    Stress and coping skills are among the most essential components of the mental health counseling field. The use of coping skills (e.g., meditation, physical activities, appropriate uses of leisure) has been identified as an effective strategy for stress management. Adventure therapy has emerged as a modality that can positively augment other therapeutic approaches by improving coping skills and assisting clients in managing stress. As with all therapies, a positive working alliance has been found to be important toward achieving clinical outcomes. This study explored how adventure therapy enhanced learned coping strategies for stress and improved therapeutic alliance. Outcomes from this exploratory research highlighted the potential of adventure therapy to decrease stress, increase coping skills, and build therapeutic rapport with the therapist

    A Mental Health Self-Screening Tool for Graduate Students

    Background: Excessive perceived stress and mental health crises are escalating among college students. About 50% meet mental health disorder criteria, and half of them receive inadequate treatment. Aim: This study aimed to evaluate the feasibility and outcomes of a 15-20 minute online, anonymous, mental health self-screening survey and resource tool for graduate students across a large midwestern university campus. Methods: A descriptive, correlational design addressed the study's aims. Recruitment occurred through multiple campus communication mechanisms and included a brief study description, quick reference (QR) code, and weblink to access the tool and survey. Data collected with the tool included demographic items and scores from standardized screening instruments measuring burnout, perceived stress, depression, anxiety, post-traumatic stress disorder (PTSD), alcohol misuse, and health behavior practices. Descriptive and correlational statistics were computed. Results: Among 778 graduate students who accessed and completed the survey, nearly 60% met the burnout threshold, 58% scored 8 (of 16) on the stress scale, 32% met the depression threshold, 47% met the anxiety threshold, 54% reported one or more symptoms of PTSD, and 38% reported poor health behaviors. Significant positive correlations (r = 0.184 to 0.615) were found between burnout, perceived stress, depression, and anxiety. Health behavior scores were significantly negatively correlated (r = -0.151 to -0.283) with burnout, perceived stress, depression, and anxiety. Scores differed by gender, race, and graduate student status (part-time vs. full-time). Female identifying students, age, and hours worked per week were associated with various outcome scores. Students commented that the tool needed more specific resources tailored to their scores. Conclusions: The data support the need for tailored coping resources based on student self-reported data. Video-based micromodules guiding individuals through coping skills (breathing, mindfulness, identifying negative thoughts, gratitude exercises) have since been developed and will be studied as a tailored resource for students

    Defective myogenesis in the absence of the muscle-specific lysine methyltransferase SMYD1

    AbstractThe SMYD (SET and MYND domain) family of lysine methyltransferases harbor a unique structure in which the methyltransferase (SET) domain is intervened by a zinc finger protein–protein interaction MYND domain. SMYD proteins methylate both histone and non-histone substrates and participate in diverse biological processes including transcriptional regulation, DNA repair, proliferation and apoptosis. Smyd1 is unique among the five family members in that it is specifically expressed in striated muscles. Smyd1 is critical for development of the right ventricle in mice. In zebrafish, Smyd1 is necessary for sarcomerogenesis in fast-twitch muscles. Smyd1 is expressed in the skeletal muscle lineage throughout myogenesis and in mature myofibers, shuttling from nucleus to cytosol during myoblast differentiation. Because of this expression pattern, we hypothesized that Smyd1 plays multiple roles at different stages of myogenesis. To determine the role of Smyd1 in mammalian myogenesis, we conditionally eliminated Smyd1 from the skeletal muscle lineage at the myoblast stage using Myf5cre. Deletion of Smyd1 impaired myoblast differentiation, resulted in fewer myofibers and decreased expression of muscle-specific genes. Muscular defects were temporally restricted to the second wave of myogenesis. Thus, in addition to the previously described functions for Smyd1 in heart development and skeletal muscle sarcomerogenesis, these results point to a novel role for Smyd1 in myoblast differentiation

    Arid3b Is Critical for B Lymphocyte Development

    Arid3a and Arid3b belong to a subfamily of ARID (AT-rich interaction domain) transcription factors. The Arid family is involved in regulating chromatin accessibility, proliferation, and differentiation. Arid3a and Arid3b are closely related and share a unique REKLES domain that mediates their homo- and hetero-multimerization. Arid3a was originally isolated as a B cell transcription factor binding to the AT rich matrix attachment regions (MARS) of the immunoglobulin heavy chain intronic enhancer. Deletion of Arid3a results in a highly penetrant embryonic lethality with severe defects in erythropoiesis and hematopoietic stem cells (HSCs). The few surviving Arid3a-/- (<1%) animals have decreased HSCs and early progenitors in the bone marrow, but all mature lineages are normally represented in the bone marrow and periphery except for B cells. Arid3b-/- animals die around E7.5 precluding examination of hematopoietic development. So it is unclear whether the phenotype of Arid3a loss on hematopoiesis is dependent or independent of Arid3b. In this study we circumvented this limitation by also examining hematopoiesis in mice with a conditional allele of Arid3b. Bone marrow lacking Arid3b shows decreased common lymphoid progenitors (CLPs) and downstream B cell populations while the T cell and myeloid lineages are unchanged, reminiscent of the adult hematopoietic defect in Arid3a mice. Unlike Arid3a-/- mice, HSC populations are unperturbed in Arid3b-/- mice. This study demonstrates that HSC development is independent of Arid3b, whereas B cell development requires both Arid3a and Arid3b transcription factors

