    Hypo- and Hyper-Virulent Listeria monocytogenes Clones Persisting in Two Different Food Processing Plants of Central Italy

    A total of 66 Listeria monocytogenes (Lm) isolated from 2013 to 2018 in a small-scale meat processing plant and a dairy facility of Central Italy were studied. Whole Genome Sequencing and bioinformatics analysis were used to assess the genetic relationships between the strains and investigate persistence and virulence abilities. The biofilm forming-ability was assessed in vitro. Cluster analysis grouped the Lm from the meat plant into three main clusters: two of them, both belonging to CC9, persisted for years in the plant and one (CC121) was isolated in the last year of sampling. In the dairy facility, all the strains grouped in a CC2 four-year persistent cluster. All the studied strains carried multidrug efflux-pumps genetic determinants (sugE, mdrl, lde, norM, mepA). CC121 also harbored the Tn6188 specific for tolerance to Benzalkonium Chloride. Only CC9 and CC121 carried a Stress Survival Islet and presented high-level cadmium resistance genes (cadA1C1) carried by different plasmids. They showed a greater biofilm production when compared with CC2. All the CC2 carried a full-length inlA while CC9 and CC121 presented a Premature Stop Codon mutation correlated with less virulence. The hypo-virulent clones CC9 and CC121 appeared the most adapted to food-processing environments; however, even the hyper-virulent clone CC2 warningly persisted for a long time. The identification of the main mechanisms promoting Lm persistence in a specific food processing plant is important to provide recommendations to Food Business Operators (FBOs) in order to remove or reduce resident Lm

    Supplemental material related to the Fermi LAT third source catalog (3FGL) (Acero et al, 2015)The gamma-ray results used in this paper were derived in the context of the 3FGL catalog (Fermi-LAT Collaboration 2015, J/ApJS/218/23). No additional analysis of the gamma-ray data was performed in the context of the present paper except for the fitting of the monthly light curves and the broadband SED fitting. The data were collected over the first 48 months of the mission, from 2008 August 4 to 2012 July 31 (MJD 54682 to 56139). Using the Australia Telescope Compact Array (ATCA) at 5 GHz and 9 GHz, Petrov et al. (2013, J/MNRAS/432/1294) detected 424 sources in the LAT error ellipses of southern unassociated 2FGL sources.     Externally Fired Micro-Gas Turbine and Organic Rankine Cycle Bottoming Cycle: Optimal Biomass/Natural Gas Combined Heat and Power Generation Configuration for Residential Energy Demand

    Small scale combined heat and power (CHP) plants present lower electric efficiency in comparison to large scale ones, and this is particularly true when biomass fuels are used. In most cases, the use of both heat and electricity to serve on-site energy demand is a key issue to achieve acceptable global energy efficiency and investment profitability. However, the heat demand follows a typical daily and seasonal pattern and is influenced by climatic conditions, in particular in the case of residential and tertiary end users. During low heat demand periods, a lot of heat produced by the CHP plant is discharged. In order to increase the electric conversion efficiency of small scale micro-gas turbine for heat and power cogeneration, a bottoming organic Rankine cycle (ORC) system can be coupled to the cycle, however, this option reduces the temperature and the amount of cogenerated heat available to the thermal load. In this perspective, the paper presents the results of a thermo-economic analysis of small scale CHP plants composed of a micro-gas turbine (MGT) and a bottoming ORC, serving a typical residential energy demand. For the topping cycle, three different configurations are examined: (1) a simple recuperative micro-gas turbine fuelled by natural gas (NG); (2) a dual fuel externally fired gas turbine (EFGT) cycle, fuelled by biomass and natural gas (50% share of energy input) (DF); and (3) an externally fired gas turbine (EFGT) with direct combustion of biomass (B). The bottoming ORC is a simple saturated cycle with regeneration and no superheating. The ORC cycle and the fluid selection are optimized on the basis of the available exhaust gas temperature at the turbine exit. The research assesses the influence of the thermal energy demand typology (residential demand with cold, mild, and hot climate conditions) and CHP plant operational strategies (baseload versus heat-driven versus electricity-driven operation mode) on the global energy efficiency and profitability of the following three configurations: (A) MGT with cogeneration; (B) MGTþ ORC without cogeneration; and (C) MGTþORC with cogeneration. In all cases, a back-up boiler is assumed to match the heat demand of the load (fed by natural gas or biomass). The research explores the profitability of bottoming ORC in view of the following trade-offs: (i) lower energy conversion efficiency and higher investment cost of biomass input with respect to natural gas; (ii) higher efficiency but higher costs and reduced heat available for cogeneration with the bottoming ORC; and (iii) higher primary energy savings and revenues from feed-in tariff available for biomass electricity fed into the gri

