    Dose-dependent RNAi-mediated geminivirus resistance in the tropical root crop cassava

    Cassava mosaic disease is a major constraint for cassava production in Africa, resulting in significant economic losses. We have engineered transgenic cassava with resistance to African cassava mosaic virus (ACMV), by expressing ACMV AC1-homologous hairpin double-strand RNAs. Transgenic cassava lines with high levels of AC1-homologous small RNAs have ACMV immunity with increasing viral load and different inoculation methods. We report a correlation between the expression of the AC1-homologous small RNAs and the ACMV resistance of the transgenic cassava lines. Characterization of the small RNAs revealed that only some of the hairpin-derived small RNAs fall into currently known small interfering RNA classes in plants.     Cassava: constraints to production and the transfer of biotechnology to African laboratories

    Knowledge and technology transfer to African institutes is an important objective to help achieve the United Nations Millennium Development Goals. Plant biotechnology in particular enables innovative advances in agriculture and industry, offering new prospects to promote the integration and dissemination of improved crops and their derivatives from developing countries into local markets and the global economy. There is also the need to broaden our knowledge and understanding of cassava as a staple food crop. Cassava (Manihot esculenta Crantz) is a vital source of calories for approximately 500 million people living in developing countries. Unfortunately, it is subject to numerous biotic and abiotic stresses that impact on production, consumption, marketability and also local and country economics. To date, improvements to cassava have been led via conventional plant breeding programmes, but with advances in molecular-assisted breeding and plant biotechnology new tools are being developed to hasten the generation of improved farmer-preferred cultivars. In this review, we report on the current constraints to cassava production and knowledge acquisition in Africa, including a case study discussing the opportunities and challenges of a technology transfer programme established between the Mikocheni Agricultural Research Institute in Tanzania and Europe-based researchers. The establishment of cassava biotechnology platform(s) should promote research capabilities in African institutions and allow scientists autonomy to adapt cassava to suit local agro-ecosystems, ultimately serving to develop a sustainable biotechnology infrastructure in African countrie

    Transgenic cassava resistance to African cassava mosaic virus is enhanced by viral DNA-A bidirectional promoter-derived siRNAs

    Expression of double-stranded RNA (dsRNA) homologous to virus sequences can effectively interfere with RNA virus infection in plant cells by triggering RNA silencing. Here we applied this approach against a DNA virus, African cassava mosaic virus (ACMV), in its natural host cassava. Transgenic cassava plants were developed to express small interfering RNAs (siRNA) from a CaMV 35S promoter-controlled, intron-containing dsRNA cognate to the common region-containing bidirectional promoter of ACMV DNA-A. In two of three independent transgenic lines, accelerated plant recovery from ACMV-NOg infection was observed, which correlates with the presence of transgene-derived siRNAs 21-24nt in length. Overall, cassava mosaic disease symptoms were dramatically attenuated in these two lines and less viral DNA accumulation was detected in their leaves than in those of wild-type plants. In a transient replication assay using leaf disks from the two transgenic lines, strongly reduced accumulation of viral single-stranded DNA was observed.     Characterization of brown streak virus-resistant cassava

    Cassava brown streak disease (CBSD) has become a major constraint to cassava production in East and Central Africa. The identification of new sources of CBSD resistance is essential to deploy CBSD mitigation strategies as the disease is progressing westwards to new geographical areas. A stringent infection method based on top cleft grafting combined with precise virus titer quantitation was utilized to screen fourteen cassava cultivars and elite breeding lines. When inoculated with mixed infections of Cassava brown streak virus (CBSV) and Ugandan cassava brown streak virus (UCBSV), the scions of elite breeding lines KBH 2006/18 and KBH 2006/26 remained symptom-free during a 16-week period of virus graft inoculation, while susceptible varieties displayed typical CBSD infection symptoms at 4 weeks after grafting. The identified CBSD resistance was stable under the co-inoculation of CBSV, UCBSV with cassava geminiviruses (CGMs). Double grafting experiments revealed that transmission of CBSV and UCBSV to CBSD susceptible top scions was delayed when using intermediate scions of elite breeding lines KBH 2006/18 and KBH 2006/26. Nonetheless, comparison of virus systemic movement using scions from KBH2006/18 and a transgenic CBSD resistant 60444 line (60444-Hp9 line) showed that both CBSV and UCBSV move at undetectable levels through the stems. Further, protoplast-based assays of virus titers over time showed that the replication of CBSVs is inhibited in the resistant line KBH2006/18, suggesting that the identified CBSD resistance is at least partially based on inhibition of virus replication.     Assessment of pre- and post-harvest anti-sprouting treatments to replace CIPC for potato storage

