Priprava vina na niskoj temperaturi s pomoću kvasca imobiliziranog na pivskom tropu bez lignina
Preservation, risk of contamination, transportation and storage costs (e.g. refrigeration) are the main problems associated with wet, active starter cultures such as yeast. To deal with these aspects, drying of commercial cultures is required. A thermally dried biocatalyst has been prepared by immobilization of the psychrotolerant yeast strain Saccharomyces cerevisiae AXAZ-1 on delignified brewer’s spent grains, followed by simple thermal drying or by air stream at 30 and 35 °C. Repeated batch fermentations of grape must (11.5 °Be, corresponding to 196 g/L of fermentable sugar) using the dried biocatalysts were performed at successively reduced temperatures (15, 10 and 5 °C). The fermented samples were analyzed for residual sugar, alcohol and free cell concentrations. The alcohol yield, alcohol productivity and sugar conversion were calculated in order to estimate the optimum conditions of drying. The volatile constituents in the produced wines were analysed by GC analysis. The results showed that drying of immobilized cells by the proposed techniques did not affect their viability and fermentative activity. High alcohol productivity, increased esters and lower concentrations of higher alcohols obtained by low-temperature fermentation using the dried biocatalysts indicate a potential improvement of product quality.Konzerviranje, rizik od onečišćenja, te cijena transporta i skladištenja (npr. hlađenja) najčešći su problemi pri korištenju vlažnih, aktivnih starter-kultura, tj. kvasaca. Stoga komercijalne kulture kvasaca treba osušiti. Sušeni je biokatalizator pripremljen imobilizacijom psihrotolerantnoga soja kvasca Saccharomyces cerevisiae AXAZ-1 na pivskom tropu, iz kojeg je prethodno uklonjen lignin, a zatim sušenjem toplinom ili strujom zraka pri 30 i 35 °C. Pomoću tako pripremljenog kvasca uspješno je provedeno nekoliko uzastopnih šaržnih postupaka vrenja mošta (11,5 °Be, proporcionalno 196 g/L fermentabilnog šećera) pri nižim temperaturama (15, 10 i 5 °C). Ispitana je koncentracija preostalog šećera, dobivenog alkohola i slobodnih stanica u fermentiranim uzorcima. Da bi se procijenili optimalni uvjeti sušenja, izračunati su ovi parametri: udio i produktivnost alkohola te konverzija šećera. Volatilni su sastojci vina analizirani plinskom kromatografijom. Rezultati pokazuju da sušenje imobiliziranih stanica kvasca predloženim metodama ne utječe na njegovu vitalnost i fermentacijsku aktivnost. Kakvoća se proizvoda može poboljšati korištenjem sušenih stanica kvasca pri nižim temperaturama, što je uočeno iz većeg udjela alkohola, veće koncentracije estera i manje koncentracije viših alkohola u vinu
Characterising rhamnolipid production in Burkholderia thailandensis E264, a non-pathogenic producer
Burkholderia thailandensis E264 is a rhamnolipid (RL)-producing gram-negative bacterium first isolated from the soils and stagnant waters of central and north-eastern Thailand. Growth of B. thailandensis E264 under two different incubation temperatures (25 and 30 °C) resulted in a significantly higher dry cell biomass production at 30 °C (7.71 g/l) than at 25 °C (4.75 g/l) after 264 h; however, incubation at the lower temperature resulted in consistently higher concentration of RL production throughout the growth period. After 264 h, the concentration of crude RL extract for the 25 °C culture was 2.79 g/l compared to 1.99 g/l for the 30 °C culture. Overall RL production concentration after 264 h was 0.258 g/g dry cell biomass (DCB) for the 30 °C culture compared to 0.587 g/g DCB for the 25 °C culture. Real-time PCR (qPCR) was also used to analyse expression of the RL biosynthesis genes throughout the incubation period at 25 °C showing that the expression of the rhlA, rhlB and rhlC genes is continuous. During the log and early stationary phases of growth, expression levels remain low and are increased upon entry to the late stationary phase. B. thailandensis E264 produces mostly di-RLs and the Di-RL C14-C14 in most abundance (41.88 %). Fermentations were also carried out in small-scale bioreactors (4 l working volume) under controlled conditions, and results showed that RL production was maintained. Our findings show that B. thailandensis E264 has excellent potential for industrial scale RL production
Use of brewers' wastes for the production of immobilized dried yeasts
The aim of the doctoral thesis is the utilization of brewers’ wastes. In
the introduction part of this dissertation the chemical composition of brewers’
wastes is reported. Furthermore the innovative methods of production and
the drying methods of starter cultures that can be used in the wine making
and brewing are also examined. In the present thesis the application of simple
thermal drying methods in low temperatures was examined. Thermal drying
was performed in thin layer, on glass plates in order to achieve a more rapid
removal of humidity and consequently high viability of cells. Thermal drying
was applied in free cells of S. cerevisiae and the fermentability that was
compared with freeze dried cells was higher and similar to that of pressed
bakers’ yeast, a fact that proves that thermal drying can be competitive to
freeze-drying, which is the drying method that is mainly applied in industry.
