    Exploring animacy as a mnemonic dimension

    There is a great deal of evidence across cognitive science that animacy, or more generally, the features that make up what it means to be a living thing, is a foundational dimension of human cognition. In perception, animates both capture attention (Pratt, Radulescu, Guo, & Abrams, 2010) and are relatively immune to change blindness (New, Cosmides, & Tooby, 2007). Developmental work places the animate-inanimate distinction as one of the first categories children learn (Opfer & Gelman, 2011). Work in neuroscience points toward a fundamental role for animacy in semantic memory (Caramazza & Mahon, 2003), and linguists have identified animacy as a “linguistic universal” (Comrie, 1989). Despite seemingly overwhelming evidence for the fundamental role animacy plays in human cognition, little effort has been made to understand the role of animacy in episodic memory. In three studies, the role of animacy as a dimension of word meaning was investigated. The collection of normative data for 1200 words on six scales believed to relate to the animacy construct in Study 1 set the stage for Studies 2 and 3, which explored the makeup of the animacy dimension and how it relates to other word dimensions (Study 2), and then how both animacy and other word dimensions predict free recall (Study 3). Results from Study 2 indicated that animacy is relatively independent of other word dimensions, and made up of two primary components, a mental component and a physical component. Study 3 collected recall norms from 800 participants, and regression and relative-weight analyses indicated that word animacy was consistently one of the primary predictors of free recall, with the physical component of animacy a larger predictor than the mental component. In addition to these primary results, the animacy advantage in free recall was independent of list composition (casting doubt on a distinctiveness explanation for the effect), age, and two potentially-relevant personality measures, Person and Thing Orientation (Graziano, Habashi, & Woodcock, 2011)

    Characterization of the replication of a baculovirus mutant lacking the DNA polymerase gene

    AbstractIn a previous study, the DNA polymerase gene (dnapol) of Autographa californica multiple nucleopolyhedrovirus (AcMNPV) was identified as one of six genes required for plasmid replication in a transient replication assay (M. Kool, C. Ahrens, R.W. Goldbach, G.F. Rohrmann, J.M. Vlak, Identification of genes involved in DNA replication of the Autographa californica, Proc. Natl. Acad. Sci. U.S.A. 91, (1994) 11212–11216); however, another study based on a similar approach reported that the virally encoded polymerase was only stimulatory (A. Lu, L.K. Miller, The roles of 18 baculovirus late expression factor genes in transcription and DNA replication, J. Virol. 69, (1995) 975–982). To reconcile the conflicting data and determine if the AcMNPV DNA polymerase is required for viral DNA replication during the course of an infection, a dnapol-null virus was generated using bacmid technology. To detect viral DNA replication, a highly sensitive assay was designed based on real-time PCR and SYBR green chemistry. Our results indicate that a bacmid in which the dnapol ORF was deleted is unable to replicate its DNA when transfected into Spodoptera frugiperda (Sf-9) cells, although when the dnapol ORF was introduced into the polyhedrin (polh) locus, this repaired virus could propagate at levels similar to the control virus. These results confirm that the AcMNPV-encoded DNA polymerase is required for viral DNA replication and the host DNA polymerases cannot substitute for the viral enzyme in this process

    Characterization of baculovirus constructs lacking either the Ac 101, Ac 142, or the Ac 144 open reading frame

    AbstractTo investigate the role of the gene products encoded from the open reading frames 101, 142, and 144 of Autographa californica multiple nucleopolyhedrovirus (AcMNPV), a set of bacmid knockout and repair constructs were generated. The repair genes were engineered to contain an HA epitope tag at their C-termini. The results of transfection–infection assays and growth curve analyses showed that the Ac 101, 142, and 144 genes were required for infectious virus production. To better characterize the role of these genes in the baculovirus replication cycle, quantitative DNA replication assays were performed and demonstrated that in cells transfected with the Ac 101, 142, or 144 knockouts, DNA replicated with similar kinetics as a control virus. Western blot analyses of budded virus from cells infected with the repair viruses showed that these proteins are associated with the viral nucleocapsid. Furthermore, immunoelectron microscopy of cells transfected with the knockout bacmids revealed defects in nucleocapsid production for all three constructs. From these results we concluded that the gene products encoded from these open reading frames are essential for virus production and may be involved in DNA processing, packaging, or nucleocapsid morphogenesis

    Impact of sequence variation in the ul128 locus on production of human cytomegalovirus in fibroblast and epithelial cells

