Tumor infiltration by chemokine receptor 7 (CCR7)+ T-lymphocytes is a favorable prognostic factor in metastatic colorectal cancer
The immune interactions occurring within the tumor microenvironment have a critical role in determining the outcome of colorectal cancer patients. We carried-out an immunohistochemical analysis of tumor infiltrating T-lymphocytes expressing chemokine receptor 7 (CCR7) in a series of colorectal cancer patients enrolled in a prospective clinical trial. We demonstrated that a high tumor infiltration score of this lymphocyte subset is predictive of longer progression free survival and overall survival. © 2012 Landes Bioscience
Observing the dynamics of super-massive black hole binaries with Pulsar Timing Arrays
Pulsar Timing Arrays are a prime tool to study unexplored astrophysical
regimes with gravitational waves. Here we show that the detection of
gravitational radiation from individually resolvable super-massive black hole
binary systems can yield direct information about the masses and spins of the
black holes, provided that the gravitational-wave induced timing fluctuations
both at the pulsar and at the Earth are detected. This in turn provides a map
of the non-linear dynamics of the gravitational field and a new avenue to
tackle open problems in astrophysics connected to the formation and evolution
of super-massive black holes. We discuss the potential, the challenges and the

Angular Resolution of the LISA Gravitational Wave Detector
We calculate the angular resolution of the planned LISA detector, a
space-based laser interferometer for measuring low-frequency gravitational
waves from galactic and extragalactic sources. LISA is not a pointed
instrument; it is an all-sky monitor with a quadrupolar beam pattern. LISA will
measure simultaneously both polarization components of incoming gravitational
waves, so the data will consist of two time series. All physical properties of
the source, including its position, must be extracted from these time series.
LISA's angular resolution is therefore not a fixed quantity, but rather depends
on the type of signal and on how much other information must be extracted.
Information about the source position will be encoded in the measured signal in
three ways: 1) through the relative amplitudes and phases of the two
polarization components, 2) through the periodic Doppler shift imposed on the
signal by the detector's motion around the Sun, and 3) through the further
modulation of the signal caused by the detector's time-varying orientation. We
derive the basic formulae required to calculate the LISA's angular resolution
for a given source. We then evaluate for
two sources of particular interest: monchromatic sources and mergers of
supermassive black holes. For these two types of sources, we calculate (in the
high signal-to-noise approximation) the full variance-covariance matrix, which
gives the accuracy to which all source parameters can be measured. Since our
results on LISA's angular resolution depend mainly on gross features of the
detector geometry, orbit, and noise curve, we expect these results to be fairly
insensitive to modest changes in detector design that may occur between now and
launch. We also expect that our calculations could be easily modified to apply

