    Energy implications of household rainwater systems

    The energy intensities of a range of household rainwater systems were monitored in Sydney and Newcastle as part of a study carried out by the Institute for Sustainable Futures (ISF) in collaboration with the Australian Commonwealth Scientific Investigation and Research Organisation (CSIRO). The study found that the energy intensity of these rainwater systems varied depending on system configuration, rainwater end uses and the water efficiency of the household. Preliminary results indicate that the energy intensity varies between 0.9 and4.9 kWh/kL with a 'typical' household rainwater system using approximately 1.5 kWh to deliver each kilo litre of rainwater

    Targeted Ī²-Phase Formation in Poly(fluorene)-Ureasil Grafted Organic-Inorganic Hybrids

The development of synthetic strategies to control the molecular organization (and inherently linked optoelectronic properties) of conjugated polymers is critical for the development of efficient light-emitting devices. Here, we report a facile route using sol-gel chemistry to promote the formation of the Ī²-phase through the covalent-grafting of poly[(9,9-dioctylfluorene)-co-(9,9-bis(8-hydroxyoctyl)fluorene)] (PFO-OH) to poly(oxyalkylene)/siloxane hybrids known as ureasils, due to the urea linkages binding the organic and inorganic components. Although grafting occurs within the siliceous domains, the degree of branching of the organic backbone determines the packing of the PFO-OH chains within the ureasil framework. Moreover, photoluminescence studies indicate that physical confinement also plays a key role in promoting the evolution of the Ī²-phase of PFO-OH as the sol-gel transition proceeds. Spectroscopic and structural analyses reveal that dibranched ureasils promote linear packing of the PFO-OH chains, while tribranched ureasils exhibit a more open, distorted structure that restricts the packing efficacy and reduces the number of covalent anchorages. These results indicate that the organic-inorganic hybrid structure induces distinct levels of Ī²-phase formation and that covalent grafting is a versatile approach to design novel poly(fluorene) hybrid materials with tailored optical properties

    Haplobanking induced pluripotent stem cells for clinical use

    The development of induced pluripotent stem cells (iPSCs) by Shinya Yamanaka and colleagues in 2006 has led to a potential new paradigm in cellular therapeutics, including the possibility of producing patient-specific, disease-specific and immune matched allogeneic cell therapies. One can envisage two routes to immunologically compatible iPSC therapies: using genetic modification to generate a ā€˜universal donorā€™ with reduced expression of Human Leukocyte Antigens (HLA) and other immunological targets or developing a haplobank containing iPSC lines specifically selected to provide HLA matched products to large portions of the population. HLA matched lines can be stored in a designated physical or virtual global bank termed a ā€˜haplobankā€™. The process of ā€˜iPSC haplobankingā€™ refers to the banking of iPSC cell lines, selected to be homozygous for different HLA haplotypes, from which therapeutic products can be derived and matched immunologically to patient populations. By matching iPSC and derived products to a patientā€™s HLA class I and II molecules, one would hope to significantly reduce the risk of immune rejection and the use of immunosuppressive medication. Immunosuppressive drugs are used in several conditions (including autoimmune disease and in transplantation procedures) to reduce rejection of infused cells, or transplanted tissue and organs, due to major and minor histocompatibility differences between donor and recipient. Such regimens can lead to immune compromise and pathological consequences such as opportunistic infections or malignancies due to decreased cancer immune surveillance.     Improved timed-mating, non-invasive method using fewer unproven female rats with pregnancy validation via early body mass increases

    For studies requiring accurate conception-timing, reliable, efficient methods of detecting oestrus reduce time and costs, whilst improving welfare. Standard methods use vaginal cytology to stage cycle, and breeders are pairedā€“up using approximately five proven females with proven males to achieve at least one conception on a specific day. We describe an alternative, fast, consistent, non-invasive method of timed-mating using detection of lordosis behaviour in Wistar and Lister-Hooded rats that used unproven females with high success rates. Rats under reverse-lighting had body masses recorded pre-mating, day (d) 3-4, d8, d10 and d18 of pregnancy. Using only the presence of the oestrus dance to time-mate females for 24-hrs, 89% Wistar and 88% Lister-Hooded rats successfully conceived. We did not observe behavioural oestrus in Sprague-Dawleys without males present. Significant body mass increases following mating distinguished pregnant from non-pregnant rats, as early as d4 of pregnancy (10% Ā± 1.0 increase cf 3% Ā± 1.2). The pattern of increases throughout gestation was similar for all pregnant rats until late pregnancy, when there were smaller increases for primi- and multiparous rats (32% Ā± 2.5; 25% Ā± 2.4), whereas nulliparous rats had highest gains (38% Ā± 1.5). This method demonstrated a distinct refinement of the previous timed-mating common practice used, as disturbance of females was minimised. Only the number required of nulli-, primi- or multiparous rats were mated, and body mass increases validated pregnancy status. This new breeding-management method is now established practice for two strains of rat and resulted in a reduction in animal use

