    The bumble bees (Hymenoptera: Apidae: Bombus) of Arkansas, fifty years later

    Many species of bumble bees (Hymenoptera: Apidae: Bombus Latreille) are declining throughout their ranges in North America, yet detecting population trends can be difficult when historical survey data are lacking.  In the present study, contemporary data is compared to a 1965 survey to detect changes in bumble bee distributions throughout Arkansas.  Using county-level records as a point of comparison to look for changes in state-wide occurrence among species over time, we find that state-level changes reflect national trends.  Contemporary records of Bombus bimaculatus Cresson and B. impatiens Cresson have more than tripled, while records for B. pensylvanicus (De Geer) show a decline to 61% of historical levels.  Although B. fervidus (Fabricius) has been reported infrequently in the state, misidentifications may have led to an overestimation of the state’s species richness.  In addition to an updated assessment of the bumble bees of Arkansas, we also provide new, localized information on the seasonal phenology and plant preferences of each species that can be used to guide conservation efforts

    From phase- to amplitude-fluctuation driven superconductivity in systems with precursor pairing

    The change-over from phase- to amplitude-fluctuation driven superconductivity is examined for a composite system of free electrons (Fermions with concentration n_F) and localized electron-pairs (hard-core Bosons with concentration n_B) as a function of doping-changing n_B. The coupling together of these two subsystems via a charge exchange term induces electron pairing below a certain T^* (showing up in form of a pseudogap) and ultimately superconductivity in the Fermionic subsystem. T^* steadily decreases with decreasing n_B. Below T^* this electron pairing leads to electron-pair resonant states (Cooperons) with quasi-particle features which strongly depend on nBn_B. For high concentrations, (n_B \simeq 0.5), correlation effects between the hard-core Bosons lead to itinerant Cooperons having a heavy mass m_p, but are long-lived. Upon reducing n_B, the mass as well as the lifetime of those Cooperons is considerably reduced. For high values of n_B, a superconducting state sets in at a T_c, being controlled by the phase stiffness D_\phi=\hbar^2 n_p/m_p of those Cooperons, where n_p denotes their density. Upon reducing n_B, the phase stiffness steadily increases, and eventually exceeds the pairing energy k_B T^*. The Cooperons loose their well defined itinerant quasi-particle features and superconductivity gets controlled by amplitude fluctuations. The resulting phase diagram with doping is reminiscent of that of the phase fluctuation scenario for high T_c superconductivity, except that in our scenario the determinant factors are the mass and the lifetime of the Cooperons rather than their density.Comment: 13 pages, 12 figure

    Convergence of Wnt signalling on the HNF4a-driven transcription in controlling liver zonation

    BACKGROUND & AIMS: In each hepatocyte, the specific repertoire of gene expression is influenced by its exact location along the portocentrovenular axis of the hepatic lobule and provides a reason for the liver functions compartmentalization defined "metabolic zonation." So far, few molecular players controlling genetic programs of periportal (PP) and perivenular (PV) hepatocytes have been identified; the elucidation of zonation mechanisms remains a challenge for experimental hepatology. Recently, a key role in induction and maintenance of the hepatocyte heterogeneity has been ascribed to Wnt/beta-catenin pathway. We sought to clarify how this wide-ranging stimulus integrates with hepatocyte specificity. METHODS: Reverse transcriptase polymerase chain reaction (RT-PCR) allowed the transcriptional profiling of hepatocytes derived from in vitro differentiation of liver stem cells. The GSK3beta inhibitor 6-bromoindirubin-3'-oxime (BIO) was used for beta-catenin stabilization. Co-immunoprecipitations were used to study biochemical protein interactions while ChIP assays allowed the in vivo inspection of PV and PP genes regulatory regions. RESULTS: We found that spontaneous differentiation of liver stem cells gives rise to PP hepatocytes that, after Wnt pathway activation, switch into PV hepatocytes. Next, we showed that the Wnt downstream player LEF1 interacts with the liver-enriched transcriptional factor HNF4alpha. Finally, we unveiled that the BIO induced activation of PV genes correlates with LEF1 binding to both its own and HNF4alpha consensus, and the repression of PP genes correlates with HNF4alpha displacement from its own consensus. CONCLUSION: Our data show a direct and hitherto unknown convergence of the canonical Wnt signaling on the HNF4alpha-driven transcription providing evidences of a mechanism controlling liver zonated gene expression

    An epistatic mini-circuitry between the transcription factors Snail and HNF4a controls liver stem cell and hepatocyte features exhorting opposite regulation on stemness-inhibiting microRNAs

    Preservation of the epithelial state involves the stable repression of EMT program while maintenance of the stem compartment requires the inhibition of differentiation processes. A simple and direct molecular mini-circuitry between master elements of these biological processes, may provide the best device to keep balanced such complex phenomena. In this work, we show that in hepatic stem cell Snail, a transcriptional repressor of the hepatocyte differentiation master gene HNF4, directly represses the expression of the epithelial microRNAs-200c and -34a, which in turn target several stem cell genes. Notably, in differentiated hepatocytes HNF4, previously identified as a transcriptional repressor of Snail, induces the microRNAs-34a and -200a, b, c that, when silenced, causes epithelial dedifferentiation and reacquisition of stem traits. Altogether these data unveiled Snail, HNF4 and microRNAs -200a, b, c and -34a as epistatic elements controlling hepatic stem cell maintenance/differentiation

