    Aspectos citoquímicos das células do sangue periférico de Oreochromis (Tilapia) niloticus. (Linnaeus, 1758) (Cichlidae, Teleostei): parte II

    Morphologicaly, seven types of cells were identified in the blood of Oreochromis niloticus: erythrocytes, thrombocytes, neutrophils, eosinophils, basophils, lymphocytes and monocytes. Glycogen was present not only in the cytoplasm of neutrophils and thrombocytes but also in some lymphocytes and monocytes. The positive reaction for myeloperoxidase and Sudan black was observed in neutrophils and eosinophils. The bromphenol blue method was strongly positive for erythrocytes and eosinophils.Morfologicamente foram identificados no sangue de Oreochromis niloticus sete tipos de células: eritrócitos, trombócitos, neutrófilos, eosinófilos, basófilos, linfócitos e monócitos. Em relação aos resultados citoquímicos foi contastada a presença de glicogênio em neutrófilos, trombócitos e em alguns linfócitos e monócitos. Os grânulos citoplasmáticos de neutrófilos e eosinófilos mostraram positividade para mieloperoxidade e Sudan black.     Highly efficient polarized polymer light-emitting diodes utilizing oriented films of beta-phase poly(9,9-dioctylfluorene)

    Uniaxially oriented films of beta-phase poly(9,9-dioctylfluorene) (PFO) were realized by a friction-transfer technique followed by thermal annealing and vapor treatments. Absorption and photoluminescence (PL) spectra show the characteristics of beta-phase: an additional absorption peak at 433 nm and redshifted PL peaks compared with those of the usual nematic (N) phase. We fabricated polarized polymer light-emitting diodes utilizing oriented films of beta-phase PFO. Highly polarized beta-phase emission with an integrated polarization ratio of 51 was observed from the devices. Molecular Pharmaceutics 20, 419 (2022); https://doi.org/10.1021/acs.molpharmaceut.2c00698.The contribution ratio of metabolic enzymes such as cytochrome P450 to in vivo clearance (fraction metabolized: fm) is a pharmacokinetic index that is particularly important for the quantitative evaluation of drug-drug interactions. Since obtaining experimental in vivo fm values is challenging, those derived from in vitro experiments have often been used alternatively. This study aimed to explore the possibility of constructing machine learning models for predicting in vivo fm using chemical structure information alone. We collected in vivo fm values and chemical structures of 319 compounds from a public database with careful manual curation and constructed predictive models using several machine learning methods. The results showed that in vivo fm values can be obtained from structural information alone with a performance comparable to that based on in vitro experimental values and that the prediction accuracy for the compounds involved in CYP induction or inhibition is significantly higher than that by using in vitro values. Our new approach to predicting in vivo fm values in the early stages of drug discovery should help improve the efficiency of the drug optimization process

    Hematological studies in Oreochromis niloticus (Linnaeus, 1758) (Cichlidae, Teleostei) - Part I

    Hemogram and cell morphology of peripheric blood samples obtained from 10 Oreochromis niloticus fishes have been evaluated. Some of these samples have been used in the accomplishment of hematimetry: eritrocytes count (x = 2.35 x 106/mm3 ±0.12), hemoblobin rate ( x = 7.04 g/dL ± 0.38), hematocrit (x = 27.85% ± 1.62), MHC (x = 30.31 pg ± 1.64), MCV (x = 118.6 p3 ± 3.53), MCHC (x = 25.78% ± 1.72), total leucocytes count (x = 16.08 x 103/mm3 ± 1.28), trombocytes count ( x = 61.69 x 103/mm3 ± 5.96). Morphological evaluation of blood cells was performed by means blood smears extension submitted to Wright staining. Both nucleated eritrocytes and trombocytes presented an elliptical shape; neutrophiles and eosinophiles were spherical with spherical nucleus presenting different sizes, they also showed profuse citoplasms which were slightly basophilic and highly acidophilous, respectively. Polimorphonuclear shapes in these cells have not been verified. Basophiles have shown spherical nucleus and metachromic granulations. Monocytes as well as lymphocytes resembled morphologically those of mammals.Avaliou-se o hemograma e a morfologia celular em amostras de sangue periférico obtidas de 10 peixes da espécie Oreochromis niloticus. Os monócitos e os linfócitos assemelharam-se morfologicamente aos dos mamíferos

    Effects of Microgravity on Cerebral Hemodynamics

    After centuries of wonder, human being finally realized the decisive possibility of going to the moon a little more than 50 years ago, and in 1969 this became a reality, no longer a dream. The human body, however, is designed to live under 1 g as on Earth. Thus, it is important to understand what happens in the human body under microgravity in order to make rapid progress in space development. One of the most serious problems produced by microgravity is a fluid shift from lower to upper body. This cephalad fluid shift may change the hemodynamics in the brain. In this review, effects of actual or simulated microgravity on cerebral blood flow, intracranial pressure, formation of brain edema, and orthostatic intolerance will be discussed. These studies provide us lots of information not only to maintain the health of astronauts but also to treat senile persons who, on rare occasions, sit or stand up

    Behavioral factors associated with SARS-CoV-2 infection in Japan.

