactants that enabled the execution of the leaching experiments.     Using terrestrial laser scanning to constrain forest ecosystem structure and functions in the Ecosystem Demography model (ED2.2)

    Terrestrial biosphere models (TBMs) are invaluable tools for studying plant-atmosphere interactions at multiple spatial and temporal scales, as well as how global change impacts ecosystems. Yet, TBM projections suffer from large uncertainties that limit their usefulness. Forest structure drives a significant part of TBM uncertainty as it regulates key processes such as the transfer of carbon, energy, and water between the land and the atmosphere, but it remains challenging to observe and reliably represent. The poor representation of forest structure in TBMs might actually result in simulations that reproduce observed land fluxes but fail to capture carbon pools, forest composition, and demography. Recent advances in terrestrial laser scanning (TLS) offer new opportunities to capture the three-dimensional structure of the ecosystem and to transfer this information to TBMs in order to increase their accuracy. In this study, we quantified the impacts of prescribing initial conditions (tree size distribution), constraining key model parameters with observations, as well as imposing structural observations of individual trees (namely tree height, leaf area, woody biomass, and crown area) derived from TLS on the state-of-the-art Ecosystem Demography model (ED2.2) of a temperate forest site (Wytham Woods, UK). We assessed the relative contributions of initial conditions, model structure, and parameters to the overall output uncertainty by running ensemble simulations with multiple model configurations. We show that forest demography and ecosystem functions as modelled by ED2.2 are sensitive to the imposed initial state, the model parameters, and the choice of key model processes. In particular, we show that: Parameter uncertainty drove the overall model uncertainty, with a mean contribution of 63 % to the overall variance of simulated gross primary production. Model uncertainty in the gross primary production was reduced fourfold when both TLS and trait data were integrated into the model configuration. Land fluxes and ecosystem composition could be simultaneously and accurately simulated with physically realistic parameters when appropriate constraints were applied to critical parameters and processes. We conclude that integrating TLS data can inform TBMs of the most adequate model structure, constrain critical parameters, and prescribe representative initial conditions. Our study also confirms the need for simultaneous observations of plant traits, structure, and state variables if we seek to improve the robustness of TBMs and reduce their overall uncertainties.Peer reviewe

    An adjudication algorithm for respiratory-related hospitalisation in idiopathic pulmonary fibrosis

    Background:There is no standard definition of respiratory-related hospitalisation, a common end-point in idiopathic pulmonary fibrosis (IPF) clinical trials. As diverse aetiologies and complicating comorbidities can present similarly, external adjudication is sometimes employed to achieve standardisation of these events. Methods:An algorithm for respiratory-related hospitalisation was developed through a literature review of IPF clinical trials with respiratory-related hospitalisation as an end-point. Experts reviewed the algorithm until a consensus was reached. The algorithm was validated using data from the phase 3 ISABELA trials (clinicaltrials.gov identifiers NCT03711162 and NCT03733444), by assessing concordance between nonadjudicated, investigator-defined, respiratory-related hospitalisations and those defined by the adjudication committee using the algorithm. Results:The algorithm classifies respiratory-related hospitalisation according to cause: extraparenchymal (worsening respiratory symptoms due to left heart failure, volume overload, pulmonary embolism, pneumothorax or trauma); other (respiratory tract infection, right heart failure or exacerbation of COPD); “definite” acute exacerbation of IPF (AEIPF) (worsening respiratory symptoms within 1 month, with radiological or histological evidence of diffuse alveolar damage); or “suspected” AEIPF (as for “definite” AEIPF, but with no radiological or histological evidence of diffuse alveolar damage). Exacerbations (“definite” or “suspected”) with identified triggers (infective, post-procedural or traumatic, drug toxicity-or aspiration-related) are classed as “known AEIPF”; “idiopathic AEIPF” refers to exacerbations with no identified trigger. In the ISABELA programme, there was 94% concordance between investigator-and adjudication committee-determined causes of respiratory-related hospitalisation. Conclusion:The algorithm could help to ensure consistency in the reporting of respiratory-related hospitalisation in IPF trials, optimising its utility as an end-point.</p

    Disturbance of Glucose Homeostasis After Pediatric Cardiac Surgery

    This study aimed to evaluate the time course of perioperative blood glucose levels of children undergoing cardiac surgery for congenital heart disease in relation to endogenous stress hormones, inflammatory mediators, and exogenous factors such as caloric intake and glucocorticoid use. The study prospectively included 49 children undergoing cardiac surgery. Blood glucose levels, hormonal alterations, and inflammatory responses were investigated before and at the end of surgery, then 12 and 24 h afterward. In general, blood glucose levels were highest at the end of surgery. Hyperglycemia, defined as a glucose level higher than 8.3 mmol/l (>150 mg/dl) was present in 52% of the children at the end of surgery. Spontaneous normalization of blood glucose occurred in 94% of the children within 24 h. During surgery, glucocorticoids were administered to 65% of the children, and this was the main factor associated with hyperglycemia at the end of surgery (determined by univariate analysis of variance). Hyperglycemia disappeared spontaneously without insulin therapy after 12–24 h for the majority of the children. Postoperative morbidity was low in the study group, so the presumed positive effects of glucocorticoids seemed to outweigh the adverse effects of iatrogenic hyperglycemia
