    Comparación de dos pruebas de campo para determinar la mastitis subclínica en bovinos en la localidad de Florida - Pomacochas, 2019

    Se evaluaron dos pruebas de campo para determinar la Mastitis Subclinica en bovinos. Se utilizó un total de 268 vacas en producción de leche, entre 1 a 10 partos, en dos sectores San Lorenzo y Miraflores de Levanto. En un sistema extensivo, su alimentación a base de pasturas naturales y cultivadas, un solo ordeño manual, y se consideró desde la segunda semana hasta 5 meses de lactación. Se utilizó el método biológico (Prueba California - Mastitis Test) y el método químico (Prueba de Whiteside), los que permitieron comprobar cuál de las pruebas fue más eficiente para la detección de la mastitis bovina. Los resultados demostraron que el 100 % de prevalencia de mastitis subclínica, según el número de partos, se reporta que el mayor porcentaje de prevalencia se muestra en el 4 parto y según los cuartos mamarios, se reporta que el anterior y posterior derecho reporta el 30 % de prevalencia. Se evidenció un 47% de efectividad en la prueba Whitside y el 53% en la prueba California Mastitis Test. Esto dificulta la confiabilidad en criadores de ganado con ambas pruebas.     Effect of a single intra-articular high molecular weight hyaluronan in a naturally occurring canine osteoarthritis model : a randomized controlled trial

    Research Areas: OrthopedicsBackground Osteoarthritis (OA) is a complex joint disease and chronic pain source, affecting a patient's quality of life and posing a financial burden. As the dog is considered a nearly ideal species for translation research of human OA and the most used model for research, exploring spontaneous dog OA under the One Health/One Medicine concept can improve both humans and dogs' health and well-being. Methods In a clinical treatment experiment, forty (N=40) joints were selected and randomly assigned to a control group (CG), which received 0.9% NaCl or a treatment (HG), which received Hylan G-F 20. Evaluations were performed on treatment day (T0), 8, 15, 30, 90, and 180 days post-treatment. They consisted of four different Clinical Metrology Instruments (CMI), evaluation of weight distribution, joint range of motion, thigh girth, radiographic and digital thermography imaging, synovial fluid interleukin-1 (IL-1), and C-reactive protein concentrations. Results were compared with repeated measures ANOVA, with a Huynh-Feldt correction, Paired samples T-test, or Wilcoxon signed-ranks test, with pResults Patients had a mean age of 6.5 +/- 2.4 years and a bodyweight of 26.6 +/- 5.2kg, and joints graded as mild (n=28, 70%), moderate (n=6, 15%), and severe OA (n=6, 15%). No differences were found between groups at T0. Symmetry index and deviation showed significant improvements in HG from 30 days (p<0.01) up to 180 days (p=0.01). Several CMI scores, particularly pain scores, improved from 90 to 180 days. Radiographic signs progressed in both groups. In both groups, increasing body weight and age corresponded to worse clinical presentation. IA hyaluronan administration produced increased lameness in six cases, which resolved spontaneously. Conclusions This study characterizes the response to treatment with Hylan G-F 20, which can produce significant functional and pain level improvements in patients with OA, even those with factors related to worse response to treatment.info:eu-repo/semantics/publishedVersio

    Gut microbiota features associated with Campylobacter burden and postnatal linear growth deficits in a Peruvian birth cohort

    BACKGROUND: Campylobacter infection is associated with impaired growth of children, even in the absence of symptoms. To examine the underlying mechanisms, we evaluated associations between Campylobacter infection, linear growth, and fecal microbial community features in a prospective birth cohort of 271 children with a high burden of diarrhea and stunting in the Amazonian lowlands of Peru. METHODS: Campylobacter was identified using a broadly reactive, genus-specific enzyme-linked immunosorbent assay. 16S rRNA-based analyses were used to identify bacterial taxa in fecal samples at ages 6, 12, 18, and 24 months (N = 928). Associations between infection, growth, and gut microbial community composition were investigated using multiple linear regression adjusting for within-child correlations, age, and breastfeeding. Indicator species analyses identified taxa specifically associated with Campylobacter burden. RESULTS: Ninety-three percent (251) of children had Campylobacter present in asymptomatic fecal samples during the follow-up period. A 10% increase in the proportion of stools infected was associated with mean reductions of 0.02 length-for-age z scores (LAZ) at 3, 6, and 9 months thereafter (P \u3c .01). We identified 13 bacterial taxa indicative of cumulative Campylobacter burden and 14 taxa significantly associated with high or low burden of enteroaggregative Escherichia coli, norovirus, or Giardia. CONCLUSIONS: Campylobacter infection is common in this cohort and associated with changes in microbial community composition. These results support the notion that disruptions to the fecal microbiota may help explain the observed effects of asymptomatic infections on growth in early life