    To avoid losses from sprouting during potato storage, the anti-sprouting agent chlorpropham [CIPC] has been widely used over the past few decades. However, the European Union recently decided not to authorize the renewal of CIPC, prompting the value chain to find alternative treatments. We assessed for three years the potential of pre- and post-harvest anti-sprouting treatments to replace CIPC using four potato-processing varieties. Pre-harvest application of maleic hydrazide [MH] and post-harvest applications of 3-decen-2-one, 1,4-dimethylnapthalene [1,4-DMN] and CIPC were performed following supplier’s recommendations. In addition, we evaluated the potential of 3-decen-2-one and 1,4-DMN to prolong the efficacy of pre-harvest MH treatment anti-sprouting activity during storage. All molecules significantly reduced sprouting after seven months of storage compared with the untreated control group. MH, 3-decen-2-one, 1,4-DMN and CIPC displayed respectively 86.9 %; 77.9 %, 73.6 % and 99.8 % of efficacy to control sprout weight and 79.4 %; 73.4 %, 68.4 % and 96.9 % of efficacy to control sprout length. Our results suggest that using 3-decen-2-one and 1,4-DMN in combination with MH do not bring additional benefit to control sprouting. Because differences in dormancies could be observed between varieties, we also showed that the efficacy of post-harvest treatments is genotype-dependent, while MH pre-harvest treatment is effective equally for all varieties. Applications of CIPC and MH led to detectable residues in tubers, while no residue of 1,4-DMN has been detected in tubers treated with this molecule (< LOQ). We concluded that treatments with MH, 1,4-DMN and 3-decen-2-one are valuable alternatives to CIPC to control sprouting of processing potatoes

    Identification of potato varieties suitable for cold storage and reconditioning: A safer alternative to anti-sprouting chemicals for potato sprouting control.

    peer reviewedLow temperature storage as an alternative to anti-sprouting chemicals in potato storage may induce reducing sugars (RS) accumulation (i.e. glucose and fructose) in potato tubers. This phenomenon is called "cold induced sweetening" (CIS) and occurs in certain varieties. CIS leads to a decrease in the organoleptic qualities and darkening of processed potato and the accumulation of toxic molecules such as acrylamide. To identify potato varieties suitable for storage at low temperatures, we screened six commercial processing varieties: Lady Claire (LC), Verdi, Kiebitz (KB), Pirol, Agria and Markies for their CIS characteristics and sprout-forming potential after storage at 4 °C and 8 °C. Our findings reveal that 4 °C storage allows for efficient sprout reduction in all six tested varieties for up to 4.5 months of storage. Three CIS-resistant varieties, namely Verdi, Lady Claire and Kiebitz, were identified as able to be stored for up to four months at 4 °C with limited increase in glucose content. Conversely, Pirol, Agria and Markies showed an increase in glucose content with a decrease in storage temperature and can be considered as CIS-susceptible varieties. After processing into crisps, the CIS-susceptible varieties displayed poor crisp color quality (brown to black color crisps) after storage for two months at 4 °C compared to the storage at 8 °C, whereas the CIS-resistant varieties had good crisp color quality (pale yellow color crisps) after storage at both 4 and 8 °C. Interestingly, the trends of total RS and/or glucose content in the CIS-resistant and in the CIS-susceptible varieties were correlated with the trends in Vacuolar Invertase (VInv) gene expression for most varieties, as well as with the trends in acrylamide content after processing. In addition, reconditioning of Markies variety after storage at 4 °C by gradually increasing the temperature to 15 °C resulted in a significant decrease of VInv transcript levels (reduction of 80 %), acrylamide content (reduction of 75 %) and glucose content when compared to a storage at 4 °C without reconditioning. Those results demonstrate that the reconditioning technique is a key factor for a sustainable potato storage and for improving the quality of processed potatoes

