    Germination responses of a dry sclerophyll forest soil-stored seedbank to fire related cues

    Fire is an integral component of many ecosystems worldwide. Many plant species require fire-related cues, primarily heat and smoke, to trigger germination. Despite the importance of this process, the responses of many Australian species to these cues are unknown. Without this knowledge fire management strategies may be developed that are inappropriate for individual species and vegetation communities. In this study we examined the responses of a dry sclerophyll forest seed bank to heat and smoke germination cues. Analysis was possible for 48 taxa within the soil seedbank with 34 of these showing a response to one or both of the germination cues. 10 species responded to the heat treatment, 11 species responded to the smoke treatment and 13 species responded to both the heat and smoke treatments. Germination cues acted independently for all species considered. Results in this study were consistent with published reports for most species, although some differences were seen at the species and genus level. The study highlights the importance of fire-related cues in enhancing germination of a large proportion of the species occurring in dry sclerophyll forests

    Bacteria and the Aging and Longevity of Caenorhabditis elegans

    The molecular genetic analysis of longevity of Caenorhabditis elegans has yielded fundamental insights into evolutionarily conserved pathways and processes governing the physiology of aging. Recent studies suggest that interactions between C. elegans and its microbial environment may influence the aging and longevity of this simple host organism. Experimental evidence supports a role for bacteria in affecting longevity through distinct mechanisms—as a nutrient source, as a potential pathogen that induces double-edged innate immune and stress responses, and as a coevolved sensory stimulus that modulates neuronal signaling pathways regulating longevity.     Characterization of a molecular switch system that regulates gene expression in mammalian cells through a small molecule

    Abstract Background Molecular switch systems that activate gene expression by a small molecule are effective technologies that are widely used in applied biological research. Nuclear receptors are valuable candidates for these regulation systems due to their functional role as ligand activated transcription factors. Previously, our group engineered a variant of the retinoid × receptor to be responsive to the synthetic compound, LG335, but not responsive to its natural ligand, 9-cis-retinoic acid. Results This work focuses on characterizing a molecular switch system that quantitatively controls transgene expression. This system is composed of an orthogonal ligand/nuclear receptor pair, LG335 and GRQCIMFI, along with an artificial promoter controlling expression of a target transgene. GRQCIMFI is composed of the fusion of the DNA binding domain of the yeast transcription factor, Gal4, and a retinoid × receptor variant. The variant consists of the following mutations: Q275C, I310M, and F313I in the ligand binding domain. When introduced into mammalian cell culture, the switch shows luciferase activity at concentrations as low as 100 nM of LG335 with a 6.3 ± 1.7-fold induction ratio. The developed one-component system activates transgene expression when introduced transiently or virally. Conclusions We have successfully shown that this system can induce tightly controlled transgene expression and can be used for transient transfections or retroviral transductions in mammalian cell culture. Further characterization is needed for gene therapy applications.</p

    Regulation of MYC Expression and Differential JQ1 Sensitivity in Cancer Cells

    High level MYC expression is associated with almost all human cancers. JQ1, a chemical compound that inhibits MYC expression is therapeutically effective in preclinical animal models in midline carcinoma, and Burkitt’s lymphoma (BL). Here we show that JQ1 does not inhibit MYC expression to a similar extent in all tumor cells. The BL cells showed a ∼90% decrease in MYC transcription upon treatment with JQ1, however, no corresponding reduction was seen in several non-BL cells. Molecularly, these differences appear due to requirements of Brd4, the most active version of the Positive Transcription Elongation Factor B (P-TEFb) within the Super Elongation Complex (SEC), and transcription factors such as Gdown1, and MED26 and also other unknown cell specific factors. Our study demonstrates that the regulation of high levels of MYC expression in different cancer cells is driven by unique regulatory mechanisms and that such exclusive regulatory signatures in each cancer cells could be employed for targeted therapeutics

    Visual spatial memory is enhanced in female rats (but inhibited in males) by dietary soy phytoestrogens

