    Skistodiaptomus pallidus (Copepoda: Diaptomidae) establishment in New Zealand natural lakes, and its effects on zooplankton community composition

    The North American calanoid copepod Skistodiaptomus pallidus is an emerging invader globally, with non-indigenous populations recorded from constructed waters in New Zealand, Germany and Mexico since 2000. We examined the effects of S. pallidus establishment on the zooplankton community of a natural lake, Lake Kereta, where it was first recorded in late-2008, coincident with releases of domestically cultured grass carp (Ctenopharyngodon idella). Although not present in any of our samples prior to August 2008, S. pallidus was found in all samples collected in the subsequent five years. ANOSIM indicated zooplankton community composition significantly differed between samples collected before and after S. pallidus invasion, whether the invader was included in the analysis or not. Zooplankton species affected most greatly were the copepods Calamoecia lucasi and Mesocyclops sp., which decreased in their relative importance, and the cladocerans Bosmina meridionalis and Daphnia galeata, which increased. Rotifer species were relatively unaffected. As the length of grass carp released were >6.5 cm, direct predatory effects by this species on the zooplankton community are unlikely. Associated reductions in macrophyte biomass could explain increases in the relative abundances of planktonic cladocerans (B. meridionalis and D. galeata). However, the effect of macrophyte reduction by grass carp on zooplankton communities is considered to be limited elsewhere, while the reduced macrophyte biomass cannot explain the decrease in relative abundance of the native planktonic calanoid copepod C. lucasi. Competition between C. lucasi and S. pallidus is the most compelling explanation for the reduction in importance of the native calanoid copepod species. Skistodiaptomus pallidus appears to have undergone a “boom-and-bust” cycle in Lake Kereta, increasing in relative abundance in the first three years following establishment, before declining in importance

    Identifying invertebrate invasions using morphological and molecular analyses: North American Daphnia ‘pulex’ in New Zealand fresh waters

    We used a DNA barcoding approach to identify specimens of the Daphnia pulex complex occurring in New Zealand lakes, documenting the establishment of non-indigenous North American Daphnia 'pulex'. Morphological delineation of species in this complex is problematic due to a lack of good morphological traits to distinguish the species, as there is a relatively high degree of morphological stasis within the group through evolutionary time. Accordingly, genetic analyses were used to determine the specific identity and likely geographic origin of this species. Morphologically, individuals most closely resembled Daphnia pulicaria or Daphnia pulex sensu lato, which cannot be separated morphologically. Furthermore, each of these taxa comprises separate species in North America and Europe, despite carrying the same names. We identified individuals using a 658 bp nucleotide portion of the mitochondrial cytochrome c oxidase subunit 1 gene (COI) as North American Daphnia 'pulex', being distinct from European Daphnia pulex sensu stricto and D. pulicaria from Europe or North America. Cellulose allozyme electrophoresis was used to confirm that individuals were not hybrids with D. pulicaria. North American Daphnia 'pulex' in New Zealand were first recorded in New Zealand from South Island lakes that are popular for overseas recreational fishers, indicating a possible source of introduction for this species (e.g. on/in fishing gear). Our study provides an additional example of how genetic techniques can be used for the accurate identification of non-indigenous taxa, particularly when morphological species determination is not possible. The growth of global databases such as GenBank and Barcode of Life Datasystems (BOLD) will further enhance this identification capacity

    In cricket, the right to make an important decision (bat first or field first) is assigned via a coin toss. These "randomised trials" allow us to examine the consistency of choices made by teams with strictly opposed preferences, and the effects of these choices upon game outcomes. Random assignment allows us to consistently aggregate across matches, ensuring that our tests have power. We find significant evidence of inconsistency, with teams often agreeing on who is to bat first. Choices are often poorly made and reduce the probability of the team winning, a surprising finding given the intense competition and learning opportunities.     How to Perform Umbilical Cord Arterial and Venous Blood Sampling in Neonatal Foals

    Umbilical cord arterial and venous blood gas analysis is a commonly performed procedure in human neonatal medicine to help ascertain a newborn infant’s oxygenation and acid-base status prior to birth. Defined protocols for performing the procedure have been described in the medical literature. The aim of this report was to describe in detail the procedure for collecting paired blood samples from the umbilical artery and vein in newborn foals so that stall-side blood gas analysis could be carried out. Thirty-five Thoroughbred foals >320 days gestation from mares at one stud farm were sampled. Paired umbilical arterial and venous whole-blood samples were obtained in n=30 foals, umbilical artery only samples obtained in n=3 and umbilical vein only samples obtained in n=2 foals. There were no adverse events or clinical outcomes associated with the sampling protocol described. The authors found that umbilical cord blood collection for blood gas analysis was a practical clinical technique that potentially could be used as a stall-side method for assessing the in utero oxygenation and acid-base status of newborn foals

    Transcriptional profiling of zebrafish identifies host factors controlling susceptibility to Shigella flexneri

    Shigella flexneri is a human-adapted pathovar of Escherichia coli that can invade the intestinal epithelium, causing inflammation and bacillary dysentery. Although an important human pathogen, the host response to S. flexneri has not been fully described. Zebrafish larvae represent a valuable model to study human infections in vivo. Here we use a Shigella-zebrafish infection model to generate mRNA expression profiles of host response to Shigella infection at the whole animal level. Immune response-related processes dominate the signature of early Shigella infection (6 hours post-infection). Consistent with its clearance from the host, the signature of late Shigella infection (24 hours post-infection) is significantly changed, and only a small set of immune-related genes remain differentially expressed, including acod1 and gpr84. Using mutant lines generated by ENU, CRISPR mutagenesis and F0 Crispants, we show that acod1- and gpr84-deficient larvae are more susceptible to Shigella infection. Together, these results highlight the power of zebrafish to model infection by bacterial pathogens and reveal the mRNA expression of the early (acutely infected) and late (clearing) host response to Shigella infection

    Towards modelling language innovation acceptance in online social networks

    Language change and innovation is constant in online and offline communication, and has led to new words entering people’s lexicon and even entering modern day dictionaries, with recent additions of ‘e-cig’ and ‘vape’. However the manual work required to identify these ‘innovations’ is both time consuming and subjective. In this work we demonstrate how such innovations in language can be identified across two different OSN’s (Online Social Networks) through the operationalisation of known language acceptance models that incorporate relatively simplistic statistical tests. From grounding our work in language theory, we identified three statistical tests that can be applied, variation in; frequency, form and meaning; each showing different success rates across the two networks (Geo-bound Twitter sample and a sample of Reddit). These tests were also applied to different community levels within the two networks allow- ing for different innovations to be identified across different community structures over the two networks, for instance: identifying regional variation across Twitter, and variation across groupings of Subreddits, where identified example in- novations included ‘casualidad’ and ‘cym’
