    Early immune innate hallmarks and microbiome changes across the gut during Escherichia coli O157: H7 infection in cattle

    The zoonotic enterohemorrhagic Escherichia coli (EHEC) O157: H7 bacterium causes diarrhea, hemorrhagic colitis, and hemolytic uremic syndrome (HUS) in humans. Cattle are primary reservoirs and EHEC O157: H7; the bacteria predominately inhabit the colon and recto-anal junctions (RAJ). The early innate immune reactions in the infected gut are critical in the pathogenesis of EHEC O157: H7. In this study, calves orally inoculated with EHEC O157: H7 showed infiltration of neutrophils in the lamina propria of ileum and RAJ at 7 and 14 days post-infection. Infected calves had altered mucin layer and mast cell populations across small and large intestines. There were differential transcription expressions of key bovine β defensins, tracheal antimicrobial peptide (TAP) in the ileum, and lingual antimicrobial peptide (LAP) in RAJ. The main Gram-negative bacterial/LPS signaling Toll-Like receptor 4 (TLR4) was downregulated in RAJ. Intestinal infection with EHEC O157: H7 impacted the gut bacterial communities and influenced the relative abundance of Negativibacillus and Erysipelotrichaceae in mucosa-associated bacteria in the rectum. Thus, innate immunity in the gut of calves showed unique characteristics during infection with EHEC O157: H7, which occurred in the absence of major clinical manifestations but denoted an active immunological niche.Fil: Larzabal, Mariano.     Study of the BΛc+ΛˉcKB^{-} \to \Lambda_{c}^{+} \bar{\Lambda}_{c}^{-} K^{-} decay

    The decay BΛc+ΛˉcKB^{-} \to \Lambda_{c}^{+} \bar{\Lambda}_{c}^{-} K^{-} is studied in proton-proton collisions at a center-of-mass energy of s=13\sqrt{s}=13 TeV using data corresponding to an integrated luminosity of 5 fb1\mathrm{fb}^{-1} collected by the LHCb experiment. In the Λc+K\Lambda_{c}^+ K^{-} system, the Ξc(2930)0\Xi_{c}(2930)^{0} state observed at the BaBar and Belle experiments is resolved into two narrower states, Ξc(2923)0\Xi_{c}(2923)^{0} and Ξc(2939)0\Xi_{c}(2939)^{0}, whose masses and widths are measured to be m(Ξc(2923)0)=2924.5±0.4±1.1MeV,m(Ξc(2939)0)=2938.5±0.9±2.3MeV,Γ(Ξc(2923)0)=0004.8±0.9±1.5MeV,Γ(Ξc(2939)0)=0011.0±1.9±7.5MeV, m(\Xi_{c}(2923)^{0}) = 2924.5 \pm 0.4 \pm 1.1 \,\mathrm{MeV}, \\ m(\Xi_{c}(2939)^{0}) = 2938.5 \pm 0.9 \pm 2.3 \,\mathrm{MeV}, \\ \Gamma(\Xi_{c}(2923)^{0}) = \phantom{000}4.8 \pm 0.9 \pm 1.5 \,\mathrm{MeV},\\ \Gamma(\Xi_{c}(2939)^{0}) = \phantom{00}11.0 \pm 1.9 \pm 7.5 \,\mathrm{MeV}, where the first uncertainties are statistical and the second systematic. The results are consistent with a previous LHCb measurement using a prompt Λc+K\Lambda_{c}^{+} K^{-} sample. Evidence of a new Ξc(2880)0\Xi_{c}(2880)^{0} state is found with a local significance of 3.8σ3.8\,\sigma, whose mass and width are measured to be 2881.8±3.1±8.5MeV2881.8 \pm 3.1 \pm 8.5\,\mathrm{MeV} and 12.4±5.3±5.8MeV12.4 \pm 5.3 \pm 5.8 \,\mathrm{MeV}, respectively. In addition, evidence of a new decay mode Ξc(2790)0Λc+K\Xi_{c}(2790)^{0} \to \Lambda_{c}^{+} K^{-} is found with a significance of 3.7σ3.7\,\sigma. The relative branching fraction of BΛc+ΛˉcKB^{-} \to \Lambda_{c}^{+} \bar{\Lambda}_{c}^{-} K^{-} with respect to the BD+DKB^{-} \to D^{+} D^{-} K^{-} decay is measured to be 2.36±0.11±0.22±0.252.36 \pm 0.11 \pm 0.22 \pm 0.25, where the first uncertainty is statistical, the second systematic and the third originates from the branching fractions of charm hadron decays.Comment: All figures and tables, along with any supplementary material and additional information, are available at https://cern.ch/lhcbproject/Publications/p/LHCb-PAPER-2022-028.html (LHCb public pages