    Diarrhea as a potential cause and consequence of reduced gut microbial diversity among undernourished children in Peru

    BACKGROUND: Detrimental effects of diarrhea on child growth and survival are well documented, but details of the underlying mechanisms remain poorly understood. Recent evidence demonstrates that perturbations to normal development of the gut microbiota in early life may contribute to growth faltering and susceptibility to related childhood diseases. We assessed associations between diarrhea, gut microbiota configuration, and childhood growth in the Peruvian Amazon. METHODS: Growth, diarrhea incidence, illness, pathogen infection, and antibiotic exposure were assessed monthly in a birth cohort of 271 children aged 0-24 months. Gut bacterial diversity and abundances of specific bacterial taxa were quantified by sequencing 16S rRNA genes in fecal samples collected at 6, 12, 18, and 24 months. Linear and generalized linear models were used to determine whether diarrhea was associated with altered microbiota and, in turn, if features of the microbiota were associated with the subsequent risk of diarrhea. RESULTS: Diarrheal frequency, duration, and severity were negatively associated with bacterial diversity and richness (P \u3c .05). Children born stunted (length-for-age z-score [LAZ] ≤ -2) who were also severely stunted (LAZ ≤ -3) at the time of sampling exhibited the greatest degree of diarrhea-associated reductions in bacterial diversity and the slowest recovery of bacterial diversity after episodes of diarrhea. Increased bacterial diversity was predictive of reduced subsequent diarrhea from age 6 to 18 months. CONCLUSIONS: Persistent, severe growth faltering may reduce the gut microbiota\u27s resistance and resilience to diarrhea, leading to greater losses of diversity and longer recovery times. This phenotype, in turn, denotes an increased risk of future diarrheal disease and growth faltering

    Car drivers' road safety performance. A benchmark across 32 countries

    The road safety performance of a country and the success of policy measures can be measured and monitored in different ways. In addition to the traditional road safety indicators based on the number of fatalities or injured people in road traffic crashes, complementary road safety performance indicators can be used in relation to vehicles, infrastructure, or road users' behaviour. The last-mentioned can be based on data from roadside surveys or from questionnaire surveys. However, results of such surveys are seldom comparable across countries due to differences in aims, scope, or methodology. This paper is based on the second edition of the E-Survey of Road Users' Attitudes (ESRA), an online survey carried out in 2018, and includes data from more than 35,000 road users across 32 countries. The objective is to present the main results of the ESRA survey regarding the four most important risky driving behaviours in traffic: driving under the influence (alcohol/drugs), speeding, mobile phone use while driving, and fatigued driving. The paper explores several aspects related to these behaviours as car driver, such as the self-declared behaviours, acceptability and risk perception, support for policy measures, and opinions on traffic rules and penalties. Results show that despite the high perception of risk and low acceptability of all the risky driving behaviours analysed, there is still a high percentage of car drivers who engage in risky behaviours in traffic in all the regions analysed. Speeding and the use of a mobile phone while driving were the most frequent self-declared behaviours. On the other hand, driving under the influence of alcohol or drugs was the least declared behaviour. Most respondents support policy measures to restrict risky behaviour in traffic and believe that traffic rules are not being checked regularly enough, and should be stricter. The ESRA survey proved to be a valuable source of information to understand the causes underlying road traffic crashes. It offers a unique database and provides policy makers and researchers with valuable insights into public perception of road safety

    The advances in hematopoietic cell transplantation (HCT) over the last decade have led to a transplant-related mortality below 15%. Hepatic sinusoidal obstruction syndrome/veno-occlusive disease (SOS/VOD) is a life-threatening complication of HCT that belongs to a group of diseases increasingly identified as transplant-related, systemic endothelial diseases. In most cases, SOS/VOD resolves within weeks; however, severe SOS/VOD results in multi-organ dysfunction/failure with a mortality rate &gt;80%. A timely diagnosis of SOS/VOD is of critical importance, given the availability of therapeutic options with favorable tolerability. Current diagnostic criteria are used for adults and children. However, over the last decade it has become clear that SOS/VOD is significantly different between the age groups in terms of incidence, genetic predisposition, clinical presentation, prevention, treatment and outcome. Improved understanding of SOS/VOD and the availability of effective treatment questions the use of the Baltimore and Seattle criteria for diagnosing SOS/VOD in children.     Efecto de dos temperaturas de dilución sobre la calidad de semen en ganado cebuino del Trópico Peruano

