    Kinetic Vlasov Simulations of collisionless magnetic Reconnection

    A fully kinetic Vlasov simulation of the Geospace Environment Modeling (GEM) Magnetic Reconnection Challenge is presented. Good agreement is found with previous kinetic simulations using particle in cell (PIC) codes, confirming both the PIC and the Vlasov code. In the latter the complete distribution functions fkf_k (k=i,ek=i,e) are discretised on a numerical grid in phase space. In contrast to PIC simulations, the Vlasov code does not suffer from numerical noise and allows a more detailed investigation of the distribution functions. The role of the different contributions of Ohm's law are compared by calculating each of the terms from the moments of the fkf_k. The important role of the off--diagonal elements of the electron pressure tensor could be confirmed. The inductive electric field at the X--Line is found to be dominated by the non--gyrotropic electron pressure, while the bulk electron inertia is of minor importance. Detailed analysis of the electron distribution function within the diffusion region reveals the kinetic origin of the non--gyrotropic terms

    Serotonin reduces inhibition via 5-HT1A receptors in area CA1 of rat hippocampal slices in vitro

    We studied the effects of serotonin (5-HT) on intrinsic and synaptic responses of hippocampal CA1 cells. The effects were partially mimicked by the 5-HT1A receptor agonist, 8-OH-DPAT, and prevented by the 5-HT1A receptor antagonist, NAN-190. Polysynaptic fast and slow inhibitory postsynaptic potentials (IPSPs) were reduced in amplitude by 60-70% following application of both 5-HT and 8-OH-DPAT. Monosynaptic fast IPSPs were reduced by 60% and slow IPSPs by 90% following application of both drugs. Since there is a temporal overlap of fast and slow IPSPs, the reduction in fast IPSPs could have arisen indirectly from the larger effect of 5-HT on slow IPSPs. To overcome this problem we blocked the slow IPSPs with new, potent GABA-B antagonists, but still observed a similar reduction in the fast IPSP with 5-HT and 8-OH-DPAT. However, the reductions in the fast IPSPs could also have arisen from the 5-HT-induced total conductance increases. Using single-electrode voltage clamp and intracellular K+ channel blockers we still observed similar changes. 5-HT and 8-OH-DPAT had no effect upon GABA-A-mediated currents evoked by iontophoretic GABA application to the dendrites or the soma of CA1 pyramidal cells, Putative inhibitory internuerons were hyperpolarized by 5-HT and their evoked EPSPs strongly reduced by 5-HT and 8-OH-DPAT. Our data indicate that 5-HT modulates fast and slow synaptic inhibition of principal cells using presynaptic mechanisms involving the inhibition of inhibitory interneurons

    Laser-induced breakdown spectrometry (LIBS) has been applied for multi-elemental analysis of slag samples from a steel plant. In order to avoid the time-consuming step of sample preparation, the liquid slag material can be filled in special probes. After cooling of the liquid slag and solidification, the samples can be analyzed with LIBS - Chemical analysis of slag is an essential input parameter used for numerical simulations to control liquid steel processing. The relative variation range of element concentrations in slag samples from steel production can amount to up to 30%. A multivariate calibration model is used to take into account matrix effects caused by these varying concentrations. By optimizing the measuring parameters as well as the calibration models, an agreement between the standard X-ray fluorescence (XRF) analysis and LIBS analysis in terms of the coefficient of determination r2 of 0.99 for the main analytes CaO, SiO2, and Fe(tot) of converter slag samples was achieved. The average repeatability of the LIBS measurement for these elements in terms of the relative standard deviation of the determined concentration is improved to less than 1.0%. With these results, the basis is established for future on-line applications of LIBS in the steel-making industry for slag analysis

    Evaluating uniform manifold approximation and projection for dimension reduction and visualization of polinsar features

