    Disambiguating the role of blood flow and global signal with partial information decomposition

    Global signal (GS) is an ubiquitous construct in resting state functional magnetic resonance imaging (rs-fMRI), associated to nuisance, but containing by definition most of the neuronal signal. Global signal regression (GSR) effectively removes the impact of physiological noise and other artifacts, but at the same time it alters correlational patterns in unpredicted ways. Performing GSR taking into account the underlying physiology (mainly the blood arrival time) has been proven to be beneficial. From these observations we aimed to: 1) characterize the effect of GSR on network-level functional connectivity in a large dataset; 2) assess the complementary role of global signal and vessels; and 3) use the framework of partial information decomposition to further look into the joint dynamics of the global signal and vessels, and their respective influence on the dynamics of cortical areas. We observe that GSR affects intrinsic connectivity networks in the connectome in a non-uniform way. Furthermore, by estimating the predictive information of blood flow and the global signal using partial information decomposition, we observe that both signals are present in different amounts across intrinsic connectivity networks. Simulations showed that differences in blood arrival time can largely explain this phenomenon, while using hemodynamic and calcium mouse recordings we were able to confirm the presence of vascular effects, as calcium recordings lack hemodynamic information. With these results we confirm network-specific effects of GSR and the importance of taking blood flow into account for improving de-noising methods. Additionally, and beyond the mere issue of data denoising, we quantify the diverse and complementary effect of global and vessel BOLD signals on the dynamics of cortical areas

    A structured design technique for distributed programs

    This report contains a non-formal motivation and description of ADL-d, a graphical design technique for parallel and distributed software. ADL-d allows a developer to construct an application in terms of communicating processes. The technique distinguishes itself from others by its use of highly orthogonal concepts, and support for automated code generation. Without being committed to one particular design method, ADL-d as a technique can be used from the early phases of application design through phases that concentrate on algorithmic design, and final implementation on some target platform. In this report, we discuss and motivate all ADL-d components, including recently incorporated features such as support for connection-oriented communication, support for modeling dynamically changing communication structures, and a formal semantical basis for each ADL-d component. Also, we discuss our ADL-d implementation, and place ADL-d in context by discussing some related work

    Movers and shakers of cognition:Hand movements, speech, task properties, and variability

    Children move their hands to explore, learn and communicate about hands-on tasks. Their hand movements seem to be “learning” ahead of speech. Children shape their hand movements in accordance with spatial and temporal task properties, such as when they feel an object or simulate its movements. Their speech does not directly correspond to these spatial and temporal task properties, however. We aimed to understand whether and how hand movements' are leading cognitive development due to their ability to correspond to spatiotemporal task properties, while speech is unable to do so. We explored whether hand movements' and speech's variability changed with a change in spatiotemporal task properties, using two variability measures: Diversity indicates adaptation, while Complexity indicates flexibility to adapt. In two experiments, we asked children (4–7 years) to predict and explain about balance scale problems, whereby we either manipulated the length of the balance scale or the mass of the weights after half of the trials. In three out of four conditions, we found a change in Complexity for both hand movements and speech between first and second half of the task. In one of these conditions, we found a relation between the differences in Complexity and Diversity of hand movements and speech. Changes in spatiotemporal task properties thus often influenced both hand movements' and speech's flexibility, but there seem to be differences in how they did so. We provided many directions for future research, to further unravel the relations between hand movements, speech, task properties, variability, and cognitive development

    Endothelial response to glucocorticoids in inflammatory diseases

    The endothelium plays a crucial role in inflammation. A balanced control of inflammation requires the action of glucocorticoids (GCs), steroidal hormones with potent cell-specific anti-inflammatory properties. Besides the classic anti-inflammatory effects of GCs on leukocytes, recent studies confirm that endothelial cells also represent an important target for GCs. GCs regulate different aspects of endothelial physiology including expression of adhesion molecules, production of pro-inflammatory cytokines and chemokines, and maintenance of endothelial barrier integrity. However, the regulation of endothelial GC sensitivity remains incompletely understood. In this review, we specifically examine the endothelial response to GCs in various inflammatory diseases ranging from multiple sclerosis, stroke, sepsis, and vasculitis to atherosclerosis. Shedding more light on the cross talk between GCs and endothelium will help to improve existing therapeutic strategies and develop new therapies better tailored to the needs of patients

