Métodos de excavación en roca en obras subterráneas.
Tesis (Ingeniero Constructor)RESUMEN: Esta investigación bibliográfica de excavación en roca en obras subterráneas tiene como
objetivo general hacer un análisis técnico y económico de los dos métodos de
excavación en roca como son Perforación y Tronadura y Excavación mecánica de la
roca.
Aquí se expone de un modo general desde los estudios previos a la construcción de una
obra subterránea, pasando por conocimientos básicos de geología general hasta los
estudios geológicos que se necesitan en toda obra de construcción de túneles.
Además se encuentra lo necesario para ejecutar las faenas de excavación en roca como
equipos, procedimientos seguros de realizar las excavaciones, sostenimientos que se
utilizan en estas faenas, actividades de apoyo como son la ventilación al avance,
topografía utilizadas en túneles hasta llegar a los rendimientos y parámetros que se
utilizan para su predicción, finalizando con los costos de cada método.
Las conclusiones obtenidas en esta investigación bibliográfica pueden ser de interés
par, el lector porque logra saber en que condiciones se puede utilizar cada método y
c~l es más viable económicamente según las condiciones del proyecto.ABTRACT: This bibliographical investigation of excavation in rock in underground works has as
general objective to make a technical and economic analysis of the two excavation
methods in rock Iike they are Perforatíon and Explosion and mechanical Excavation of
the rock.
Here it is exposed in a general way from the previous studies to the construction of an
underground work, going by basic knowledge of general geology until the geologic
studies that are needed in all work of construction of tunnels.
Was also the necessary thing to execute the excavation tasks in rock Iike teams, sure
procedures of carrying out the excavations, maintenances that are used in these tasks,
support activities like they are the ventilation to the advance, topography used in tunnels
until arriving to the yields and parameters that are used for their prediction, concluding
with the costs of each method.
The conclusfons obtained in this bibliographical investigation can being of interest for the
reader because it is able to know in that condition you can use each method and which is
viable economically according to the conditions of the project

Caracterización de la proteína FNR de A. ferroxidans, estructural y funcionalmente.
Tesis (Doctor en Biociencias Moleculares)RESUMEN: En este estudio se demostró la capacidad de la cepa de A. ferrooxidans ATCC23270
de vivir en condiciones anaeróbicas de crecimiento utilizando S0 y Fe+3 como dador y
aceptor de electrones respectivamente.
Se identificó en el genoma de la bacteria un gen fnr que podría codificar para el
regulador global de la anaerobiosis, la proteína FNR (Fumarate Nitrate Reduction). Al
comparar la secuencia aminoacídica de FNRAF, con la del ortólogo mejor caracterizado, la
proteína FNR de E. coli, se observó grandes similitudes en relación a los dominios, motivos
y aminoácidos importantes para la función de esta clase de proteínas. Entre éstos últimos,
se identificó cuatro residuos de cisteína fundamentales para la unión de un centro [4Fe-
4S]+2 en condiciones anaeróbicas en la proteína. Al modelar estructuralmente FNRAF en
base a la estructura cristalina de un miembro relacionado de la familia, la proteína CRP
(Proteína activadora del catabolismo), se observó una distribución espacial similar de los
residuos de cisteína encargados de unir el centro [4Fe-4S]+2, al observado en la proteína
FNR de E. coli. Todos estos resultados sugieren fuertemente que FNRAF tiene una función
similar al de otras proteínas FNRs descritas en otras bacterias.
La proteína FNRAF purificada anaeróbicamente mostró distintas características
como color, absorbancia UV-Visible y cantidad de hierro que sugieren fuertemente la unión
de un centro [4Fe-4S]+2, el cual es un cofactor esencial para su función regulatoria. Una
característica interesante encontrada en FNRAF fue la menor sensibilidad a oxígeno que
presentó el centro [4Fe-4S]+2 en comparación a proteínas ortólogas, lo cual no ha sido
descrito anteriormente en otras bacterias y podría dar cuenta del funcionamiento de la
proteína tanto en condiciones aeróbicas como anaeróbicas de crecimiento, permitiendo de
este modo, una mejor transición de la bacteria entre ambos tipos de ambientes.
También se logro identificar en la bacteria distintos sitios de unión de FNRAF
ubicados en regiones promotoras de genes pertenecientes a distintas vías metabólicas tales
como transporte de membrana, stress, reacciones redox, regulación transcripcional y
metabolismo energético. En esta última categoría se identificó un complejo
multienzimático de proteínas de membrana denominado SreABCD, el cual ha sido asociado
en otros microorganismos a la reducción anaeróbica de azufre. Ensayos de RT-qPCR y
proteómica demostraron que tanto a nivel transcripcional como traduccional, los niveles de
expresión de sreABCD se vieron incrementados en fase estacionaria del crecimiento
anaeróbico. Análisis de co-transcripción mediante RT-PCR demostraron que sreABCD se
expresó como una sola unidad transcripcional constituyendo por ende un operón. La
ubicación de este sitio de unión a FNR es característico de promotores tipo II (sitio de
unión de FNR ubicado rio arriba de caja -35 de unión a la RNA polimerasa), los cuales...ABSTRACT: This study investigated molecular genetic aspects of the growth of the
chemolithoautotrophic acidophile Acidithiobacillus ferrooxidans ATCC 23270 in anaerobic
growth conditions using S0 and Fe+3 as electron donor and acceptor respectively.