    Measurement of the ratios of branching fractions R(D)\mathcal{R}(D^{*}) and R(D0)\mathcal{R}(D^{0})

    The ratios of branching fractions R(D)B(BˉDτνˉτ)/B(BˉDμνˉμ)\mathcal{R}(D^{*})\equiv\mathcal{B}(\bar{B}\to D^{*}\tau^{-}\bar{\nu}_{\tau})/\mathcal{B}(\bar{B}\to D^{*}\mu^{-}\bar{\nu}_{\mu}) and R(D0)B(BD0τνˉτ)/B(BD0μνˉμ)\mathcal{R}(D^{0})\equiv\mathcal{B}(B^{-}\to D^{0}\tau^{-}\bar{\nu}_{\tau})/\mathcal{B}(B^{-}\to D^{0}\mu^{-}\bar{\nu}_{\mu}) are measured, assuming isospin symmetry, using a sample of proton-proton collision data corresponding to 3.0 fb1{ }^{-1} of integrated luminosity recorded by the LHCb experiment during 2011 and 2012. The tau lepton is identified in the decay mode τμντνˉμ\tau^{-}\to\mu^{-}\nu_{\tau}\bar{\nu}_{\mu}. The measured values are R(D)=0.281±0.018±0.024\mathcal{R}(D^{*})=0.281\pm0.018\pm0.024 and R(D0)=0.441±0.060±0.066\mathcal{R}(D^{0})=0.441\pm0.060\pm0.066, where the first uncertainty is statistical and the second is systematic. The correlation between these measurements is ρ=0.43\rho=-0.43.     Mycobacterium bovis Dmce2 double deletion mutant protects cattle against challenge with virulent M. bovis

    A Mycobacterium bovis strain deleted in mce2A and mce2B genes (M. bovis Δmce2) was tested as an experimental vaccine in cattle challenged with a virulent M. bovis strain. Three-and-a-half-month old calves (n = 5 to 6 per group) were vaccinated and challenged with a virulent strain of M. bovis by the intratracheal route 9 weeks after vaccination. A non-vaccinated group and a group vaccinated with BCG were included as controls. Blood samples were collected to measure IFN-γ by an interferon-gamma release assay (IGRA), cytometry and cytokine responses of bovine purified protein derivative (PPD) restimulated peripheral blood mononuclear cells (PBMCs). The IGRA test showed IFN-γ values similar to pre-vaccination except for the animals vaccinated with M. bovis Δmce2, where a significant increase was observed at 30 days post-vaccination. The expression of IL-2R on CD4+ cells in response to PPD from the animals vaccinated with Δmce2 increased at 15 days post-vaccination compared to cells from non-vaccinated group. Vaccination of cattle with M. bovis Δmce2 induced the highest (P < 0.05) expression of IFN-γ and IL-17 mRNA upon PPD stimulation of PBMCs compared to vaccination with BCG or that for the non-vaccinated group. There was a weak positive correlation between the production of these proinflammatory cytokines post-vaccination and reduced pathology scores post-challenge. The animals were euthanized and necropsied 100 days after challenge. The group vaccinated with M. bovis Δmce2 displayed a significantly lower histopathological score for lesions in lungs and pulmonary lymph nodes than for the other groups (P < 0.05). A marked positive reaction to tuberculin intradermal test was observed post-vaccination in animals vaccinated with M. bovis Δmce2 compared to those vaccinated with BCG or the non-vaccinated group. In contrast, after challenge, non-vaccinated animals had greater skin test responses than the vaccinated animals.     Decays of Ξb\Xi_{b}^{-} and Ωb\Omega_{b}^{-} baryons to Λc+hh\Lambda_{c}^{+} h^- h^{\prime -} final states, with hhh^- h^{\prime -} being ππ\pi^{-}\pi^{-}, KπK^{-}\pi^{-} and KKK^{-}K^{-} meson pairs, are searched for using data collected with the LHCb detector. The data sample studied corresponds to an integrated luminosity of 8.7fb18.7 \rm fb^{-1} of pppp collisions collected at centre-of-mass energies s=7\sqrt{s} = 7, 88 and 13TeV13 \rm TeV. The products of the relative branching fractions and fragmentation fractions for each signal mode, relative to the BΛc+pˉπB^{-} \rightarrow \Lambda_{c}^{+} \bar{p} \pi^{-} mode, are measured, with ΞbΛc+Kπ\Xi_{b}^{-} \rightarrow\Lambda_{c}^{+} K^{-} \pi^{-}, ΞbΛc+KK\Xi_{b}^{-} \rightarrow\Lambda_{c}^{+} K^{-} K^{-} and ΩbΛc+KK\Omega_{b}^{-} \rightarrow\Lambda_{c}^{+} K^{-} K^{-} decays being observed at over 5σ5\,\sigma significance. The ΞbΛc+Kπ\Xi_{b}^{-} \rightarrow\Lambda_{c}^{+} K^{-} \pi^{-} mode is also used to measure the Ξb\Xi_{b}^{-} production asymmetry, which is found to be consistent with zero.     A measurement of the transverse polarization of the Λ\Lambda and Λˉ\bar{\Lambda}hyperons in ppNe fixed-target collisions at sNN\sqrt{s_{NN}}=68.4 GeV is presented using data collected by the LHCb detector.     Tracking of charged particles with nanosecond lifetimes at LHCb