    Self-demodulation effect on subharmonic response of ultrasound contrast agent

    In this work the use of the self-demodulation (S-D) signal as a mean of microbubble excitation at the subharmonic (SH) frequency to enhance the SH emission of ultrasound contrast agent (UCA) is studied. SH emission from the UCA is of interest since it is produced only by the UCA and is free of the artifacts produced in harmonic imaging modes. The S-D wave is a low-frequency signal produced by nonlinear propagation of an ultrasound wave in the medium. Single element transducer experiments and numerical simulations were conducted at 10 MHz to study the effect of the S-D signal on the SH response of the UCA by modifying the envelope of the excitation bursts. For 6 and 20 transmitted cycles, the SH response is increased up to 25 dB and 22 dB because of the S-D stimulation for a burst with a rectangular envelope compared with a Gaussian envelope burst. Such optimized excitations were used in an array-based micro-ultrasound system (Vevo 2100, VisualSonics Inc., Toronto, ON, Canada) at 18 MHz for in vitro validation of SH imaging. This study suggests that a suitable design of the envelope of the transmit excitation to generate a S-D signal at the SH frequency can enhance the SH emission of UCA and real-time SH imaging is feasible with shorter transmit burst (6- cycle) and low acoustic pressure (-150 KPa) at high frequencies (>15 MHz)

    Plasmodium berghei NK65 in Combination with IFN-Îł Induces Endothelial Glucocorticoid Resistance via Sustained Activation of p38 and JNK.

    Malaria-associated acute respiratory distress syndrome (MA-ARDS) is an often lethal complication of malaria. Currently, no adequate therapy for this syndrome exists. Although glucocorticoids (GCs) have been used to improve clinical outcome of ARDS, their therapeutic benefits remain unclear. We previously developed a mouse model of MA-ARDS, in which dexamethasone treatment revealed GC resistance. In the present study, we investigated GC sensitivity of mouse microvascular lung endothelial cells stimulated with interferon-Îł (IFN-Îł) and Plasmodium berghei NK65 (PbNK65). Upon challenge with IFN-Îł alone, dexamethasone inhibited the expression of CCL5 (RANTES) by 90% and both CCL2 (MCP-1) and CXCL10 (IP-10) by 50%. Accordingly, whole transcriptome analysis revealed that dexamethasone differentially affected several gene clusters and in particular inhibited a large cluster of IFN-Îł-induced genes, including chemokines. In contrast, combined stimulation with IFN-Îł and PbNK65 extract impaired inhibitory actions of GCs on chemokine release, without affecting the capacity of the GC receptor to accumulate in the nucleus. Subsequently, we investigated the effects of GCs on two signaling pathways activated by IFN-Îł. Dexamethasone left phosphorylation and protein levels of signal transducer and activator of transcription 1 (STAT1) unhampered. In contrast, dexamethasone inhibited the IFN-Îł-induced activation of two mitogen-activated protein kinases (MAPK), JNK, and p38. However, PbNK65 extract abolished the inhibitory effects of GCs on MAPK signaling, inducing GC resistance. These data provide novel insights into the mechanisms of GC actions in endothelial cells and show how malaria may impair the beneficial effects of GCs

    Photoacoustic imaging of carotid artery atherosclerosis

    We introduce a method for photoacoustic imaging of the carotid artery, tailored toward detection of lipidrich atherosclerotic lesions. A common human carotid artery was obtained at autopsy, embedded in a neck mimicking phantom and imaged with a multimodality imaging system using interstitial illumination. Light was delivered through a 1.25-mm-diameter optical probe that can be placed in the pharynx, allowing the carotid artery to be illuminated from within the body. Ultrasound imaging and photoacoustic signal detection is achieved by an external 8-MHz linear array coupled to an ultrasound imaging system. Spectroscopic analysis of photoacoustic images obtained in the wavelength range from 1130 to 1250 nm revealed plaque-specific lipid accumulation in the collagen structure of the artery wall.