Consequently biocatalysts that were prepared with immobilized cells
S. cerevisiae on brewers’ spent grains were thermally dried in an incubator,
thermally dried in air stream and under vacuum, in low and high
temperatures. Thermally dried immobilized biocatalysts presented high
fermentation ability and lower fermentation time in comparison with
thermally dried free cells. The effect of temperature at the duration of drying
didn’t influence the biocatalysts’ activity. Thermally dried biocatalyst is
resistant at higher temperatures up to 70°C. Fermentations at 15°C were
performed by thermally dried free cells that were dried at temperature up to
45°C, resulted to much higher fermentation time. Also, storage for 4 months
of thermally dried biocatalysts didn’t influence particularly their fermentation
ability.
In next section, immobilised biocatalysts that were dried at low
temperatures were used for the production of wine at extremely low
temperatures. Thermally dried biocatalysts showed stability at repeated
fermentation batches. Fermentation time achieved by thermally dried
biocatalysts even at 5°C is lower in comparison with the time of natural
fermentations at 25-30°C. The quality of wine that is produced is fine, as
volatile by-products are within the desirable limits. Consequently, thermally
dried biocatalysts can be considered as good winemaking yeasts of low cost.
Therefore, it is obvious that brewers’ spent grains can be used as
immobilisation support for the production of thermally dried biocatalysts.
Liquid waste of brewery can be utilized with recovery of brewers’ yeast and
production of thermally dried immobilized biocatalysts, as previously
described. Thermally dried biocatalysts that were produced from liquid and
solid wastes of brewery were used for the production of wine and beer. The
activity of these biocatalysts depends on viability of rejected yeast of brewery.
The results of repeated batch fermentations of grape must show that the
activity of these biocatalysts is high, particularly at extremely low
temperatures, corresponding with that of thermally dried biocatalysts that
were produced with immobilization of S. cerevisiae on delignified brewers’
spent grains. Consequently thermally dried biocatalysts that were prepared
by waste of brewery can be used as starter cultures for vinification and
brewing and give products of fine quality. Biocatalysts are of low cost due to
the low cost of raw material and the low cost of the production method.
Then brewers’ spent grains were examined for production of sugars
with degradation of starch with use of Aspergillus awamori, as well as for
production of bioethanol with use of co-culture of A. awamori and S. cerevisiae.
Low concentration of starch of brewers’ spent grains produced low
concentrations of sugars and bioethanol. However the chemical composition
of brewers’ spent grains depends on many factors, and for this reason, higher
concentrations of starch has been reported. The application of this method
would be efficient on brewers’ spent grains with higher concentrations of
starch.
Finally, brewers’ spent grains were used in solid state fermentations
for upgrading of brewers’ spent grains through single cell protein production.
