    Applications Of Monoclonal Antibodies To Studies Of Structure/function Relationships In Escherichia Coli F(1)-atpase

    Twenty-one monoclonal antibodies to Escherichia coli F{dollar}\sb 1{dollar}-ATPase were characterized and used in structural studies of the enzyme. Solid phase competition assays placed antibodies recognizing the same or overlapping epitopes into competition subgroups. Immunoprecipitation and membrane-binding studies identified those antibodies recognizing external epitopes on soluble and membrane-bound forms of the holoenzyme and demonstrated that the {dollar}\delta{dollar} and {dollar}\epsilon{dollar} subunits are substantially exposed in F{dollar}\sb 1{dollar}F{dollar}\sb 0{dollar}.;Analysis of partially cleaved {dollar}\alpha{dollar} subunit using Western blots mapped antibody epitopes to its carboxyl-half. Analysis of CNBr/weak acid cleaved {dollar}\beta{dollar} located the non-inhibitory B-II antibody epitopes between Asp-345 and Met-380 and the inhibitory B-I antibody epitopes between Asp-381 and the carboxyl terminus. The {dollar}\epsilon{dollar}-binding site on {dollar}\beta{dollar} was mapped to the same region as the B-I antibodies. The Fab fragment of a B-I antibody and {dollar}\epsilon{dollar} were cross-linked indicating the proximity of their binding sites in native F{dollar}\sb 1{dollar}.;Cross-linking studies revealed neither differences in quaternary structure between soluble and membrane-bound F{dollar}\sb 1{dollar} nor any association between {dollar}\delta{dollar} or {dollar}\epsilon{dollar} and any F{dollar}\sb 0{dollar} subunit. Cross-linking studies of F{dollar}\sb 1{dollar} in the presence of the ATPase activators ethylene glycol and antibody {dollar}\epsilon{dollar}-4 examined the mechanism of {dollar}\epsilon{dollar}-inhibition. Ethylene glycol displaced {dollar}\epsilon{dollar} only from its site of 1-ethyl-3- (3-(dimethylamino)propyl) carbodiimide (EDC) cross-linkage to {dollar}\beta{dollar} demonstrating that {dollar}\epsilon{dollar}-inhibition is mediated through {dollar}\beta{dollar}. In contrast, antibody {dollar}\epsilon{dollar}-4 did not alter EDC cross-linking of {dollar}\epsilon{dollar} to {dollar}\beta{dollar} showing that this interaction is not always inhibitory. {dollar}\epsilon{dollar} could also be cross-linked to {dollar}\beta{dollar} by EDC in F{dollar}\sb 1{dollar}F{dollar}\sb 0{dollar} where it is not inhibitory. The inhibitory effect of {dollar}\epsilon{dollar} and the B-I antibodies binding to the carboxyl-terminal region of {dollar}\beta{dollar} region suggested a function for this region.;The antibodies were used to characterize an {dollar}\alpha{dollar}-{dollar}\delta{dollar} cross-link produced during column centrifugation of F{dollar}\sb 1{dollar} by oxidation of sulfhydryls. This cross-link affected neither the enzyme\u27s activity nor susceptibility to {dollar}\epsilon{dollar}-inhibition, casting doubt on rotational models of F{dollar}\sb 1{dollar} catalysis. The {dollar}\alpha{dollar}-{dollar}\delta{dollar} cross-link also resulted in a loss of binding affinity between F{dollar}\sb 1{dollar} and F{dollar}\sb 0{dollar}

    A double-blind, placebo-controlled study of the short term effects of a spring water supplemented with magnesium bicarbonate on acid/base balance, bone metabolism and cardiovascular risk factors in postmenopausal women

    <p>Abstract</p> <p>Background</p> <p>A number of health benefits including improvements in acid/base balance, bone metabolism, and cardiovascular risk factors have been attributed to the intake of magnesium rich alkaline mineral water. This study was designed to investigate the effects of the regular consumption of magnesium bicarbonate supplemented spring water on pH, biochemical parameters of bone metabolism, lipid profile and blood pressure in postmenopausal women.</p> <p>Findings</p> <p>In this double-blind, placebo-controlled, parallel-group, study, 67 postmenopausal women were randomised to receive between 1500 mL and 1800 mL daily of magnesium bicarbonate supplemented spring water (650 mg/L bicarbonate, 120 mg/L magnesium, pH 8.3-8.5) (supplemented water group) or spring water without supplements (control water group) over 84 days. Over this period biomarkers of bone turnover (serum parathyroid hormone (PTH), 1,25-dihydroxyvitamin D, osteocalcin, urinary telopeptides and hydroxyproline), serum lipids (total cholesterol, HDL-cholesterol, LDL-cholesterol and triglycerides), venous and urinary pH were measured together with measurements of standard biochemistry, haematology and urine examinations.</p> <p>Serum magnesium concentrations and urinary pH in subjects consuming the magnesium bicarbonate supplemented water increased significantly at Day 84 compared to subjects consuming the spring water control (magnesium - p = 0.03; pH - p = 0.018). The consumption of spring water led to a trend for an increase in parathyroid hormone (PTH) concentrations while the PTH concentrations remained stable with the intake of the supplemented spring water. However there were no significant effects of magnesium bicarbonate supplementation in changes to biomarkers of bone mineral metabolism (n-telopeptides, hydroxyproline, osteocalcin and 1,25-dihydroxyvitamin D) or serum lipids or blood pressure in postmenopausal women from Day 0 to Day 84.</p> <p>Conclusions</p> <p>Short term regular ingestion of magnesium bicarbonate supplemented water provides a source of orally available magnesium. Long term clinical studies are required to investigate any health benefits.</p> <p>Trial registration</p> <p>ACTRN12609000863235</p

