    Comparative In Vitro Toxicity Evaluation of Heavy Metals (Lead, Cadmium, Arsenic, and Methylmercury) on HT-22 Hippocampal Cell Line

    Heavy metals are considered some of the most toxic environmental pollutants. Exposure to heavy metals including lead (Pb), cadmium (Cd), arsenic (As), and methyl mercury (MeHg) has long been known to cause damage to human health. Many recent studies have supported the hippocampus as the major target for these four metals for inflicting cognitive dysfunction. In the present study, we proposed hippocampal relevant in vitro toxicity of Pb, Cd, As, and MeHg in HT-22 cell line. This study reports, initially, cytotoxic effects in acute, subchronic, chronic exposures. We further investigated the mechanistic potency of DNA damage and apoptosis damage with the observed cytotoxicity. The genotoxicity and apoptosis were measured by using the comet assay, annexin-V FTIC / propidium iodide (PI) assay, respectively. The results of cytotoxicity assay clearly demonstrated significant concentration and time-dependent effects on HT-22 cell line. The genotoxic and apoptosis effects also concentration-dependent fashion with respect to their potency in the range of IC10-IC30, maximal level of damage observed in MeHg. In conclusion, the obtained result suggests concentration and potency-dependent response; the maximal level of toxicity was observed in MeHg. These novel findings support that Pb, Cd, As, and MeHg induce cytotoxic, genotoxic, and apoptotic effects on HT-22 cells in potency-dependent manner; MeHg> As> Cd> Pb. Therefore, the toxicity of Pb, Cd, As, and MeHg could be useful for knowing the common underlying molecular mechanism, and also for estimating the mixture impacts on HT-22 cell line

    DEVELOPMENT OF OSMOTICALLY CONTROLLED ORAL DRUG DELIVERY SYSTEM FOR NATEGLINIDE AN ANTI-DIABETIC DRUG

    Objective: The purpose of the present study was to develop an oral push-pull osmotic drug delivery system for the drug Nateglinide which is a bio pharmaceutics classification system (BCS) class II drug. Methods: The tablets were prepared by the wet granulation method using ingredients microcrystalline cellulose (Adsorbent), potassium chloride (Osmotic agent), poly ethylene glycol (4000 and 6000) (Hydrophilic polymer, Plasticizer), starch (Disintegrant), and aerosil. The granules were compacted by double compression method and were coated with eudragit by dipping method. Different batches were prepared to study the effect of the various ingredients and their effect on the release of the drug from the system by varying the concentrations of the ingredients in each batch. Dissolution was assessed using USP dissolution apparatus 2 in phosphate buffer pH 6.8 for 12 h. Results: Certain key findings observed includes a decrease in micro crystalline cellulose content reduced the release of the drug due to the reduction of the hydrophilic content in the tablet which complements the uptake of water from the surroundings, and increase in the ethylene glycol leads to decrease in the release which resulted due to excess swelling and increase in the osmotic agent concentration lead to satisfactory release of the drug and followed zero-order release. Conclusion: To conclude, the push-pull osmotic tablet of Nateglinide was able to deliver the drug in a controlled pattern for a prolonged period of time. This type of formulation can be used in conditions like hyperglycemia where the patient compliance can improve by reducing the dosing frequency and the plasma drug levels can be maintained, the total drug load is also reduced so that the dose related side-effects are also reduced. Keywords: Controlled release, Push-pull osmotic pump, Nateglinid

    RNA interference in marine and freshwater sponges

    Background: The marine sponge Tethya wilhelma and the freshwater sponge Ephydatia muelleri are emerging model organisms to study evolution, gene regulation, development, and physiology in non-bilaterian animal systems. Thus far, functional methods (i.e., loss or gain of function) for these organisms have not been available. Results: We show that soaking developing freshwater sponges in double-stranded RNA and/or feeding marine and freshwater sponges bacteria expressing double-stranded RNA can lead to RNA interference and reduction of targeted transcript levels. These methods, first utilized in C. elegans, have been adapted for the development and feeding style of easily cultured marine and freshwater poriferans. We demonstrate phenotypic changes result from ‘knocking down’ expression of the actin gene. Conclusion: This technique provides an easy, efficient loss-of-function manipulation for developmental and gene regulatory studies in these important non-bilaterian animals

    Is Split Manufacturing Secure?

    Abstract-Split manufacturing of integrated circuits (IC) is being investigated as a way to simultaneously alleviate the cost of owning a trusted foundry and eliminate the security risks associated with outsourcing IC fabrication. In split manufacturing, a design house (with a low-end, in-house, trusted foundry) fabricates the Front End Of Line (FEOL) layers (transistors and lower metal layers) in advanced technology nodes at an untrusted high-end foundry. The Back End Of Line (BEOL) layers (higher metal layers) are then fabricated at the design house's trusted low-end foundry. Split manufacturing is considered secure (prevents reverse engineering and IC piracy) as it hides the BEOL connections from an attacker in the FEOL foundry. We show that an attacker in the FEOL foundry can exploit the heuristics used in typical floorplanning, placement, and routing tools to bypass the security afforded by straightforward split manufacturing. We developed an attack where an attacker in the FEOL foundry can connect 96% of the missing BEOL connections correctly. To overcome this security vulnerability in split manufacturing, we developed a fault analysis-based defense. This defense improves the security of split manufacturing by deceiving the FEOL attacker into making wrong connections

    Bedside Nurses\u27 Perceptions of Effective Nurse-Physician Communication in General Medical Units: A Qualitative Study

    Background There is a dearth of research on successful interventions to improve nurse-physician communication (NPC). An important step is identifying what matters to bedside nurses and their perceptions of effective NPC communications and actions. Methods We conducted three focus groups with a total of 19 medical unit nurses across two hospitals in one academic medical center in the United States. Using a convenience sampling strategy, five to eight nurses voluntarily participated in each focus group. The recording was transcribed verbatim and two independent coders performed coding and resolved any discrepancies in codes. Qualitative content analysis was pursued to identify themes and associated quotes. Results The presence of direct communication between physicians and nurses was identified as the first theme and perceived by nurses as very important. Additional themes related to physician communication and attributes emerged including collegiality and respect (e.g., engaging nurses as partners in patient care), attentiveness and responsiveness (e.g., listening carefully and addressing concerns), and directness and support (e.g., backing nurses up in difficult situations). Effective NPC is further facilitated by organizational structure, relationship development separate from patient care, and consistent/timely use of technology. Conclusions Hospital bedside nurses provided valuable insight into improved physician communication and what attributes contribute to more effective NPC. Most importantly, they emphasized the significance of physicians in supporting them with difficult patients