Various factors, such as low temperature, low pH and spore concentration A.
awamori, were examined. It was observed that the aerobic microorganism was
grown on the surface of brewers’ spent grains. Therefore, when a thin layer of
brewers’ spent grains was used, the production of proteins was higher. Thus
brewers’ spent grains after the enrichment in proteins give a new added value
product that can be used as animal feed. The microorganism that is used,
even if it belongs in the family of Aspergillus, is not toxic.Η διδακτορική διατριβή έχει ως στόχο την αξιοποίηση των αποβλήτων
της ζυθοποιίας για τη δημιουργία προστιθέμενης αξίας. Στη βιβλιογραφική
ανασκόπηση της διατριβής περιγράφονται η σύσταση και οι μέχρι τώρα
χρήσεις των αποβλήτων της ζυθοποιίας, καθώς και οι καινοτόμες μέθοδοι
παραγωγής και οι μέθοδοι ξήρανσης των αρχικών καλλιεργειών που μπορούν
να χρησιμοποιηθούν στην οινοποιία και ζυθοποιία. Στο πειραματικό μέρος
και στη συζήτηση των αποτελεσμάτων εξετάζονται (i) η παραγωγή θερμικά
ξηρανθέντων βιοκαταλυτών με χρήση των αποβλήτων της ζυθοποιίας, (ii) η
σακχαροποίηση του αμύλου και η ζύμωση με συγκαλλιέργεια, των στερεών
αποβλήτων της ζυθοποιίας και (iii) ο εμπλουτισμός με πρωτεΐνες των στερεών
αποβλήτων. Αναλυτικότερα, στην παρούσα διατριβή εξετάστηκε η εφαρμογή
απλών μεθόδων θερμικής ξήρανσης σε χαμηλές θερμοκρασίες. Η ξήρανση
πραγματοποιήθηκε σε λεπτό στρώμα, σε γυάλινες πλάκες ώστε η
απομάκρυνση της υγρασίας να είναι ταχύτερη και επομένως η διατήρηση της
βιωσιμότητας των κυττάρων να είναι η μέγιστη δυνατή. Η ξήρανση σε
θερμοθάλαμο εξετάστηκε αρχικά σε ελεύθερα κύτταρα S. cerevisiae και η
ικανότητα ζύμωσης των θερμικά ξηρανθέντων κυττάρων σε σύγκριση με τα
λυοφιλιωμένα κύτταρα ήταν υψηλότερη και όμοια με εκείνη της πιεστής
ζύμης αρτοποιίας. Αυτό δείχνει ότι η ξήρανση σε θερμοθάλαμο μπορεί να
είναι ανταγωνιστική της λυοφιλίωσης, που είναι η μέθοδος ξήρανσης που
εφαρμόζεται κυρίως στη βιομηχανία.
Στη συνέχεια εξετάστηκαν βιοκαταλύτες που παρασκευάστηκαν με
ακινητοποίηση κυττάρων S. cerevisiae σε απολιγνινοποιημένα υπολείμματα
κριθαριού της ζυθοποιίας που υπέστησαν ξήρανση λεπτής στιβάδας σε
θερμοθάλαμο, σε ρεύμα αέρα και υπό κενό σε χαμηλές και υψηλές
θερμοκρασίες. Οι θερμικά ξηρανθέντες ακινητοποιημένοι βιοκαταλύτες,
κυρίως σε θερμοθάλαμο και ρεύμα αέρα, παρουσίασαν υψηλή ικανότητα
ζύμωσης και μικρότερους χρόνους ζύμωσης σε σχέση με τα θερμικά
ξηρανθέντα ελεύθερα κύτταρα. Η επίδραση της θερμοκρασίας κατά τη
διάρκεια της ξήρανσης δεν επηρέασε τη δραστικότητα των βιοκαταλυτών. Οι
θερμικά ξηρανθέντες βιοκαταλύτες είναι ανθεκτικοί στις υψηλότερες
θερμοκρασίες έως 70°C. Τα θερμικά ξηρανθέντα ελεύθερα κύτταρα που
ξηράνθηκαν σε θερμοκρασίες μέχρι 45°C, πραγματοποίησαν ζυμώσεις στους
15°C, ωστόσο οι χρόνοι ζύμωσης ήταν πολύ μεγαλύτεροι από εκείνους των
ακινητοποιημένων κυττάρων στην ίδια θερμοκρασία. Επίσης η αποθήκευση
για 4 μήνες των θερμικά ξηρανθέντων βιοκαταλύτων δεν επηρέασε ιδιαίτερα
την ικανότητα ζύμωσης τους.