    BFORE: The B-mode Foreground Experiment

    The B-mode Foreground Experiment (BFORE) is a proposed NASA balloon project designed to make optimal use of the sub-orbital platform by concentrating on three dust foreground bands (270, 350, and 600 GHz) that complement ground-based cosmic microwave background (CMB) programs. BFORE will survey ~1/4 of the sky with 1.7 - 3.7 arcminute resolution, enabling precise characterization of the Galactic dust that now limits constraints on inflation from CMB B-mode polarization measurements. In addition, BFORE's combination of frequency coverage, large survey area, and angular resolution enables science far beyond the critical goal of measuring foregrounds. BFORE will constrain the velocities of thousands of galaxy clusters, provide a new window on the cosmic infrared background, and probe magnetic fields in the interstellar medium.     Spinal Locomotor Circuits Develop Using Hierarchical Rules Based on Motorneuron Position and Identity

    SummaryThe coordination of multi-muscle movements originates in the circuitry that regulates the firing patterns of spinal motorneurons. Sensory neurons rely on the musculotopic organization of motorneurons to establish orderly connections, prompting us to examine whether the intraspinal circuitry that coordinates motor activity likewise uses cell position as an internal wiring reference. We generated a motorneuron-specific GCaMP6f mouse line and employed two-photon imaging to monitor the activity of lumbar motorneurons. We show that the central pattern generator neural network coordinately drives rhythmic columnar-specific motorneuron bursts at distinct phases of the locomotor cycle. Using multiple genetic strategies to perturb the subtype identity and orderly position of motorneurons, we found that neurons retained their rhythmic activity—but cell position was decoupled from the normal phasing pattern underlying flexion and extension. These findings suggest a hierarchical basis of motor circuit formation that relies on increasingly stringent matching of neuronal identity and position

    Behavioural responses of pasture based dairy cows to short term management in tie-stalls

The objective of this study was to establish effects of transfer from pasture, to tie-stalls in a metabolism house, then back to pasture, on lying behaviour and locomotion score of lactating cows: Holstein-Friesian (H, n = 16), Jersey (J, n = 16) and H × J (HJ, n = 16). Cows were transferred to tie-stalls on d 1 for 12 days, and were offered freshly cut ryegrass according to herbage allowance (HERB) and genotype: J low = 14; J high = 17; H and HJ low = 16; and H and HJ high = 20 kg DM/d. Lying behaviour was recorded on four days: −2, −1 (Pre-confinement), 3 (Early confinement), 10, 11 (Late confinement), 13 and 14 (Post-confinement) relative to transfer (d 1) using dataloggers, and was also video-recorded during the first 15 h. Locomotion score was recorded on days −4, −3, 12 and 16. No effects of HERB on lying variables were observed during the first 15 h in confinement, but J cows made more lying intentions (21.0 vs. 12.2; P < 0.05) and tended (P = 0.07) to have a shorter latency to lie. Cows spent less (P < 0.001) time lying in early confinement (07:22:29 h/d) than on any of the other occasions (9:12:50 h/d). Cows had more (P < 0.001) and shorter (P < 0.001) lying bouts in confinement than while at pasture. Low HERB cows spent more time lying than high HERB cows (09:54:55 vs. 09:09:33 h/d; P < 0.01). J had higher locomotion scores than H (9.2 ± 0.2 vs.7.8 ± 0.2; P < 0.001), and tended (P = 0.09) to have higher scores than HJ (8.5 ± 0.2) cows. Locomotion scores were lowest pre confinement, highest at turnout (d 12), and intermediate after that at pasture (d 16) (7.6 ± 0.2, 9.3 ± 0.2 and 8.6 ± 0.3, respectively; P < 0.01). On transfer to the metabolism house cows showed disrupted patterns of lying although daily lying time returned to levels similar to pasture by late confinement. Confinement also resulted in a short-term deterioration in locomotory ability, which although improving, was still evident 4 days following the cows return to pasture with Jersey cows being more affected than the other genotypes. These findings suggest that longer adaptation periods and temporary release to loafing areas may improve both the validity of data collected and cow welfare
