    Loss of the arabidopsis protein kinases ANPs affects root cell wall composition, and triggers the cell wall damage syndrome

    The Arabidopsis NPK1-related Protein kinases ANP1, ANP2 and ANP3 belong to the MAP kinase kinase kinase (MAPKKK) superfamily and were previously described to be crucial for cytokinesis, elicitor-induced immunity and development. Here we investigate the basis of their role in development by using conditional β-estradiol-inducible triple mutants to overcome lethality. In seedlings, lack of ANPs causes root cell bulging, with the transition zone being the most sensitive region. We uncover a role of ANPs in the regulation of cell wall composition and suggest that developmental defects of the triple mutants, observed at the cellular level, might be a consequence of the alterations of the pectic and cellulosic cell wall components. Lack of ANPs also induced a typical cell wall damage syndrome (CWDS) similar to that observed in plants treated with the cellulose biosynthesis inhibitor isoxaben (ISX). Moreover, anp double mutants and plants overexpressing single ANPs (ANP1 or ANP3) respectively showed increased and reduced accumulation of jasmonic acid and PDF1.2 transcripts upon ISX treatment, suggesting that ANPs are part of the pathway targeted by this inhibitor and play a role in cell wall integrity surveillance

    Stenosis triggers spread of helical Pseudomonas biofilms in cylindrical flow systems

    Biofilms are multicellular bacterial structures that adhere to surfaces and often endow the bacterial population with tolerance to antibiotics and other environmental insults. Biofilms frequently colonize the tubing of medical devices through mechanisms that are poorly understood. Here we studied the helicoidal spread of Pseudomonas putida biofilms through cylindrical conduits of varied diameters in slow laminar flow regimes. Numerical simulations of such flows reveal vortical motion at stenoses and junctions, which enhances bacterial adhesion and fosters formation of filamentous structures. Formation of long, downstream-flowing bacterial threads that stem from narrowings and connections was detected experimentally, as predicted by our model. Accumulation of bacterial biomass makes the resulting filaments undergo a helical instability. These incipient helices then coarsened until constrained by the tubing walls, and spread along the whole tube length without obstructing the flow. A three-dimensional discrete filament model supports this coarsening mechanism and yields simulations of helix dynamics in accordance with our experimental observations. These findings describe an unanticipated mechanism for bacterial spreading in tubing networks which might be involved in some hospital-acquired infections and bacterial contamination of catheters

    Glenoid fossa position in Class II malocclusion associated with mandibular retrusion

    Abstract Objective: To assess the position of the glenoid fossa in subjects with Class II malocclusion associated with mandibular retrusion and normal mandibular size in the mixed dentition. Materials and Methods: A sample of 30 subjects (16 male, 14 female), age 9 years ± 6 months, with skeletal and dental Class II malocclusion associated with mandibular retrusion, normal skeletal vertical relationships, and normal mandibular dimensions, was compared with a matched group of 37 subjects (18 male, 19 female) with skeletal and dental Class I relationships. The comparisons between the Class II group and the control group on the cephalometric measures for the assessment of glenoid fossa position were performed by means of a nonparametric test for independent samples (Mann-Whitney U-test, P < .05). Results: Subjects with Class II malocclusion presented with a significantly more distal position of the glenoid fossa, when compared with the control group as measured by means of three parameters (GF-S on FH, GF-Ptm on FH, and GF-FMN). Conclusions: A posteriorly displaced glenoid fossa is a possible diagnostic feature of Class II malocclusion associated with mandibular retrusion. An effective cephalometric measurement to evaluate glenoid fossa position is the distance from the glenoid fossa to the frontomaxillonasal suture (GF-FMN)

    Full Counting Statistics of Non-Commuting Variables: the Case of Spin Counts

    We discuss the Full Counting Statistics of non-commuting variables with the measurement of successive spin counts in non-collinear directions taken as an example. We show that owing to an irreducible detector back-action, the FCS in this case may be sensitive to the dynamics of the detectors, and may differ from the predictions obtained with using a naive version of the Projection Postulate. We present here a general model of detector dynamics and path-integral approach to the evaluation of FCS. We concentrate further on a simple "diffusive" model of the detector dynamics where the FCS can be evaluated with transfer-matrix method. The resulting probability distribution of spin counts is characterized by anomalously large higher cumulants and substantially deviates from Gaussian Statistics.Comment: 11 pages, 3 figure

    Minerva: A File-Based Ransomware Detector

    Ransomware is a rapidly evolving type of malware designed to encrypt user files on a device, making them inaccessible in order to exact a ransom. Ransomware attacks resulted in billions of dollars in damages in recent years and are expected to cause hundreds of billions more in the next decade. With current state-of-the-art process-based detectors being heavily susceptible to evasion attacks, no comprehensive solution to this problem is available today. This paper presents Minerva, a new approach to ransomware detection. Unlike current methods focused on identifying ransomware based on process-level behavioral modeling, Minerva detects ransomware by building behavioral profiles of files based on all the operations they receive in a time window. Minerva addresses some of the critical challenges associated with process-based approaches, specifically their vulnerability to complex evasion attacks. Our evaluation of Minerva demonstrates its effectiveness in detecting ransomware attacks, including those that are able to bypass existing defenses. Our results show that Minerva identifies ransomware activity with an average accuracy of 99.45% and an average recall of 99.66%, with 99.97% of ransomware detected within 1 second.Comment: 19 pages, 3 figure