Μετά οι ακινητοποιημένοι βιοκαταλύτες που ξηράνθηκαν σε χαμηλές
θερμοκρασίες χρησιμοποιήθηκαν για την παραγωγή οίνου σε εξαιρετικά
χαμηλές θερμοκρασίες. Οι θερμικά ξηρανθέντες βιοκαταλύτες έδειξαν
σταθερότητα κατά τις επαναλαμβανόμενες ζυμώσεις. Οι χρόνοι ζύμωσης
ακόμη και στους 5°C ήταν μικρότεροι από τους χρόνους των φυσικών
ζυμώσεων που πραγματοποιούνται στους 25-30°C. Η ποιότητα του οίνου που
παράγεται είναι καλή, αφού ο σχηματισμός των πτητικών παραπροϊόντων
είναι εντός των επιθυμητών ορίων. Συνεπώς οι θερμικά ξηρανθέντες
βιοκαταλύτες αποτελούν καλές οινοποιητικές ζύμες, χαμηλού κόστους
παραγωγής.
Σύμφωνα με τα παραπάνω αποτελέσματα είναι φανερό ότι τα
υπολείμματα κριθαριού της ζυθοποιίας αποτελούν καλό φορέα
ακινητοποίησης για την παρασκευή θερμικά ξηρανθέντων βιοκαταλύτων. Για
την αξιοποίηση των υγρών αποβλήτων της ζυθοποιίας έγινε ανάκτηση της
ζύμης και παρασκευάστηκαν θερμικά ξηρανθέντες ακινητοποιημένοι
βιοκαταλύτες με βάση το μοντέλο βιοκαταλυτών που εξετάστηκαν
προηγουμένως. Οι θερμικά ξηρανθέντες βιοκαταλύτες από υγρά και στερεά
απόβλητα της ζυθοποιίας χρησιμοποιήθηκαν για την παρασκευή οίνου και
ζύθου. Η δραστικότητα των βιοκαταλυτών αυτών εξαρτάται από την
καθαρότητα και ζωτικότητα της απορριπτόμενης ζύμης ζυθοποιίας. Τα
αποτελέσματα των επαναλαμβανόμενων ζυμώσεων γλεύκους και
ζυθογλεύκους δείχνουν ότι η δραστικότητα των βιοκαταλυτών αυτών είναι
υψηλή ιδιαίτερα στις πολύ χαμηλές θερμοκρασίες, αντίστοιχη με εκείνη των
ξηρανθέντων βιοκαταλυτών που προκύπτουν με ακινητοποίηση κυττάρων S.
cerevisiae. Συνεπώς οι θερμικά ξηρανθέντες βιοκαταλύτες που
παρασκευάζονται από απόβλητα της ζυθοποιίας μπορούν να
χρησιμοποιηθούν ως αρχικές καλλιέργειες οινοποίησης και ζυθοποίησης,
δίνοντας προϊόντα καλής ποιότητας. Είναι χαμηλού κόστους λόγω του σχεδόν
μηδενικού κόστους της πρώτης ύλη και του χαμηλού κόστους της μεθόδου
παρασκευής.
Στη συνέχεια εξετάστηκε η χρήση των υπολειμμάτων κριθαριού της
ζυθοποιίας για σακχαροποίηση του αμύλου από τον Aspergillus awamori,
καθώς επίσης και για παραγωγή βιοαιθανόλης με συγκαλλιέργεια A. awamori-
S. cerevisiae. Η μικρή περιεκτικότητα αμύλου των υπολειμμάτων κριθαριού
της ζυθοποιίας έδωσε μικρές συγκεντρώσεις σακχάρου και βιοαιθανόλης.
Ωστόσο η σύσταση των υπολειμμάτων εξαρτάται από πολλούς παράγοντες
και γι’ αυτό υπάρχουν αναφορές για μεγαλύτερες συγκεντρώσεις αμύλου στα
υπολείμματα, όπου η μέθοδός θα ήταν πιο αποδοτική.
