    Poly(I:C) induces intense expression of c-IAP2 and cooperates with an IAP inhibitor in induction of apoptosis in cancer cells

    <p>Abstract</p> <p>Background</p> <p>There is increasing evidence that the toll-like receptor 3 (TLR3) is an interesting target for anti-cancer therapy. Unfortunately, most laboratory investigations about the impact of TLR3 stimulation on human malignant cells have been performed with very high concentrations - 5 to 100 ÎŒg/ml - of the prototype TLR3 ligand, poly(I:C). In a previous study focused on a specific type of human carcinoma - nasopharyngeal carcinoma - we have shown that concentrations of poly(I:C) as low as 100 ng/ml are sufficient to induce apoptosis of malignant cells when combined to a pharmacological antagonist of the IAP family based on Smac mimicry.</p> <p>Methods</p> <p>This observation prompted us to investigate the contribution of the IAP family in cell response to poly(I:C) in a variety of human malignant cell types.</p> <p>Results</p> <p>We report a rapid, intense and selective increase in c-IAP2 protein expression observed under stimulation by poly(I:C)(500 ng/ml) in all types of human malignant cells. In most cell types, this change in protein expression is underlain by an increase in c-IAP2 transcripts and dependent on the TLR3/TRIF pathway. When poly(I:C) is combined to the IAP inhibitor RMT 5265, a cooperative effect in apoptosis induction and/or inhibition of clonogenic growth is obtained in a large fraction of carcinoma and melanoma cell lines.</p> <p>Conclusions</p> <p>Currently, IAP inhibitors like RMT 5265 and poly(I:C) are the subject of separate therapeutic trials. In light of our observations, combined use of both types of compounds should be considered for treatment of human malignancies including carcinomas and melanomas.</p

    Testing gravity using galaxy-galaxy lensing and clustering amplitudes in KiDS-1000, BOSS and 2dFLenS

    The physics of gravity on cosmological scales affects both the rate of assembly of large-scale structure, and the gravitational lensing of background light through this cosmic web. By comparing the amplitude of these different observational signatures, we can construct tests that can distinguish general relativity from its potential modifications. We used the latest weak gravitational lensing dataset from the Kilo-Degree Survey, KiDS-1000, in conjunction with overlapping galaxy spectroscopic redshift surveys BOSS and 2dFLenS, to perform the most precise existing amplitude-ratio test. We measured the associated E_G statistic with 15-20% errors, in five dz = 0.1 tomographic redshift bins in the range 0.2 < z < 0.7, on projected scales up to 100 Mpc/h. The scale-independence and redshift-dependence of these measurements are consistent with the theoretical expectation of general relativity in a Universe with matter density Omega_m = 0.27 +/- 0.04. We demonstrate that our results are robust against different analysis choices, including schemes for correcting the effects of source photometric redshift errors, and compare the performance of angular and projected galaxy-galaxy lensing statistics.Comment: 23 pages, 14 figures, version accepted for publication by A&

    The IDENTIFY study: the investigation and detection of urological neoplasia in patients referred with suspected urinary tract cancer - a multicentre observational study

    Objective To evaluate the contemporary prevalence of urinary tract cancer (bladder cancer, upper tract urothelial cancer [UTUC] and renal cancer) in patients referred to secondary care with haematuria, adjusted for established patient risk markers and geographical variation. Patients and Methods This was an international multicentre prospective observational study. We included patients aged ≄16 years, referred to secondary care with suspected urinary tract cancer. Patients with a known or previous urological malignancy were excluded. We estimated the prevalence of bladder cancer, UTUC, renal cancer and prostate cancer; stratified by age, type of haematuria, sex, and smoking. We used a multivariable mixed-effects logistic regression to adjust cancer prevalence for age, type of haematuria, sex, smoking, hospitals, and countries. Results Of the 11 059 patients assessed for eligibility, 10 896 were included from 110 hospitals across 26 countries. The overall adjusted cancer prevalence (n = 2257) was 28.2% (95% confidence interval [CI] 22.3–34.1), bladder cancer (n = 1951) 24.7% (95% CI 19.1–30.2), UTUC (n = 128) 1.14% (95% CI 0.77–1.52), renal cancer (n = 107) 1.05% (95% CI 0.80–1.29), and prostate cancer (n = 124) 1.75% (95% CI 1.32–2.18). The odds ratios for patient risk markers in the model for all cancers were: age 1.04 (95% CI 1.03–1.05; P < 0.001), visible haematuria 3.47 (95% CI 2.90–4.15; P < 0.001), male sex 1.30 (95% CI 1.14–1.50; P < 0.001), and smoking 2.70 (95% CI 2.30–3.18; P < 0.001). Conclusions A better understanding of cancer prevalence across an international population is required to inform clinical guidelines. We are the first to report urinary tract cancer prevalence across an international population in patients referred to secondary care, adjusted for patient risk markers and geographical variation. Bladder cancer was the most prevalent disease. Visible haematuria was the strongest predictor for urinary tract cancer