    Evaluation of demand in a rural English hospital emergency department

    The purpose of this article is to analyse the patient demand placed on a rural district general hospital (DGH) emergency department within the context of the Purpose, Process, People (PPP) framework used in the private sector. This analysis was undertaken to inform wider evaluation of the implementation of the enterprise cultureā€”the NHS policy to adopt private sector best practice to produce resource use, quality and efficiency improvements. The article concludes with a view that the PPP framework provides methods of calculating the level of discharge necessary to meet the four-hour wait target. Data describing the characteristics and patterns of attending patients can be used to develop an emergency departmentā€™s processes and people to achieve its time-based target

    Constructing and testing hypotheses of dinosaur foot motions from fossil tracks using digitization and simulation

    Whilst bones present a static view of extinct animals, fossil footprints are a direct record of the activity and motion of the track maker. Deep footprints are a particularly good record of foot motion. Such footprints rarely look like the feet that made them; the sediment being heavily disturbed by the foot motion. Because of this, such tracks are often overlooked or dismissed in preference for more footā€like impressions. However, the deeper the foot penetrates the substrate, the more motion is captured in the sediment volume. We have used deep, penetrative, Jurassic dinosaur tracks which have been naturally split into layers, to reconstruct foot motions of animals living over 200 million years ago. We consider these reconstructions to be hypotheses of motion. To test these hypotheses, we use the Discrete Element Method, in which individual particles of substrate are simulated in response to a penetrating foot model. Simulations that produce virtual tracks morphologically similar to the fossils lend support to the motion being plausible, while simulations that result in very different final tracks serve to reject the hypothesis of motion and help generate a new hypothesis

    Forefoot pathology in rheumatoid arthritis identified with ultrasound may not localise to areas of highest pressure: cohort observations at baseline and twelve months

    BackgroundPlantar pressures are commonly used as clinical measures, especially to determine optimum foot orthotic design. In rheumatoid arthritis (RA) high plantar foot pressures have been linked to metatarsophalangeal (MTP) joint radiological erosion scores. However, the sensitivity of foot pressure measurement to soft tissue pathology within the foot is unknown. The aim of this study was to observe plantar foot pressures and forefoot soft tissue pathology in patients who have RA.Methods A total of 114 patients with established RA (1987 ACR criteria) and 50 healthy volunteers were assessed at baseline. All RA participants returned for reassessment at twelve months. Interface foot-shoe plantar pressures were recorded using an F-ScanĀ® system. The presence of forefoot soft tissue pathology was assessed using a DIASUS musculoskeletal ultrasound (US) system. Chi-square analyses and independent t-tests were used to determine statistical differences between baseline and twelve months. Pearsonā€™s correlation coefficient was used to determine interrelationships between soft tissue pathology and foot pressures.ResultsAt baseline, RA patients had a significantly higher peak foot pressures compared to healthy participants and peak pressures were located in the medial aspect of the forefoot in both groups. In contrast, RA participants had US detectable soft tissue pathology in the lateral aspect of the forefoot. Analysis of person specific data suggests that there are considerable variations over time with more than half the RA cohort having unstable presence of US detectable forefoot soft tissue pathology. Findings also indicated that, over time, changes in US detectable soft tissue pathology are out of phase with changes in foot-shoe interface pressures both temporally and spatially.Conclusions We found that US detectable forefoot soft tissue pathology may be unrelated to peak forefoot pressures and suggest that patients with RA may biomechanically adapt to soft tissue forefoot pathology. In addition, we have observed that, in patients with RA, interface foot-shoe pressures and the presence of US detectable forefoot pathology may vary substantially over time. This has implications for clinical strategies that aim to offload peak plantar pressures

    Group cognitive behavioural therapy for stroke survivors with depression and their carers

    Background: Depression in stroke survivors is common, leads to poorer outcomes and often not treated. A group cognitive behavioural therapy (CBT) program (Brainstorm) for stroke survivors with depression, and their carers has been running as part of usual care since 2007. Objective: To evaluate the implementation and acceptability of Brainstorm, a closed group intervention consisting of up to 10 sessions of education, activity planning, problem solving and thought challenging. Methods: Participating stroke survivors and their carers complete assessment measures at baseline, post-treatment and 1-month and 6-months follow-up. A mixed models for repeated measures data was conducted with depression and anxiety scores for stroke survivors (Beck Depression Inventory-II; Hospital Anxiety and Depression Scale) and the assessment of depression, anxiety and carer burden for carers. Acceptability was assessed by session attendance and written and open participant feedback upon completion of the program. Results: Forty-eight community dwelling stroke survivors and 34 carers attended Brainstorm, with a median attendance of 88% of sessions. Follow-up assessments were completed by 77% (post-treatment), 46% (1-month) and 38% (6-month) of stroke survivors. Stroke survivorsā€™ depression scores decreased from baseline to post-treatment (p<.001); maintained at 1-month (p<.001) but not at 6-month (p=.056). Anxiety scores decreased between baseline and 1-month (p=.013). Carer burden, depression and anxiety scores at 1-month and 6-month follow-up, for carers, were all reduced when compared with baseline (p<.05). Conclusion: The Brainstorm group intervention for depression in stroke survivors appears to have been effectively implemented and is acceptable to stroke survivors and carers