Metabolic and hormonal adaptation in Bubalus bubalis around calving and early lactation
Pregnancy and lactation are physiological periods that result in an increased metabolic demand that, if not satisfied, could provoke a threat to homeostasis. In this study changes in the values of Non-Esterified Fatty Acids (NEFA) and β-hydroxybutyrate (BHB), glucose, insulin, thyroid hormones, milk composition and yield were investigated in buffaloes during the late pregnancy and early lactation. From a total of 50 buffaloes, blood samples were collected -7±5 days before expected calving; +7±5; +30±5 and +50±5 days post-partum; milk samples were collected at the same post-partum time points. On serum samples, the values of Non-Esterified Fatty Acids (NEFA), β-hydroxybutyrate (BHB), glucose, insulin, Triiodothyronine (T3), Thyroxine (T4) and Thyroid-stimulating hormone (TSH) were evaluated. On milk, fat %, protein %, lactose %, somatic cells score (SCS), milk yield and daily milk production (DMP) were assessed. Peripartum period significantly influenced all studied parameters (P<0.05). Milk constituents and productivity statistically changed throughout monitoring period (P<0.005). Milk yield resulted positively correlated with insulin and TSH values, negatively correlated with NEFA and BHB. Insulin was negatively correlated with lactose % and positively correlated with SCS. The obtained results showed that the peripartum period and lactation are accompanied by marked changes in some biochemical variables and in the thyroid hormones values in Italian Mediterranean Buffaloes. Moreover, the relationship found between TSH values and milk yield seems to suggest a possible role of thyroid gland on the maintenance of lactogenesis. This study underlines the importance of monitoring the hormonal status of buffalo during the transition period in order to understand when adjustments of regulatory mechanisms break through physiological limits predisposing the buffalo to metabolic problems
Relative Sea-Level Rise Projections and Flooding Scenarios for 2150 CE for the Island of Ustica (Southern Tyrrhenian Sea, Italy)
The island of Ustica (Italy) is constantly exposed to the effects of sea-level rise, which is threatening its coastal zone. With the aim of assessing the sea levels that are anticipated by 2150 CE under the climatic projections shown in the AR6 report from the IPCC, a detailed evaluation of potential coastal flooding under different climatic scenarios and the ongoing land subsidence has been carried out for three coastal zones. Scenarios are based on the determination of the current coastline position, a high-resolution digital terrain and marine model, and the SSP1-2.6, SSP3-7.0, and SSP5-8.5 climatic projections. Relative sea-level rise projections allowed the mapping of the potential inundated surfaces for 2030, 2050, 2100, and 2150. The results show rising sea levels for 2150, ranging from a minimum of 66 ± 40 cm (IPCC AR6 SSP2.6 scenario) to a maximum of 128 ± 52 cm (IPCC AR6 SSP8.5 scenario). In such conditions, considering the SSP8.5 scenario during storm surges with return times (RTs) of 1 and 100 years, the expected maximum wave run-up along the island may vary from 3 m (RT = 1) to 14 m (RT = 100), according to the coastal morphology. Our results show that adaptation and mitigation actions are required to protect the touristic and harbor installations of the island
High Resolution Observations using Adaptive Optics: Achievements and Future Needs
Over the last few years, several interesting observations were obtained with
the help of solar Adaptive Optics (AO). In this paper, few observations made
using the solar AO are enlightened and briefly discussed. A list of
disadvantages with the current AO system are presented. With telescopes larger
than 1.5m are expected during the next decade, there is a need to develop the
existing AO technologies for large aperture telescopes. Some aspects of this
development are highlighted. Finally, the recent AO developments in India are
also presented
Global entrainment of transcriptional systems to periodic inputs
This paper addresses the problem of giving conditions for transcriptional
systems to be globally entrained to external periodic inputs. By using
contraction theory, a powerful tool from dynamical systems theory, it is shown
that certain systems driven by external periodic signals have the property that
all solutions converge to a fixed limit cycle. General results are proved, and
the properties are verified in the specific case of some models of
transcriptional systems. The basic mathematical results needed from contraction
theory are proved in the paper, making it self-contained
Extracting galactic binary signals from the first round of Mock LISA Data Challenges
We report on the performance of an end-to-end Bayesian analysis pipeline for
detecting and characterizing galactic binary signals in simulated LISA data.
Our principal analysis tool is the Blocked-Annealed Metropolis Hasting (BAM)
algorithm, which has been optimized to search for tens of thousands of
overlapping signals across the LISA band. The BAM algorithm employs Bayesian
model selection to determine the number of resolvable sources, and provides
posterior distribution functions for all the model parameters. The BAM
algorithm performed almost flawlessly on all the Round 1 Mock LISA Data
Challenge data sets, including those with many highly overlapping sources. The
only misses were later traced to a coding error that affected high frequency
sources. In addition to the BAM algorithm we also successfully tested a Genetic
Algorithm (GA), but only on data sets with isolated signals as the GA has yet

Contribution of cysteine residues in the extracellular domain of the F protein of human respiratory syncytial virus to its function
The mature F protein of all known isolates of human respiratory syncytial virus (HRSV) contains fifteen absolutely conserved cysteine (C) residues that are highly conserved among the F proteins of other pneumoviruses as well as the paramyxoviruses. To explore the contribution of the cysteines in the extracellular domain to the fusion activity of HRSV F protein, each cysteine was changed to serine. Mutation of cysteines 37, 313, 322, 333, 343, 358, 367, 393, 416, and 439 abolished or greatly reduced cell surface expression suggesting these residues are critical for proper protein folding and transport to the cell surface. As expected, the fusion activity of these mutations was greatly reduced or abolished. Mutation of cysteine residues 212, 382, and 422 had little to no effect upon cell surface expression or fusion activity at 32°C, 37°C, or 39.5°C. Mutation of C37 and C69 in the F2 subunit either abolished or reduced cell surface expression by 75% respectively. None of the mutations displayed a temperature sensitive phenotype
Progress on stochastic background search codes for LIGO
One of the types of signals for which the LIGO interferometric gravitational
wave detectors will search is a stochastic background of gravitational
radiation. We review the technique of searching for a background using the
optimally-filtered cross-correlation statistic, and describe the state of plans
to perform such cross-correlations between the two LIGO interferometers as well
as between LIGO and other gravitational-wave detectors, in particular the