    Introduction : L endomĂ©triose est dĂ©finie par la prĂ©sence, hors de l utĂ©rus, de tissu endomĂ©trial constituĂ© de glandes et de chorion cytogĂšne. Les mĂ©talloproteinases matricielles (MMPs) et leurs inhibiteurs endogĂšnes (TIMPs), les protĂ©ines de l apoptose, le rĂ©cepteur c-kit et des protĂ©ines de la famille Ă  rĂ©cepteurs Ă  l EGF ont un rĂŽle dans la progression tumorale mais leur rĂŽle dans l endomĂ©triose a Ă©tĂ© peu ou pas Ă©tudiĂ©. MatĂ©riel et mĂ©thodes : L objectif de ces travaux Ă©tait d Ă©tudier en immunohistochimie l expression de MMP-2, MMP-3, MMP-11, TIMP-1, TIMP-2, c-kit, des protĂ©ines de l apoptose, HER1 et HER2 dans l endomĂštre de patients atteintes ou non d endomĂ©triose et dans les lĂ©sions d endomĂ©triose pĂ©ritonĂ©ale, ovarienne et colo-rectale. HER2 a de plus Ă©tĂ© Ă©tudiĂ© par FISH dans les lĂ©sions d endomĂ©triose rectale et dans des prĂ©lĂšvements de carcinome endomĂ©trioĂŻde de l ovaire. De plus, des techniques de microdissection et de CGH (Hybridation GĂ©nomique Comparative) ont Ă©tĂ© effectuĂ©es pour rechercher des anomalies cytogĂ©nĂ©tiques dans les lĂ©sions d endomĂ©triose. RĂ©sultats : Dans l endomĂštre des patientes atteintes d endomĂ©triose, comparĂ© Ă  l endomĂštre de patientes non endomĂ©triosiques, une diminution de l expression de MMP-2 et une perte complĂšte de l expression de MMP-3 ont Ă©tĂ© observĂ©es. L expression de MMP-2, -3, -11, TIMP-1 et 2 Ă©tait la mĂȘme dans l endomĂštre de patientes endomĂ©triosiques et dans les lĂ©sions d endomĂ©triose pĂ©ritonĂ©ale. L expression de MMP-2, -3 et 11 Ă©tait plus Ă©levĂ©e dans les lĂ©sions colo-rectales que dans les lĂ©sions ovariennes et pĂ©ritonĂ©ales. Inversement, l expression de TIMP-2 Ă©tait plus faible dans les lĂ©sions colo-rectales et ovariennes que dans les lĂ©sions pĂ©ritonĂ©ales. L expression de p53 et p21 Ă©tait plus importante dans les endomĂ©triomes que dans l endomĂ©triose pĂ©ritonĂ©ale et l endomĂ©triose colorectale. L expression de bcl-2 Ă©tait plus faible dans les endomĂ©triomes que dans l endomĂ©triose pĂ©ritonĂ©ale et l endomĂ©triose colorectale. L'expression de c-kit Ă©tait plus Ă©levĂ©e dans les lĂ©sions colo-rectales que dans les lĂ©sions pĂ©ritonĂ©ales et ovariennes. Il n a pas Ă©tĂ© retrouvĂ© d expression d HER1 et 2 dans les diffĂ©rentes lĂ©sions d endomĂ©triose et il n y avait pas de surexpression d HER2 dans les lĂ©sions d endomĂ©triose colorectales et dans les carcinomes endomĂ©trioĂŻdes de l ovaire.