    A function-based typology for Earth’s ecosystems

    As the United Nations develops a post-2020 global biodiversity framework for the Convention on Biological Diversity, attention is focusing on how new goals and targets for ecosystem conservation might serve its vision of ‘living in harmony with nature’(1,2). Advancing dual imperatives to conserve biodiversity and sustain ecosystem services requires reliable and resilient generalizations and predictions about ecosystem responses to environmental change and management(3). Ecosystems vary in their biota(4), service provision(5) and relative exposure to risks(6), yet there is no globally consistent classification of ecosystems that reflects functional responses to change and management. This hampers progress on developing conservation targets and sustainability goals. Here we present the International Union for Conservation of Nature (IUCN) Global Ecosystem Typology, a conceptually robust, scalable, spatially explicit approach for generalizations and predictions about functions, biota, risks and management remedies across the entire biosphere. The outcome of a major cross-disciplinary collaboration, this novel framework places all of Earth’s ecosystems into a unifying theoretical context to guide the transformation of ecosystem policy and management from global to local scales. This new information infrastructure will support knowledge transfer for ecosystem-specific management and restoration, globally standardized ecosystem risk assessments, natural capital accounting and progress on the post-2020 global biodiversity framework

    Logging Affects Fledgling Sex Ratios and Baseline Corticosterone in a Forest Songbird

    Silviculture (logging) creates a disturbance to forested environments. The degree to which forests are modified depends on the logging prescription and forest stand characteristics. In this study we compared the effects of two methods of group-selection (“moderate” and “heavy”) silviculture (GSS) and undisturbed reference stands on stress and offspring sex ratios of a forest interior species, the Ovenbird (Seiurus aurocapilla), in Algonquin Provincial Park, Canada. Blood samples were taken from nestlings for corticosterone and molecular sexing. We found that logging creates a disturbance that is stressful for nestling Ovenbirds, as illustrated by elevated baseline corticosterone in cut sites. Ovenbirds nesting in undisturbed reference forest produce fewer male offspring per brood (proportion male = 30%) while logging with progressively greater forest disturbance, shifted the offspring sex ratio towards males (proportion male: moderate = 50%, heavy = 70%). If Ovenbirds in undisturbed forests usually produce female-biased broods, then the production of males as a result of logging may disrupt population viability. We recommend a broad examination of nestling sex ratios in response to anthropogenic disturbance to determine the generality of our findings

    Developing a core outcome set for fistulising perianal Crohn's disease

    OBJECTIVE: Lack of standardised outcomes hampers effective analysis and comparison of data when comparing treatments in fistulising perianal Crohn's disease (pCD). Development of a standardised set of outcomes would resolve these issues. This study provides the definitive core outcome set (COS) for fistulising pCD. DESIGN: Candidate outcomes were generated through a systematic review and patient interviews. Consensus was established via a three-round Delphi process using a 9-point Likert scale based on how important they felt it was in determining treatment success culminating in a final consensus meeting. Stakeholders were recruited nationally and grouped into three panels (surgeons and radiologists, gastroenterologists and IBD specialist nurses, and patients). Participants received feedback fromtheir panel(in the second round) andall participants(in the third round) to allow refinement of their scores. RESULTS: A total of 295 outcomes were identified from systematic reviews and interviews that were categorised into 92 domains. 187 stakeholders (response rate 78.5%) prioritised 49 outcomes through a three-round Delphi study.The final consensus meeting of 41 experts and patients generated agreement on an eight domain COS. The COS comprised three patient-reported outcome domains (quality of life, incontinence and a combined score of patient priorities) and five clinician-reported outcome domains (perianal disease activity, development of new perianal abscess/sepsis, new/recurrent fistula, unplanned surgery and faecal diversion). CONCLUSION: A fistulising pCD COS has been produced by all key stakeholders. Application of the COS will reduce heterogeneity in outcome reporting, thereby facilitating more meaningful comparisons between treatments, data synthesis and ultimately benefit patient care
