    Root, soil water and nitrogen dynamics in a catch crop - soil system in the Wageningen Rhizolab

    Catch crops (winter rye [Secale cereale] and fodder radish [Raphanus sativus]) were grown on lysimeters with rhizotron facilities in Wageningen, Netherlands, from September-March (1993-94) and August-March (1994-95) in order to study root growth and water and nitrogen dynamics under different regimes of irrigation and N supply. Catch crops took up 20-30 g N/msuperscript 2, of which 37-48% was present in dead leaves in March. Rooting depth increased by 2.6 cm/day for both species at the start of the growing season. Catch cropping reduced the NO-3-N concentration in the soil considerably, initially in the top layers and then further down the soil profile. The reduction in total leached N was similar to the total crop N uptake. Nitrate-N concentrations in leached water were reduced by 49-85 mg/litre (62-99%), depending upon N availability and irrigation. Due to catch cropping the NO-3-N concentration in the percolate decreased with increasing irrigation (or precipitation), whereas the amount of N leached increased with irrigation. Evapotranspiration from a catch cropped soil was close to the potential evapotranspiration under optimal growth conditions

    Background: Molecular characterisation of single circulating tumour cells (CTCs) holds considerable promise for predictive biomarker assessment and to explore CTC heterogeneity. We evaluate a new method, the DEPArray system, that allows the dielectrophoretic manipulation and isolation of single and 100% purified groups of CTCs from pre-enriched blood samples and explore the feasibility of their molecular characterisation.Methods:Samples containing known numbers of two cell populations were used to assess cell loss during sample loading. Cultured breast cancer cells were isolated from spiked blood samples using CellSearch CTC and Profile kits. Single tumour cells and groups of up to 10 tumour cells were recovered with the DEPArray system and subjected to transcriptional and mutation analysis.Results:On average, 40% cell loss was observed when loading samples to the DEPArray system. Expected mutations in clinically relevant markers could be obtained for 60% of single recovered tumour cells and all groups of tumour cells. Reliable gene expression profiles were obtained from single cells and groups of up to 10 cells for 2 out of 3 spiked breast cancer cell lines.Conclusion:We describe a semiautomated workflow for the isolation of small groups of 1 to 10 tumour cells from whole blood samples and provide proof of principle for the feasibility of their comprehensive molecular characterisation

    A new thin film photochromic material: Oxygen-containing yttrium hydride

    In this work we report on photochromism in transparent thin film samples of oxygen-containing yttrium hydride. Exposure to visible and ultraviolet (UV) light at moderate intensity triggers a decrease in the optical transmission of visible and infrared (IR) light. The photo-darkening is colour-neutral. We show that the optical transmission of samples of 500 nm thickness can be reduced by up to 50% after one hour of illumination with light of moderate intensity. The reaction is reversible and samples that are left in the dark return to the initial transparent state. The relaxation time in the dark depends on the temperature of the sample and the duration of the light exposure. The photochromic reaction takes place under ambient conditions in the as-deposited state of the thin-film samples.Comment: Accepted for publication in Solar Energy Materials and Solar Cell

    Molecular characterization of MRSA collected during national surveillance between 2008 and 2019 in the Netherlands

    Background.Although the Netherlands is a country with a low endemic level, methicillin-resistant Staphylococcus aureus (MRSA) poses a significant health care problem. Therefore, high coverage national MRSA surveillance has been in place since 1989. To monitor possible changes in the type-distribution and emergence of resistance and virulence, MRSA isolates are molecularly characterized.Methods.All 43,321 isolates from 36,520 persons, collected 2008–2019, were typed by multiple-locus variable number tandem repeats analysis (MLVA) with simultaneous PCR detection of the mecA, mecC and lukF-PV genes, indicative for PVL. Next-generation sequencing data of 4991 isolates from 4798 persons were used for whole genome multi-locus sequence typing (wgMLST) and identification of resistance and virulence genes.Results.We show temporal change in the molecular characteristics of the MRSA population with the proportion of PVL-positive isolates increasing from 15% in 2008–2010 to 25% in 2017–2019. In livestock-associated MRSA obtained from humans, PVL-positivity increases to 6% in 2017–2019 with isolates predominantly from regions with few pig farms. wgMLST reveals the presence of 35 genogroups with distinct resistance, virulence gene profiles and specimen origin. Typing shows prolonged persistent MRSA carriage with a mean carriage period of 407 days. There is a clear spatial and a weak temporal relationship between isolates that clustered in wgMLST, indicative for regional spread of MRSA strains.Conclusions.Using molecular characterization, this exceptionally large study shows genomic changes in the MRSA population at the national level. It reveals waxing and waning of types and genogroups and an increasing proportion of PVL-positive MRSA

    Graviton and scalar propagations on AdS(4) space in f(R) gravities

    We investigate propagations of graviton and additional scalar on four-dimensional anti de Sitter (AdS4_4) space using f(R)f(R) gravity models with external sources. It is shown that there is the van Dam-Veltman-Zakharov (vDVZ) discontinuity in f(R)f(R) gravity models because f(R)f(R) gravity implies GR with additional scalar.     Rhesus macaques self-curing from a schistosome infection can display complete immunity to challenge

    To date there is only one single drug with modest efficacy and no vaccine available to protect from schistosomiasis. Here, Amaral et al. characterize the self-cure process of rhesus macaques following primary infection and secondary challenge with Schistosoma mansoni to inform future vaccine development studies.The rhesus macaque provides a unique model of acquired immunity against schistosomes, which afflict >200 million people worldwide. By monitoring bloodstream levels of parasite-gut-derived antigen, we show that from week 10 onwards an established infection with Schistosoma mansoni is cleared in an exponential manner, eliciting resistance to reinfection. Secondary challenge at week 42 demonstrates that protection is strong in all animals and complete in some. Antibody profiles suggest that antigens mediating protection are the released products of developing schistosomula. In culture they are killed by addition of rhesus plasma, collected from week 8 post-infection onwards, and even more efficiently with post-challenge plasma. Furthermore, cultured schistosomula lose chromatin activating marks at the transcription start site of genes related to worm development and show decreased expression of genes related to lysosomes and lytic vacuoles involved with autophagy. Overall, our results indicate that enhanced antibody responses against the challenge migrating larvae mediate the naturally acquired protective immunity and will inform the route to an effective vaccine.Cancer Signaling networks and Molecular Therapeutic

    Search for composite and exotic fermions at LEP 2

    A search for unstable heavy fermions with the DELPHI detector at LEP is reported. Sequential and non-canonical leptons, as well as excited leptons and quarks, are considered. The data analysed correspond to an integrated luminosity of about 48 pb^{-1} at an e^+e^- centre-of-mass energy of 183 GeV and about 20 pb^{-1} equally shared between the centre-of-mass energies of 172 GeV and 161 GeV. The search for pair-produced new leptons establishes 95% confidence level mass limits in the region between 70 GeV/c^2 and 90 GeV/c^2, depending on the channel.