    In this paper, the nonlinear dimension reduction algorithm Uniform Manifold Approximation and Projection (UMAP) is investigated to visualize information contained in high dimensional feature representations of Polarimetric Interferometric Synthetic Aperture Radar (PolInSAR) data. Based on polarimetric parameters, target decomposition methods and interferometric coherences a wide range of features is extracted that spans the high dimensional feature space. UMAP is applied to determine a representation of the data in 2D and 3D euclidean space, preserving local and global structures of the data and still suited for classification. The performance of UMAP in terms of generating expressive visualizations is evaluated on PolInSAR data acquired by the F-SAR sensor and compared to that of Principal Component Analysis (PCA), Laplacian Eigenmaps (LE) and t-distributed Stochastic Neighbor embedding (t-SNE). For this purpose, a visual analysis of 2D embeddings is performed. In addition, a quantitative analysis is provided for evaluating the preservation of information in low dimensional representations with respect to separability of different land cover classes. The results show that UMAP exceeds the capability of PCA and LE in these regards and is competitive with t-SNE

    Molecular Dynamics Simulations Reveal an Interplay between SHAPE Reagent Binding and RNA Flexibility

    The function of RNA molecules usually depends on their overall fold and on the presence of specific structural motifs. Chemical probing methods are routinely used in combination with nearest-neighbor models to determine RNA secondary structure. Among the available methods, SHAPE is relevant due to its capability to probe all RNA nucleotides and the possibility to be used in vivo. However, the structural determinants for SHAPE reactivity and its mechanism of reaction are still unclear. Here molecular dynamics simulations and enhanced sampling techniques are used to predict the accessibility of nucleotide analogs and larger RNA structural motifs to SHAPE reagents. We show that local RNA reconformations are crucial in allowing reagents to reach the 2'-OH group of a particular nucleotide and that sugar pucker is a major structural factor influencing SHAPE reactivity

    Anti-phospholipid-antibodies in patients with relapsing polychondritis

    Relapsing polychondritis (RP) is an extremly rare multisystemic disease thought to be of autoimmune origin. In order to assess if RP is associated with anti-phospholipid antibodies (aPL), clinical data and sera of 21 patients with RP were collected in a multicentre study. Concentration of anti-cardiolipin antibodies (aCL) (IgG-, IgM-and IgA-isotypes), anti-phosphatidylserine-antibodies (aPS) (IgG-and IgM-isotypes) and anti-β-2-glycoprotein I-antibodies (aβ2 GPI) were measured by ELISA. In eight patients aCL were found to be elevated. One patient had elevated aPS. No patient had elevated aβ2 GPI. No patient had clinical signs and symptoms of a aPL syndrome. Interestingly, the two RP patients with the highest aPL had concomitant systemic lupus erythematosus (SLE). Therefore the presence of elevated aPL in RP is probably more closely related to an associated SLE than to RP itself. There is no convincing evidence that aPL are associated with RP

    The effect of a regional increase in ocean surface roughness on the tropospheric circulation: a GCM experiment

    The sensitivity of the atmospheric circulation to an increase in ocean surface roughness in the Southern Hemisphere storm track is investigated in a paired general circulation model experiment. Such a change in sea roughness could be induced by ocean waves generated by storms. Two extended permanent-July runs are made. One with standard sea surface roughness, the other with ten times as a large surface roughness over open sea poleward of 40-degrees-S. The regional increase in ocean surface roughness significantly modifies the tropospheric circulation in the Southern Hemisphere. The strongest effect is the reduction of tropospheric winds (by 2 m/s or 100%) above the area with increased roughness. The poleward eddy momentum flux is reduced in the upper troposphere and the meridional eddy sensible heat flux is reduced in the lower troposphere. Zonal mean and eddy kinetic energy are consistently reduced

    INI1 mutations in meningiomas at a potential hotspot in exon 9

    Rhabdoid tumours have been shown to carry somatic mutations in the INI1 (SMARCB1/hSNF5) gene. A considerable fraction of these tumours exhibit allelic losses on chromosome 22. Allelic loss on 22q also is characteristic for meningiomas, however most of these alterations are considered to be associated with mutations of the NF2 gene. We examined a series of 126 meningiomas for alterations in the INI1 gene. Four identical somatic mutations in exon 9 were detected resulting in an exchange of Arg to His in position 377 of INI1. Our observations were reproduced both by using DNA from a new round of extraction and by employing overlapping primers. This mutational hotspot therefore appears to be an important target in the formation of a fraction of meningiomas. In addition, 4 novel polymorphisms of INI1 were characterized. Our data indicate that the INI1 is a second tumour suppressor gene on chromosome 22 that may be important for the genesis of meningiomas. © 2001 Cancer Research Campaign http://www.bjcancer.co