    The bull is a factor of genetic and economic importance in cattle breed, since the next generations will depend on it; therefore, the evaluation of the seminal quality and the cryopreservation of its genetic ma­terial are essential. The aim of the study was to evaluate the effect of two dilution temperatures on the quality of semen from zebuine cattle from the Peruvian Tropics. The samples were placed in a water bath at 34°C and the macroscopic and microscopic characteristics of semen were analyzed. Only the ejaculates with &gt;60% motility and &lt;20% abnormalities were processed, separating them into two treatments. T1 consisted of a 1:1 predilu­tion at 34°C, followed by a final dilution of 30 minutes by adding the dilutor at 34°C. T2 consisted of a 1:1 predilution at 34°C and after 30 minutes of stabilization, refrigeration was carried out together with the dilutor, in order to carry out the final dilution at 5°C. The seminal quality was analyzed after 16 hours of refrigeration. The percentage of vitality in T1 (80.89%) was greater than in T2 (71.22%) (p&lt;0.05), the positive reaction to the hypoosmotic problem was greater in T1 (71.11%) than in T2 (60%) (p&lt;0.05). Acrosomal integrity and morphometric features did not vary according to treatments. In conclusion, the initial and final dilution at 34°C in Gyr bulls positively influences the semen quality before the freezing of semen packets.El toro es un factor de importancia genética y económica en un hato ganadero, ya que de este dependerán las próximas generaciones; por lo tanto, es esencial la evaluación de la calidad seminal y la criopreservación de su material genético. El objetivo del estudio fue evaluar el efecto de dos temperaturas de dilución sobre la calidad de semen de ganado cebuino del Trópico Peruano. Dos toros Gyr fueron colectados semanalmente mediante electroeyaculación. Las muestras se colocaron en baño maría a 34°C y se analizaron las características macroscópicas y microscópicas del semen. Solo los eyaculados con &gt;60% de motilidad y &lt;20% de anormalidades fueron procesados, separándolos en dos tratamientos. El T1 consistió en predilución 1:1 a 34°C, luego de 30 minutos una dilución final agregando el dilutor a 34°C. El T2 consistió en predilución 1:1 a 34°C y a los 30 minutos de estabilización se llevó a refrigeración conjuntamente con el dilutor, para luego realizar la dilución final a 5°C. La calidad seminal fue analizada luego de 16 horas de refrigeración. El porcentaje de vitalidad en T1 (80,89%) fue mayor que en T2 (71,22%) (p&lt;0,05), la reacción positiva a la prueba hipoosmótica fue mayor en T1 (71,11%) que en T2 (60%) (p&lt;0,05). La integridad acrosomal y los rasgos morfométricos no variaron según tratamientos. En conclusión, la dilución inicial y final a 34°C en toros Gyr influye positivamente sobre la calidad seminal antes de la congelación de pajillas de semen

    Cloning, Expression, and Purification of Histidine-Tagged Escherichia coli Dihydrodipicolinate Reductase

DHDPR catalyzes the NAD(P)H dependent reduction of 2,3-dihydrodipicolinate to the cyclic imine L-2,3,4,5,-tetrahydropicolinic acid. The dapB gene that encodes dihydrodipicolinate reductase has previously been cloned, but the expression of the enzyme is low and the purification is time consuming. Therefore the E. coli dapB gene was cloned into the pET16b vector to improve the protein expression and simplify the purification. The dapB gene sequence was utilized to design forward and reverse oligonucleotide primers that were used to PCR the gene from Escherichia coli genomic DNA. The primers were designed with NdeI or BamHI restriction sites on the 5’and 3’ terminus respectively. The PCR product was sequenced to confirm the identity of dapB. The gene was cloned into the expression vector pET16b through NdeI and BamHI restriction endonuclease sites. The resulting plasmid containing dapB was transformed into the bacterial strain BL21 (DE3). The transformed cells were utilized to grow and express the histidine-tagged reductase and the protein was purified using Ni-NTA affinity chromatography. SDS/PAGE gel analysis has shown that the protein was 95% pure and has approximate subunit molecular weight of 28 kDa. The protein purification is completed in one day and 3 liters of culture produced approximately 40–50 mgs of protein, an improvement on the previous protein expression and multistep purification.Yeshttp://www.plosone.org/static/editorial#pee