A gene denominated fnrAF was identified in the genome of A. ferrooxidans that is
predicted to encode the global regulator FNR (Fumarate Nitrate Reductase). A comparison
of the amino acid sequence of FNRAF with the FNR of Escherichia coli, revealed conserved
domains, motifs and amino acids important for the function of this class of proteins
including four critically positioned cysteine residues predicted to be involved in the binding
of a [4Fe-4S]+2 center under anaerobic conditions. Molecular modeling of the three
dimensional structure of FNRAF using the structurally related CRP (Catabolite Activator
Protein) crystal structure as a template demonstrated significant conservation of the spatial
distribution of the critical cysteine residues. These results strongly suggest that FNRAF has
a similar function to other reported FNRs.
FNRAF, purified under anaerobic conditions, exhibited properties such as color, UVvisible
absorbance and amount of iron that are characteristic of an [4Fe-4S]+2 center, an
essential cofactor for the regulatory function of FNRAF. An interesting feature of FNRAF
was the diminished sensibility of the [4Fe-4S]+2 center to oxygen compared with
orthologous FNRs. This has not previously been described in other bacteria and could
provide a mechanism that permits A. ferrooxidans to live aerobically or anaerobically and
to transition between the two states.
In addition, FNR binding sites were predicted using bioinformatic tools in promoter
regions of genes belonging to a number of metabolic pathways such membrane transport
functions, stress and redox reactions, transcriptional regulation and energy metabolism.
Among the latter is the SreABCD complex involved in the anaerobic reduction of sulfur in
other bacteria. RT-qPCR and proteomic analyses demonstrated that the transcription of the
sreABCD gene cluster and the levels of SreABCD encoded proteins were increased during
stationary phase under anaerobic conditions. PCR analysis demonstrated that the sreABCD
gene cluster was expressed as a single transcriptional unit and is therefore an operon. An
FNR DNA binding site was identified upstream of the promoter of the operon using HMM
and PSSM bioinformatics tools for pattern-matching. The promoter exhibited sequence
characteristics of type II promoters responsible for activation of downstream genes.
Activation of SreABCD by FNRAF was experimentally validated through in vivo assays
with a reporter gene in a heterologous host (E. coli).
Combining the above results allowed a systems biology approach to be developed
that predicted a network of interacting genes and pathways used in anaerobic metabolism
under the control of the master regulator FNR. This network predicts genetic connections
not previously described in other bacteria for regulators of the FNR class such as the
connections between hupSL, involved in oxidation of molecular hydrogen coupled to the
reduction of sulfur (sreABCD) and cysteine formation (cysJIHDNG); the latter is involved..
Prediction of abnormal wine fermentations using computational intelligent techniques
The early detection abnormal fermentations (sluggish and stuck) is one of the main problems that appear in wine production, due to the signi cant impacts in wine quality and utility. This situation is specially important in Chile, which is one of the top ten worldwide wine production countries. In last years, two di erent methods coming from Computational Intelligence have been applied to solve this problem: Arti cial Neural Networks and Support Vector Machines. In this work we present the main results that have been obtained to detect abnormal wine fermentations applying these approaches. The Support Vector Machine method with radial basis kernel present the best results for the time cuto s considered (72 [hr] and 96 [hr]) over all the techniques studied with respect to prediction rates and number of the training sets.Facultad de Informátic
Microalgae and Cyanobacteria as Green Molecular Factories: Tools and Perspectives
Cyanobacteria and eukaryotic microalgae are phototrophic microorganisms capable of producing organic compounds using solar energy. Owing to their fast growth, low cost cultivation, and the diversity of high-value chemical substances produced, they are considered an attractive target to be exploited by the biotechnology industry. While genetic modulation of these organisms has been extensively proved in the laboratory, present-day microalgal industry uses mainly non-transgenic strains. Although some unicellular cyanobacteria can be successfully engineered, many commercial bioproducts are synthesized preferably by eukaryotic microalgae or filamentous cyanobacteria to take advantage of their better-suited natural physiological characteristics. The successful genetic engineering of these microorganisms is not limited to the understanding of the gene expression machinery (e.g., promoters, codon usage, ribosome binding sites), but it must also include other subjects, such as defense mechanisms against the intrusion of foreign DNA. This chapter reviews current strategies in microalgae biotechnology and analyzes the most frequent problems we faced to genetically manipulate these microorganisms, including their transformation and selection methodologies. In summary, we attempt to provide a comprehensive review of the relevant information and tools required for optimal engineering of a photosynthetic microorganism employed in sustainable biotechnology applications
Marioneta y de-formación tensiva en Suspensions (1976 - 1980) de Sterlac: en torno al individuo y subjetividad en el cuerpo
Este artículo propone una aproximación al trabajo performático de Stelarc (1976-1980), llamado Suspensions, el cual aborda la problematización de la “auto-afección” de la experiencia estética en relación con la configuración espacial del cuerpo, cuya implicancia es la intervención del orden simbólico y gravitacional de éste a través de las tensiones de zonas de la piel. Aquella intervención plantea la cuestión sobre la subjetivación del cuerpo –o su interiorización inmanente– por la auto-afección y la exteriorización de la noción de individuo, en relación con los conceptos tiempo y espacio para la interioridad del ‘sí mismo’, asociados a la subjetividad de la estética moderna. Así, Stelarc inicia en Suspensions su trayectoria hacia la fortaleza de la individuación
