    Research in plant pathology often relies on testing interactions between a fungicide and a pathogen in vitro and extrapolating from these results what may happen in planta. Likewise, results from glasshouse experiments are used to estimate what will happen if the fungicide is applied in the field. However, it is difficult to obtain conditions in vitro and in the glasshouse which reflect the conditions where the fungicide may eventually be used, in the field.     Exploring Clinical Educator and Student Perspectives on the Implementation of the Calderdale Framework as a Model of Occupational Therapy Clinical Education in Australia: a Pilot Study

    Purpose: This paper describes the exploration of clinical educator and student perspectives on the implementation of the Calderdale Framework (CF) as a model for pre-entry occupational therapy clinical placement. Setting: The study was undertaken within a regional health service in Queensland, Australia. The orthopaedic inpatient ward at the regional hospital and mixed inpatient and outpatient community based caseloads in rural hospitals were the focus of this study. Methods: The Calderdale Framework is a seven stage process which involves: engaging staff with the framework; identifying which clinical tasks are carried out within the service; analysing which tasks can be allocated to students; setting up supervision systems; developing Clinical Task Instructions (CTIs); providing competency training and assessment; and developing organisational systems to sustain the new ways of working. In the first week of placement, students were ‘taught’ background and theoretical information for nine Clinical Task Instructions. In the remainder of their placement they achieved competence in these tasks and implemented clinical interventions with patients. Results: Separate focus groups and in-depth interviews were used to explore clinical educator and student opinions about their experience of the application of the Calderdale Framework in clinical education. Five themes emerged from the data analysis: experience of new model; adjustment to change in approach to the placement model; impact of the Calderdale Framework Clinical Task Instructions; impact of the Calderdale Framework learning model on student confidence and impact of the Calderdale Framework placement model on service delivery. Conclusion: In this pilot study, the application of the Calderdale Framework in clinical education was supported by both clinical educators and students as a model that supported student learning and safe contribution to patient service delivery. The time invested in planning, resource development and implementing this application of the Calderdale Framework in clinical education has resulted in a model which can continue to be implemented within our health service and could be applied to other professions or replicated elsewhere

    Effects and mechanisms of an allied health research position in a Queensland regional and rural health service: a descriptive case study

    The aim of the present case study is to illustrate the outcomes of a dedicated allied health (AH) research position within a large Queensland regional and rural health service. The secondary aim of the case study is to describe the enabling and hindering mechanisms to the success of the role. Semistructured interviews were conducted with the Executive Director of Allied Health and the current AH research fellow incumbent within the health service. A focus group was also undertaken with six stakeholders (e.g. clinicians, team leaders) who had engaged with the research position. Outcomes of the AH research fellow included clinical and service improvements, enhanced research culture and staff up-skilling, development of research infrastructure and the formation of strategic research collaborations. Despite being a sole position in a geographically expansive health service with constrained resources, key enabling mechanisms to the success of the role were identified, including strong advocacy and regular communication with the Executive. In conclusion, the case study highlights the potential value of an AH research position in building research capacity within a large non-metropolitan health service. Factors to facilitate ongoing success could include additional research and administrative funding, as well as increased use of technology and team-based research. What is known about the topic?: Dedicated research positions embedded within health care settings are a well cited strategy to increase research capacity building of allied health professionals (AHPs). However the majority of these positions are within metropolitan health settings and unique challenges exist for these roles in regional and rural areas. Few studies have described the impact of dedicated AH research positions within regional health centres or the factors which facilitate or hinder their role. What does this paper add?: Dedicated research positions within a non-metropolitan Australian health service may have a positive impact on AH clinical services, research culture, staff upskilling, research infrastructure and research collaborations. Key enabling mechanisms to support the role may include advocacy from higher level management, strong networks and communication channels. Additional research and administrative funding, the use of technology and team based research may enhance sustainability of such roles. What are the implications for practitioners?: AH research positions have potential value in building research capacity within a large non-metropolitan health service. Health managers and researchers should be aware of the unique challenges to these roles and consider mechanisms that may best enhance and sustain outcomes of the positions including: the development of infrastructure (i.e. technology, website of resources), networks, and communication strategies (i.e. regular meetings with leadership and promotion internally)