    A method is presented to reconstruct charged particles with lifetimes between 10 ps and 10 ns, which considers a combination of their decay products and the partial tracks created by the initial charged particle. Using the Ξ\Xi^- baryon as a benchmark, the method is demonstrated with simulated events and proton-proton collision data at s=13\sqrt{s}=13 TeV, corresponding to an integrated luminosity of 2.0 fb1{}^{-1} collected with the LHCb detector in 2018. Significant improvements in the angular resolution and the signal purity are obtained. The method is implemented as part of the LHCb Run 3 event trigger in a set of requirements to select detached hyperons. This is the first demonstration of the applicability of this approach at the LHC, and the first to show its scaling with instantaneous luminosity

By considering the Λb0Λc+D0K\Lambda_b^0 \rightarrow \Lambda_c^{+} \overline{D}^0 K^{-} decay as reference channel, the following branching fraction ratios are measured to be, B(Λb0Σc++DK)B(Λb0Λc+D0K)=0.282±0.016±0.016±0.005,B(Λb0Σc++DK)B(Λb0Σc++DK)=0.460±0.052±0.028,B(Λb0Σc++DK)B(Λb0Σc++DK)=2.261±0.202±0.129±0.046,B(Λb0Σc++DK)B(Λb0Σc++DK)=0.896±0.137±0.066±0.018,\frac{\cal{B} (\Lambda_{b}^{0} \rightarrow \Sigma_{c}^{++} \rm{D}^{-} {K}^{-})}{\cal{B}(\Lambda_{b}^{0} \rightarrow \Lambda_c^{+} \rm \overline{D}^0 {K}^{-})} = {0.282}\pm{0.016}\pm{0.016}\pm{0.005}, \frac{\cal{B}(\Lambda_{b}^{0} \rightarrow \Sigma_{c}^{*++} \rm {D}^{-} {K}^{-})}{\cal{B}(\Lambda_{b}^{0} \rightarrow \Sigma_c^{++} \rm {D}^{-} {K}^{-})} = {0.460}\pm{0.052}\pm{0.028}, \frac{\cal{B}(\Lambda_{b}^{0} \rightarrow \Sigma_{c}^{++} \rm {D}^{*-} {K}^{-})}{\cal{B}(\Lambda_{b}^{0} \rightarrow \Sigma_c^{++} \rm {D}^{-} {K}^{-})} = {2.261}\pm{0.202}\pm{0.129}\pm{0.046}, \frac{\cal{B}(\Lambda_{b}^{0} \rightarrow \Sigma_{c}^{*++} \rm D^{*-} K^{-})}{\cal{B}(\Lambda_{b}^{0} \rightarrow \Sigma_c^{++} \rm D^{-} K^{-})} = {0.896}\pm{0.137}\pm{0.066}\pm{0.018}, where the first uncertainties are statistical, the second are systematic, and the third are due to uncertainties in the branching fractions of intermediate particle decays. These initial observations mark the beginning of pentaquark searches in these modes, with more data set to become available following the LHCb upgrade