    BACKGROUND: In learning and memory tasks, requiring visual spatial memory (VSM), males exhibit superior performance to females (a difference attributed to the hormonal influence of estrogen). This study examined the influence of phytoestrogens (estrogen-like plant compounds) on VSM, utilizing radial arm-maze methods to examine varying aspects of memory. Additionally, brain phytoestrogen, calbindin (CALB), and cyclooxygenase-2 (COX-2) levels were determined. RESULTS: Female rats receiving lifelong exposure to a high-phytoestrogen containing diet (Phyto-600) acquired the maze faster than females fed a phytoestrogen-free diet (Phyto-free); in males the opposite diet effect was identified. In a separate experiment, at 80 days-of-age, animals fed the Phyto-600 diet lifelong either remained on the Phyto-600 or were changed to the Phyto-free diet until 120 days-of-age. Following the diet change Phyto-600 females outperformed females switched to the Phyto-free diet, while in males the opposite diet effect was identified. Furthermore, males fed the Phyto-600 diet had significantly higher phytoestrogen concentrations in a number of brain regions (frontal cortex, amygdala & cerebellum); in frontal cortex, expression of CALB (a neuroprotective calcium-binding protein) decreased while COX-2 (an inducible inflammatory factor prevalent in Alzheimer's disease) increased. CONCLUSIONS: Results suggest that dietary phytoestrogens significantly sex-reversed the normal sexually dimorphic expression of VSM. Specifically, in tasks requiring the use of reference, but not working, memory, VSM was enhanced in females fed the Phyto-600 diet, whereas, in males VSM was inhibited by the same diet. These findings suggest that dietary soy derived phytoestrogens can influence learning and memory and alter the expression of proteins involved in neural protection and inflammation in rats

    Survival, density, and abundance of common bottlenose dolphins in Barataria Bay (USA) following the Deepwater Horizon oil spill

    To assess potential impacts of the Deepwater Horizon oil spill in April 2010, we conducted boat-based photo-identification surveys for common bottlenose dolphins Tursiops truncatus in Barataria Bay, Louisiana, USA (~230 km2, located 167 km WNW of the spill center). Crews logged 838 h of survey effort along pre-defined routes on 10 occasions between late June 2010 and early May 2014. We applied a previously unpublished spatial version of the robust design capture-recapture model to estimate survival and density. This model used photo locations to estimate density in the absence of study area boundaries and to separate mortality from permanent emigration. To estimate abundance, we applied density estimates to saltwater (salinity > ~8 ppt) areas of the bay where telemetry data suggested that dolphins reside. Annual dolphin survival varied between 0.80 and 0.85 (95% CIs varied from 0.77 to 0.90) over 3 yr following the Deepwater Horizon spill. In 2 non-oiled bays (in Florida and North Carolina), historic survival averages approximately 0.95. From June to November 2010, abundance increased from 1300 (95% CI ± ~130) to 3100 (95% CI ± ~400), then declined and remained between ~1600 and ~2400 individuals until spring 2013. In fall 2013 and spring 2014, abundance increased again to approximately 3100 individuals. Dolphin abundance prior to the spill was unknown, but we hypothesize that some dolphins moved out of the sampled area, probably northward into marshes, prior to initiation of our surveys in late June 2010, and later immigrated back into the sampled area.Publisher PDFPeer reviewe

    Polarised radio pulsations from a new T dwarf binary

    Brown dwarfs display Jupiter-like auroral phenomena such as magnetospheric Hα\alpha emission and coherent radio emission. Coherent radio emission is a probe of magnetospheric acceleration mechanisms and provides a direct measurement of the magnetic field strength at the emitter's location, both of which are difficult to access by other means. Observations of the coldest brown dwarfs (spectral types T and Y) are particularly interesting as their magnetospheric phenomena may be very similar to those in gas-giant exoplanets. Here we present 144 MHz radio and infrared adaptive optics observations of the brown dwarf WISEP J101905.63+652954.2 made using the LOFAR and Keck telescopes respectively. The radio data shows pulsed highly circularly polarised emission which yields a rotation rate of 0.32±0.030.32\pm0.03 hr1^{-1}. The infrared imaging reveals the source to be a binary with a projected separation of 423.0±1.6423.0\pm1.6 mas between components of spectral type T5.5±0.55\pm0.5 and T7.0±0.50\pm0.5. With a simple "toy model" we show that the radio emission can in principle be powered by the interaction between the two dwarfs with a mass-loss rates of at least 2525 times the Jovian value. WISEP J101905.63+652954.2 is interesting because it is the first pulsed methane dwarf detected in a low radio-frequency search. Unlike previous gigahertz-frequency searches that were only sensitive to objects with kiloGauss fields, our low-frequency search is sensitive to surface magnetic fields of 50\approx 50 Gauss and above which might reveal the coldest radio-loud objects down to planetary mass-scales.Comment: Accepted for publication in A&