    Synthesis and characterisation of hop derived compounds, and their application in quantitative high performance liquid chromatography

    High Performance Liquid Chromatography (HPLC) has been extensively used in the analysis of hop derived components in beer, but the scope of these methods is limited by the range of attainable reference compounds. The synthesis and purification of a range of relevant components has been described by several authors, but in our laboratory these procedures fail to give compounds of sufficient purity. Modification of published procedures are described for the preparation of humulinic acids, dehydrohumulinic acids, hulupones, and colupulone. These procedures provide facile routes for preparation of these compounds in higher yields than was hitherto reported, and also in sufficient purity for their use as standards in quantitative HPLC analysis. Although bitterness in beer is largely due to iso - a - acids, other compounds such as hulupones are considered to have similar bitter properties. Alternative routes to hulupones using the readily available and cheap 6-acids extracts as starting material are therefore examined. Oxidation of Bacids in the presence of sodium sulphite was established as the most feasible route to these compounds. The structural authenticity of all preparations was confirmed by infra-red, and nuclear magnetic resonance analysis. Purity of the compounds was established using HPLC in conjunction with photodiode array detection. Developed HPLC methods, and synthesised pure reference standards are used in the quantitative analysis of humulinic acids in hop iso-a-acid extracts before, and after acid hydrolysis. HPLC methods are also developed for the quantitative analysis of hulupone formed from the oxidation of B-acids solutions. The stability of all compounds synthesised was investigated so that satisfactory conditions for their long-term storage could be identified

    Direct Interaction of Pericentrin with Cytoplasmic Dynein Light Intermediate Chain Contributes to Mitotic Spindle Organization

    Pericentrin is a conserved protein of the centrosome involved in microtubule organization. To better understand pericentrin function, we overexpressed the protein in somatic cells and assayed for changes in the composition and function of mitotic spindles and spindle poles. Spindles in pericentrin-overexpressing cells were disorganized and mispositioned, and chromosomes were misaligned and missegregated during cell division, giving rise to aneuploid cells. We unexpectedly found that levels of the molecular motor cytoplasmic dynein were dramatically reduced at spindle poles. Cytoplasmic dynein was diminished at kinetochores also, and the dynein-mediated organization of the Golgi complex was disrupted. Dynein coimmunoprecipitated with overexpressed pericentrin, suggesting that the motor was sequestered in the cytoplasm and was prevented from associating with its cellular targets. Immunoprecipitation of endogenous pericentrin also pulled down cytoplasmic dynein in untransfected cells. To define the basis for this interaction, pericentrin was coexpressed with cytoplasmic dynein heavy (DHCs), intermediate (DICs), and light intermediate (LICs) chains, and the dynamitin and p150Glued subunits of dynactin. Only the LICs coimmunoprecipitated with pericentrin. These results provide the first physiological role for LIC, and they suggest that a pericentrin–dynein interaction in vivo contributes to the assembly, organization, and function of centrosomes and mitotic spindles

    Turbulence Nonlinearities Shed Light on Geometric Asymmetry in Tokamak Confinement Transitions

    A comprehensive study of fully frequency-resolved nonlinear kinetic energy transfer has been performed for the first time in a diverted tokamak, providing new insight into the parametric dependences of edge turbulence transitions. Measurements using gas puff imaging in the turbulent L-mode state illuminate the source of the long known but as yet unexplained "favorable-unfavorable" geometric asymmetry of the power threshold for transition to the turbulence-suppressed H mode. Results from the recently discovered I mode point to a competition between zonal flow (ZF) and geodesic-acoustic modes (GAM) for turbulent energy, while showing new evidence that the I-to-H transition is still dominated by ZFs. The availability of nonlinear drive for the GAM against net heat flux through the edge corresponds very well to empirical scalings found experimentally for accessing the I mode