    Interference of dibutylphthalate on human prostate cell viability

    Dibutylphthalate (DBP) is an environmental pollutant widely used as plasticizer in a variety of industrial applications worldwide. This agent can be found in personal-care products, children's toy, pharmaceuticals, food products. Exposure to DBP can occur via ingestion and inhalation as well as intravenous or skin contact. DBP belongs to the family of endocrine disrupting chemicals (EDCs) and its effects on reproductive system were demonstrated both in vivo and in vitro. In the present study we evaluated the effects of DBP on human prostate adenocarcinoma epithelial cells (LNCaP) in order to highlight xenoestrogens influence on human prostate. Moreover, we have compared DBP effects with 17β-estradiol action in order to investigate possible mimetical behaviour. We have assessed the effects of both compounds on the cell viability. After then, we have evaluated the expression of genes and proteins involved in cell cycle regulation. Furthermore, we have observed the expression and the cell localization of estrogen (ERs) and androgen (AR) receptors. In conclusion, we have demonstrated that DBP interacts with estrogen hormonal receptor pathway but differently from E2. DBP alters the normal gland physiology and it is involved in the deregulation of prostate cell cycle

    Alkyphenol exposure alters steroidogenesis in male lizard podarcis siculus

    Background: Nonylphenol (NP) and Octylphenol (OP) are persistent and non-biodegradable environmental contaminants classified as endocrine disruptor chemicals (EDCs). These compounds are widely used in several industrial applications and present estrogen-like properties, which have extensively been studied in aquatic organisms. The present study aimed to verify the interference of these compounds alone, and in mixture, on the reproductive cycle of the male terrestrial vertebrate Podarcis siculus, focusing mainly on the steroidogenesis process. Methods: Male lizards have been treated with different injections of both NP and OP alone and in mixture, and evaluation has been carried out using a histological approach. Results: Results obtained showed that both substances are able to alter both testis histology and localization of key steroidogenic enzymes, such as 3β-hydroxysteroid dehydrogenase (3β-HSD), 17β-hydroxysteroid dehydrogenase (17β-HSD) and P450 aromatase. Moreover, OP exerts a preponderant effect, and the P450 aromatase represents the major target of both chemicals. Conclusions: In conclusion, NP and OP inhibit steroidogenesis, which in turn may reduce the reproductive capacity of the specimens

    Conditional score-based diffusion models for Bayesian inference in infinite dimensions

    Since their first introduction, score-based diffusion models (SDMs) have been successfully applied to solve a variety of linear inverse problems in finite-dimensional vector spaces due to their ability to efficiently approximate the posterior distribution. However, using SDMs for inverse problems in infinite-dimensional function spaces has only been addressed recently and by learning the unconditional score. While this approach has some advantages, depending on the specific inverse problem at hand, in order to sample from the conditional distribution it needs to incorporate the information from the observed data with a proximal optimization step, solving an optimization problem numerous times. This may not be feasible in inverse problems with computationally costly forward operators. To address these limitations, in this work we propose a method to learn the posterior distribution in infinite-dimensional Bayesian linear inverse problems using amortized conditional SDMs. In particular, we prove that the conditional denoising estimator is a consistent estimator of the conditional score in infinite dimensions. We show that the extension of SDMs to the conditional setting requires some care because the conditional score typically blows up for small times contrarily to the unconditional score. We also discuss the robustness of the learned distribution against perturbations of the observations. We conclude by presenting numerical examples that validate our approach and provide additional insights

    Combinatorial experimental protocols for Erbicin-derived immunoagents and Herceptin

    Erbicin is a human anti-ErbB2 single-chain antibody fragment with high affinity and selectivity for ErbB2-positive cancer cells. Two anti-ErbB2 immunoconjugates, called Erb-hRNase and Erb-hcAb, have been prepared and found to be selectively cytotoxic on ErbB2-positive cancer cells in vitro and vivo. In Erb-hRNase, Erbicin is linked to a human RNase and in Erb-hcAb it is linked to the key structural and functional regions of a human IgG. Herceptin is an anti-ErbB2 humanised antibody successfully used in the immunotherapy of breast cancer. We report here that the Erbicin-derived immunoagents target on breast cancer cells an ErbB2 epitope different than that of Herceptin. This finding led us to verify the effects of Herceptin on breast cancer cells when it was used in combination with the Erbicin-derived immunoagents. The results indicated that in combination experiments the antitumour action of Herceptin and that of the novel agents were significantly increased in an additive fashion. An inspection of the mechanism of action of Erb-hRNase or Erb-hcAb combined with Herceptin provided evidence that the antibody combinations engendered an increased downregulation of the ErbB2 receptor, and led to an enhanced apoptotic cell death