    Towards green chemicals and edible coatings from barks and peels with near critical extraction of suberin

    peer reviewedDue to their complex structures, industrial suberoligneous by-products such as barks and peels remain poorly exploited. In this study, we demonstrated the efficient extraction of suberin from barks (11.3%) and peels (20.85%) via supercritical and near-supercritical transesterification without the need for catalysts. When applied in non-isocyanate polyurethane (NIPU) foams, suberinic extracts allowed the increase of the biomass content by up to 20% and facilitated the foaming process by improving pre-curing viscosity. When sprayed on fruits, the suberinic extracts effectively reduced the water loss rate by 25%, extending the shelf-life of the produce. A life cycle analysis for the latter application confirmed the competitive potential of our process against plastic sealing. This work uncovers the potential of suberoligneous biomass (SBM) to provide sustainable solutions to emerging societal challenges

    Potato sprouting during storage occurs after a break in dormancy, leading to a decrease in quality and consequently economic losses. We used 3379 records from multi-year and multi-environment trials of 537 potato varieties to identify the main factors driving potato dormancy and to develop predictive models for an efficient sprouting forecast. The variety explained the majority of the dormancy variability (60.3%), followed by the year (13.9%) and the location (5.4%). About 250 predictors were considered to develop a predictive model of potato dormancy. The selected model had a validation precision of 14.59 days; it used the variety class and the sum of the daily maximum temperatures in the air during the period from planting to harvest as predictors. The predictions of the selected model were supported by results of the in vivo trial using dormancy measurements from potato varieties grown under different temperature regimes in greenhouse conditions. With the growing impact of climate change on crop production, predictive models as developed here can provide an efficient and cost-effective tool to optimize the control of potato sprouting during storage.2. Strict phytosanitary measures are important to limit the impact of diseases as illustrated by the potato seed system in Europe. Cassava brown streak disease (CBSD) is a devastating disease caused by two viral species collectively named cassava brown streak viruses (CBSVs). CBSD can cause substantial root yield losses of up to 100% in the worst affected areas and is easily transmitted through stem cuttings. In Eastern and Central Africa, the epidemiology of CBSVs in the local socio-economical context of production remains poorly known while a better understanding would be an asset to properly manage the disease. This lack of information explains partially the limited efficiency of current regulatory schemes in increasing the availability of quality seed to smallholders and mitigating the spread of pests and diseases. This study surveyed the epidemiology of CBSVs in Uvira territory, Eastern D.R. Congo, and its drivers using a multivariate approach combining farmer’s interview, field observation, sampling and molecular detection of CBSVs. Investigation on the epidemiology of CBSD revealed that three clusters in the study area could be identified using five most significant factors: (i) symptoms incidence, (ii) number of whiteflies, (iii) types of foliar symptoms, (iv) cutting’s pathways and (v) plant age. Among the three clusters identified, one proved to be potentially interesting for seed multiplication activities since the disease pressure was the lowest. Through risk assessment, we also identified several key socio-economic determinants on disease epidemy: (i) factors related to farmer’s knowledge and awareness (knowledge of cassava pests and diseases, knowledge of management practices, support from extension services and management strategies applied), (ii) factors related to the geographical location of farmer’s fields (proximity to borders, proximity to town, distance to acquire cuttings), as well as (iii) the pathways used to acquire cuttings.2.