    The human cytomegalovirus (HCMV) virion envelope contains a complex consisting of glycoproteins gH and gL plus proteins encoded by the UL128 locus (UL128L): pUL128, pUL130, and pUL131A. UL128L is necessary for efficient infection of myeloid, epithelial, and endothelial cells but limits replication in fibroblasts. Consequently, disrupting mutations in UL128L are rapidly selected when clinical isolates are cultured in fibroblasts. In contrast, bacterial artificial chromosome (BAC)-cloned strains TB40-BAC4, FIX, and TR do not contain overt disruptions in UL128L, yet no virus reconstituted from them has been reported to acquire mutations in UL128L in vitro. We performed BAC mutagenesis and reconstitution experiments to test the hypothesis that these strains contain subtle mutations in UL128L that were acquired during passage prior to BAC cloning. Compared to strain Merlin containing wild-type UL128L, all three strains produced higher yields of cell-free virus. Moreover, TB40-BAC4 and FIX spread cell to cell more rapidly than wild-type Merlin in fibroblasts but more slowly in epithelial cells. The differential growth properties of TB40-BAC4 and FIX (but not TR) were mapped to single-nucleotide substitutions in UL128L. The substitution in TB40-BAC4 reduced the splicing efficiency of UL128, and that in FIX resulted in an amino acid substitution in UL130. Introduction of these substitutions into Merlin dramatically increased yields of cell-free virus and increased cell-to-cell spread in fibroblasts but reduced the abundance of pUL128 in the virion and the efficiency of epithelial cell infection. These substitutions appear to represent mutations in UL128L that permit virus to be propagated in fibroblasts while retaining epithelial cell tropism

    The Effect of Psychological Distance on Willingness to Engage in Ideologically Based Violence

    Research on the cognitive and decision-making processes of individuals who choose to engage in ideologically based violence is vital. Our research examines how abstract and concrete construal mindsets affect likelihood to engage in ideologically based violence. Construal Level Theory (CLT) states that an abstract mindset (high-level construal), as opposed to a concrete mindset (low-level construal), is associated with a greater likelihood of engaging in goal-oriented, value-motivated behaviors. Assuming that ideologically based violence is goal-oriented, we hypothesized that high-level construal should result in an increased likelihood of engaging in ideologically based violence. In the pilot study we developed and tested 24 vignettes covering controversial topics and assessed them on features such as relatability, emotional impact, and capacity to elicit a violent reaction. The ten most impactful vignettes were selected for use in the primary investigations. The two primary investigations examined the effect of high- and low-level construal manipulations on self-reported likelihood of engaging in ideologically based violence. Self-reported willingness was measured through an ideological violence assessment. Data trends implied that participants were engaged in the study, as they reported a higher willingness to engage in ideologically based violence when they had a higher passion for the vignette's social issue topic. Our results did not indicate a significant relationship between construal manipulations and level of passion for a topic

    A high level of sustained personal plaque control is fundamental for successful treatment outcomes in patients with active periodontal disease and, hence, oral hygiene instructions are the cornerstone of periodontal treatment planning. Other risk factors for periodontal disease also should be identified and modified where possible. Many restorative dental treatments in particular require the establishment of healthy periodontal tissues for their clinical success. Failure by patients to control dental plaque because of inappropriate designs and materials for restorations and prostheses will result in the long-term failure of the restorations and the loss of supporting tissues. Periodontal treatment planning considerations are also very relevant to endodontic, orthodontic and osseointegrated dental implant conditions and proposed therapies.E. Corbet and R. Smale

    Unilateral cross bite treated by corticotomy-assisted expansion: two case reports

    <p>Abstract</p> <p>Background</p> <p>True unilateral posterior crossbite in adults is a challenging malocclusion to treat. Conventional expansion methods are expected to have some shortcomings. The aim of this paper is to introduce a new technique for treating unilateral posterior crossbite in adults, namely, corticotomy-assisted expansion (CAE) applied on two adult patients: one with a true unilateral crossbite and the other with an asymmetrical bilateral crossbite, both treated via modified corticotomy techniques and fixed orthodontic appliances.</p> <p>Methods</p> <p>Two cases with asymmetric maxillary constriction were treated using CAE.</p> <p>Results</p> <p>In both cases, effective asymmetrical expansion was achieved using CAE, and functional occlusion was established as well.</p> <p>Conclusions</p> <p>Unilateral CAE presents an effective and reliable technique to treat true unilateral crossbite.</p