    Null-Flux Coils in Permanent Magnets Bearings

    In this paper, the stability analysis of a new permanent magnets (PMs) bearings is presented and discussed. The suspension is assured by the repulsive force of properly shaped PMs placed on both the stator and the rotor. Then, exploiting currents induced on a system of null-flux coils attached to the stator, a stabilizing force for the translation of the center of mass of the rotor is obtained. The performance of the proposed bearing is investigated by a research code, previously developed at DESTEC and capable to simulate six degrees of freedom electromechanical devices

    Surgery on breast cancer in pregnancy

    Pregnancy-associated breast cancer (PABC) is defined as breast cancer develops either during or within 1 year after pregnancy, it is a rare disease arising in 1:3,000 to 1:10,000 pregnant women. Prognosis of this tumor is influenced by local or systemic treatment, which might be conditioned by gestational age and limited by the concern on potential adverse impact on fetus. The aim of this literature review is to analyze the main topics regarding surgical treatment of patients diagnosed with breast cancer in pregnancy: anesthesia and maternal-fetal monitoring, type of breast surgery, immediate breast reconstruction after radical surgery and management of the axilla. Some important topics remain controversial since the relative rarity of PABC precludes the feasibility of large studies leading to a lack of literature data. Multi-institutional collaboration is warranted to collect women with PABC, in order to best define surgical treatment in view of associated maternal and fetal risks

    HBV vaccination with Fendrix is effective and safe in pre-dialysis CKD population

    Background: Patients with chronic kidney disease have a poor response to hepatitis B vaccine due to the immunodeficiency conferred from chronic uremia. A recombinant HB vaccine containing an improved adjuvant system AS04 (HBV-AS04) has been manufactured but scarce evidence exists on HBV-AS04 use among patients with CKD. Aim: To assess efficacy and safety of an adjuvanted recombinant vaccine (HBV-AS04) in a large cohort of CKD patients at pre-dialysis stage (with susceptibility to HBV infection). Methods: Patients were prospectively enrolled to receive four 20-mcg doses of HBV-AS04 by intramuscular route (deltoid muscle) at months 1, 2, 3, and 4. Anti-HBs surface antibody concentrations were tested at intervals of 1, 2, 3, 4, and 12 months. Multivariate analyses were performed to assess the parameters, which predicted immunologic response to HBV-AS04 vaccine. Results: One hundred and seven patients were included and 102 completed the study. At completion of vaccine schedule, the frequency of responders (anti-HBs titers 65 10 mIU/mL) was 95% (97/102) (mean anti-HBs antibody titers, 688.9 \ub1 385 mIU/mL), according to per-protocol analysis. Serum haemoglobin levels were greater in responder than non- or low-responder patients to HBV-AS04 (P = 0.04) and this was confirmed by multivariate analysis. The seroprotection rate at month 50 was 88% (30/34) with lower anti-HBs antibody titers (218.5 \ub1 269.6 mIU/mL, P = 0.001). No major side effects were observed. Conclusions: Our prospective study performed in a real-world setting showed a high immunogenicity and safety of HBV-AS04 vaccine in patients with CKD not yet on maintenance dialysis. Studies provided with longer follow-ups are under way to assess the durability of seroprotection in responders

    Colony Breeding Structure of Reticulitermes (Isoptera: Rhinotermitidae) in Northwest Arkansas

    Termites, as social insects, have a complicated life cycle in which the colony breeding structure, that is the number of and origin of reproductives in a colony, can vary in relation to age and environmental factors. In this study, we used genetic methods to characterize the breeding structure of three species of Reticulitermes from three sites in northwest Arkansas and compared two habitats: undeveloped, forested sites and developed, agricultural sites. We found 57.1% of R. flavipes (Kollar) in northwest Arkansas (n = 28) were simple families, 39.3% were extended families and 3.6% were mixed families. Similarly, for R. hageni Banks (n = 23), we found 58.3% simple families, 33.3% extended families, and 8.3% mixed families. All of the R. virginicus (Banks) samples (n = 5) were simple families. For R. flavipes and R. hageni, the percentage of extended families is intermediate to southeastern and northern USA populations, corresponding to the intermediate seasonality and climate in Arkansas. The level of inbreeding in Arkansas, estimated via FIT, was relatively high and similar to northern populations of Reticulitermes. There were significantly more extended family colonies at the developed site compared to the two undeveloped sites which contained more simple family colonies. This difference may occur as a strategy to cope with sparse resources in urban environments or as a consequence of different abiotic factors

    The insulin-like growth factor 2 receptor (IGF2R) plays a major role in binding and regulating the circulating and tissue levels of the mitogenic peptide insulin-like growth factor 2 (IGF2). IGF2/IGF2R interaction influences cell growth, survival, and migration in normal tissue development, and the deregulation of IGF2R expression has been associated with growth-related disease and cancer. IGF2R overexpression has been implicated in heart and muscle disease progression. Recent research findings suggest novel approaches to target IGF2R action. This review highlights recent advances in the understanding of the IGF2R structure and pathways related to muscle homeostasis