Τέλος, τα υπολείμματα κριθαριού της ζυθοποιίας χρησιμοποιήθηκαν σε
ζυμώσεις στερεάς κατάστασης για παραγωγή μονοκυτταρικής πρωτεΐνης.
Εξετάστηκαν διάφοροι παράγοντες, όπως η χαμηλή θερμοκρασία, το χαμηλό
pH και η συγκέντρωση του A. awamori. Παρατηρήθηκε ότι ο
μικροοργανισμός, που είναι αερόβιος, αναπτύσσεται στην επιφάνεια των
υπολειμμάτων. Όταν λοιπόν χρησιμοποιήθηκε πολύ λεπτό στρώμα
υπολειμμάτων κριθαριού, η παραγωγή των πρωτεϊνών ήταν μεγάλη. Έτσι τα
υπολείμματα κριθαριού της ζυθοποιίας μετά τον εμπλουτισμό τους σε
πρωτεΐνες δίνουν ένα νέο προϊόν με προστιθέμενη αξία, που μπορεί να
χρησιμοποιηθεί ως ζωοτροφή, αφού ο μικροοργανισμός που χρησιμοποιείται,
παρόλο που ανήκει στην οικογένεια Aspergillus, δεν είναι τοξικός
Bioethanol production from brewers' solid wastes by co-immobilized co-culture Aspergillus awamori-Saccharomyces cerevisiae
The signing of the Kyoto Protocol (KP) in 1997 commands the substitution of fossil fuels by alternative fuels for the reduction of the emission of greenhouse gases. The Environmental strategy (EC, 1997) necessitated the increase of energy production from renewables. Utilization of Brewers’ Spent Grain (BSG) for starch hydrolysis and bioethanol production was studied. BSG is a solid agro-industrial waste material produced from the brewing process. Member countries of the European Community generate millions of tons annually, but their utilization still remains limited. Aspergillus awamori was used for starch conversion and Saccharomyces cerevisiae for sugar conversion to bioethanol. HPLC and GC-FID analysis were performed for the determination of produced sugars and bioethanol, respectively. Pretreatments of BSG were examined for the enhancement of hydrolysis rate and bioethanol concentration. Finally, a co-immobilized co-culture A. awamori–S. cerevisiae was used for bioethanol production. This system reduces the required time and the equipment cost. Both microorganisms were immobilized on delignified BSG. Electron microscope spectroscopy was used for the observation of cells immobilization. High conversion rate of BSG starch to bioethanol was achieved by this biological, low-cost and environmentally friendly method that could be applied in the utilization of starchy agro-industrial wastes

Unusual N Gene Dropout and Ct Value Shift in Commercial Multiplex PCR Assays Caused by Mutated SARS-CoV-2 Strain
Several SARS-CoV-2 variants have emerged and early detection for monitoring their prevalence is crucial. Many identification strategies have been implemented in cases where sequencing data for confirmation is pending or not available. The presence of B.1.1.318 among prevalent variants was indicated by an unusual amplification pattern in various RT-qPCR commercial assays. Positive samples for SARS-CoV-2, as determined using the Allplex SARS-CoV-2 Assay, the Viasure SARS-CoV-2 Real Time Detection Kit and the GeneFinder COVID-19 Plus RealAmp Kit, presented a delay or failure in the amplification of the N gene, which was further investigated. Whole-genome sequencing was used for variant characterization. The differences between the mean Ct values for amplification of the N gene vs. other genes were calculated for each detection system and found to be at least 14 cycles. Sequencing by WGS revealed that all the N gene dropout samples contained the B.1.1.318 variant. All the isolates harbored three non-synonymous mutations in the N gene, which resulted in four amino acid changes (R203K, G204R, A208G, Met234I). Although caution should be taken when the identification of SARS-CoV-2 variants is based on viral gene amplification failure, such patterns could serve as a basis for rapid and cost-effective screening, functioning as indicators of community circulation of specific variants, requiring subsequent verification via sequencing
