    The Arabidopsis AtRaptor genes are essential for post-embryonic plant growth

    BACKGROUND: Flowering plant development is wholly reliant on growth from meristems, which contain totipotent cells that give rise to all post-embryonic organs in the plant. Plants are uniquely able to alter their development throughout their lifespan through the generation of new organs in response to external signals. To identify genes that regulate meristem-based growth, we considered homologues of Raptor proteins, which regulate cell growth in response to nutrients in yeast and metazoans as part of a signaling complex with the target of rapamycin (TOR) kinase. RESULTS: We identified AtRaptor1A and AtRaptor1B, two loci predicted to encode Raptor proteins in Arabidopsis. Disruption of AtRaptor1B yields plants with a wide range of developmental defects: roots are thick and grow slowly, leaf initiation and bolting are delayed and the shoot inflorescence shows reduced apical dominance. AtRaptor1A AtRaptor1B double mutants show normal embryonic development but are unable to maintain post-embryonic meristem-driven growth. AtRaptor transcripts accumulate in dividing and expanding cells and tissues. CONCLUSION: The data implicate the TOR signaling pathway, a major regulator of cell growth in yeast and metazoans, in the maintenance of growth from the shoot apical meristem in plants. These results provide insights into the ways in which TOR/Raptor signaling has been adapted to regulate plant growth and development, and indicate that in plants, as in other eukaryotes, there is some Raptor-independent TOR activity

    Sound studies and Deaf studies may seem at first impression to operate in worlds apart. We argue in this article, however, that similar renderings of hearing, deafness, and seeing as ideal types - and as often essentialized sensory modes - make it possible to read differences between Sound studies and Deaf studies as sites of possible articulation. We direct attention to four zones of productive overlap, attending to how sound is inferred in deaf and Deaf practice, how reimagining sound in the register of low-frequency vibration can upend deafhearing dichotomies, how “deaf futurists“ champion cyborg sound, and how signing and other non-spoken communicative practices might undo phonocentric models of speech. The definitive version was published in Deep Sea Research Part II: Topical Studies in Oceanography 55 (2008): 485-499, doi:10.1016/j.dsr2.2007.11.006.A key hypothesis guiding the U.S. Southern Ocean Global Ocean Ecosystems Dynamics (U.S. SO GLOBEC) program is that deep across-shelf troughs facilitate the transport of warm and nutrient-rich waters onto the continental shelf of the Western Antarctic Peninsula, resulting in enhanced winter production and prey availability to top predators. We tested aspects of this hypothesis during austral winter by assessing the distribution of the resident pack-ice top predators in relation to these deep across-shelf troughs and by investigating associations between top predators and their prey. Surveys were conducted July-August 2001 and August-September 2002 in Marguerite Bay, Antarctica, with a focus on the main across-shelf trough in the bay, Marguerite Trough. The common pack-ice seabird species were snow petrel (Pagodroma nivea, 1.2 individuals km-2), Antarctic petrel (Thalassoica antarctica, 0.3 individuals km-2), and Adélie penguin (Pygoscelis adeliae, 0.5 individuals km-2). The most common pack-ice pinniped was crabeater seal (Lobodon carcinophagus). During both winters, snow and Antarctic petrels were associated with low sea ice concentrations independent of Marguerite Trough, while Adélie penguins occurred in association with this trough. Krill concentrations, both shallow and deep, were also associated with Adélie penguin and snow petrel distributions. During both winters, crabeater seal occurrence was associated with deep krill concentrations and with regions of lower chlorophyll concentration. The area of lower chlorophyll concentrations occurred in an area with complex bathymetry close to land and heavy ice concentrations. Complex or unusual bathymetry via its influence on physical and biological processes appears to be one of the keys to understanding how top predators survive during the winter in this Antarctic region.This material is based upon work supported by the National Science Foundation under Grants No. OPP-9910096 (to C.     Effects of Aversive Stimuli on Prospective Memory. An Event-Related fMRI Study

    Prospective memory (PM) describes the ability to execute a previously planned action at the appropriate point in time. Although behavioral studies clearly showed that prospective memory performance is affected by the emotional significance attributed to the intended action, no study so far investigated the brain mechanisms subserving the modulatory effect of emotional salience on PM performance. The general aim of the present study was to explore brain regions involved in prospective memory processes when PM cues are associated with emotional stimuli. In particular, based on the hypothesised critical role of the prefrontal cortex in prospective memory in the presence of emotionally salient stimuli, we expected a stronger involvement of aPFC when the retrieval and execution of the intended action is cued by an aversive stimulus. To this aim BOLD responses of PM trials cued by aversive facial expressions were compared to PM trials cued by neutral facial expressions. Whole brain analysis showed that PM task cued by aversive stimuli is differentially associated with activity in the right lateral prefrontal area (BA 10) and in the left caudate nucleus. Moreover a temporal shift between the response of the caudate nucleus that preceded that of aPFC was observed. These findings suggest that the caudate nucleus might provide an early analysis of the affective properties of the stimuli, whereas the anterior lateral prefrontal cortex (BA10) would be involved in a slower and more deliberative analysis to guide goal-directed behaviour

    AI is a viable alternative to high throughput screening: a 318-target study

    : High throughput screening (HTS) is routinely used to identify bioactive small molecules. This requires physical compounds, which limits coverage of accessible chemical space. Computational approaches combined with vast on-demand chemical libraries can access far greater chemical space, provided that the predictive accuracy is sufficient to identify useful molecules. Through the largest and most diverse virtual HTS campaign reported to date, comprising 318 individual projects, we demonstrate that our AtomNet® convolutional neural network successfully finds novel hits across every major therapeutic area and protein class. We address historical limitations of computational screening by demonstrating success for target proteins without known binders, high-quality X-ray crystal structures, or manual cherry-picking of compounds. We show that the molecules selected by the AtomNet® model are novel drug-like scaffolds rather than minor modifications to known bioactive compounds. Our empirical results suggest that computational methods can substantially replace HTS as the first step of small-molecule drug discovery