    BACKGROUND: The relative burden of COVID-19 has been less severe in Japan. One reason for this may be the uniquely strict restrictions imposed upon bars/restaurants. To assess if this approach was appropriately targeting high-risk individuals, we examined behavioral factors associated with SARS-CoV-2 infection in the community. METHODS: This multicenter case-control study involved individuals receiving SARS-CoV-2 testing in June-August 2021. Behavioral exposures in the past 2 weeks were collected via questionnaire. SARS-CoV-2 PCR-positive individuals were cases, while PCR-negative individuals were controls. RESULTS: The analysis included 778 individuals (266 [34.2%] positives; median age [interquartile range] 33 [27-43] years). Attending three or more social gatherings was associated with SARS-CoV-2 infection (adjusted odds ratio [aOR] 2.00 [95% CI 1.31-3.05]). Attending gatherings with alcohol (aOR 2.29 [1.53-3.42]), at bars/restaurants (aOR 1.55 [1.04-2.30]), outdoors/at parks (aOR 2.87 [1.01-8.13]), at night (aOR 2.07 [1.40-3.04]), five or more people (aOR 1.81 [1.00-3.30]), 2 hours or longer (aOR 1.76 [1.14-2.71]), not wearing a mask during gatherings (aOR 4.18 [2.29-7.64]), and cloth mask use (aOR 1.77 [1.11-2.83]) were associated with infection. Going to karaoke (aOR 2.53 [1.25-5.09]) and to a gym (aOR 1.87 [1.11-3.16]) were also associated with infection. Factors not associated with infection included visiting a cafe with others, ordering takeout, using food delivery services, eating out by oneself, and work/school/travel-related exposures including teleworking. CONCLUSIONS: We identified multiple behavioral factors associated with SARS-CoV-2 infection, many of which were in line with the policy/risk communication implemented in Japan.     Activation of Sympathetic Signaling in Macrophages Blocks Systemic Inflammation and Protects against Renal Ischemia-Reperfusion Injury

    Background: The sympathetic nervous system regulates immune cell dynamics. However, the detailed role of sympathetic signaling in inflammatory diseases is still unclear because it varies according to the disease situation and responsible cell types. This study focused on identifying the functions of sympathetic signaling in macrophages in LPS-induced sepsis and renal ischemia-reperfusion injury (IRI).Methods: We performed RNA sequencing of mouse macrophage cell lines to identify the critical gene that mediates the anti-inflammatory effect of β2-adrenergic receptor (Adrb2) signaling. We also examined the effects of salbutamol (a selective Adrb2 agonist) in LPS-induced systemic inflammation and renal IRI. Macrophage-specific Adrb2 conditional knockout (cKO) mice and the adoptive transfer of salbutamol-treated macrophages were used to assess the involvement of macrophage Adrb2 signaling.Results: In vitro, activation of Adrb2 signaling in macrophages induced the expression of T cell Ig and mucin domain 3 (Tim3), which contributes to anti-inflammatory phenotypic alterations. In vivo, salbutamol administration blocked LPS-induced systemic inflammation and protected against renal IRI; this protection was mitigated in macrophage-specific Adrb2 cKO mice. The adoptive transfer of salbutamol-treated macrophages also protected against renal IRI. Single-cell RNA sequencing revealed that this protection was associated with the accumulation of Tim3-expressing macrophages in the renal tissue.Conclusions: The activation of Adrb2 signaling in macrophages induces anti-inflammatory phenotypic alterations partially via the induction of Tim3 expression, which blocks LPS-induced systemic inflammation and protects against renal IRI

    島根県西部地区出身看護学生の就職先選択要因に関する調査

    島根県西部地区出身の看護学生の就職先選択要因について明らかにすることを目的とし、過去5年間に島根県立大学短期大学部看護学科を卒業した西部地区就職者4名、東部地区就職者9名の合計13名にインタビュー調査を行った。就職先を選択した要因、西部地区の病院に就職しなかった要因、就職先を選択して良かったこと、西部地区への就職率増加につながると思うこと、島根県立大学に求める支援の5つの視点に沿って分析した。その結果から、病院との連携をさらに図りながら現在のキャリア支援を継続していくとともに、卒業後のフォローアップ体制についてもさらに検討していく必要があると考える