    Detection of Vibrio splendidus and related species in Chamelea gallina sampled in the Adriatic along the Abruzzi coastline

    Vibrio species are an important and widespread component of marine microbial communities. Some Vibrio strains are potentially pathogenic to marine vertebrates and invertebrates. The aim of this study was to identify vibrios, in particular Vibrio splendidus and related species, isolated from clams (Chamelea gallina) collected along the coasts of the Abruzzi region from May to October 2007. The isolates obtained were phenotyped and classified as belonging to the genus Vibrio. The strains underwent biochemical testing in accordance with Alsina’s scheme for V. splendidus identification. Molecular analysis of the 16S-23S intergenic space region and recA gene was used to identify V. splendidus and related species. All the samples examined were found to contain halophylic Vibrio species, with V. alginolyticus, V. splendidus-related species and V. mediterranei most commonly found. A polymerase chain reaction of the 16S-23S intergenic space region and sequencing of the recA gene from isolates confirmed that phenotyping of Vibrio species is not sufficient to distinguish between different species. Differentiation of the highly related species among V. splendidus-related clusters remains an important issue. In this regard, our data suggests sequencing the recA genes was far more discriminatory than sequencing 16S rDNA for this purpose

    Autoantibodies to the GM2-1 islet ganglioside and to GAD-65 at type 1 diabetes onset

    The GM2-1 islet ganglioside has been sequenced, found to be a novel ganglioside structure with a sialic acid moiety in the terminal position and two residues of non-acetylated galactosamine and also shown to be a target of autoantibodies in a subset of ICA(+) relatives of type 1 diabetic patients who subsequently progressed to the overt disease. In the present study we determined whether antibodies to GM2-1 or to other pancreatic gangliosides (a) are also expressed at disease onset and (b) are correlated with other diabetes-associated autoantibodies. Pancreatic gangliosides were extracted from human pancreas and purified by thin layer chromatography (TLC). Anti-ganglioside autoantibodies were determined using an indirect immunoperoxidase technique performed directly on TLC plates in the following groups of patients: (a) newly diagnosed type 1 diabetic subjects before insulin therapy (n=45); all were tested for GAD65 autoantibodies in a fluid-phase RIA using S-35-methionine-labelled recombinant human GAD65. Of these patients, 24 were also tested for insulin autoantibodies (IAA) by a competitive fluid phase radioimmunoassay and 21 were tested for GAD67 reactivity. (b) Forty-two age-and sex-matched normal control subjects. Autoantibodies to GM2-1, but not to other pancreatic gangliosides (GM3, GD3, GD1a), were expressed in 31 of 45 new-onset type 1 diabetic subjects and in one of 42 normal controls (P<0.01), while anti-GAD65, IAA and anti-GAD67 were found in 31 of 45, 12 of 24 and three of 21 patients respectively, but not in the control group of subjects. Interestingly, occurrence of GM2-1 autoantibodies was significantly correlated (P<0.005) with positivity for GAD65 autoantibodies, but not for IAA or GAD67 autoantibodies. It is of note that both GAD and gangliosides are mainly expressed in islets and in neuronal tissues and, therefore, type 1 diabetes may be regarded as a neuroendocrine autoimmune disease.     Listeria monocytogenes (Lm) is the causative agent of listeriosis, an invasive disease primarily affecting immunocompromised people, the elderly, children and pregnant women, with high hospitalization (98.6%) and fatality rates (13.8%) 1. The disease is most commonly caused by eating contaminated food, in particular ready-to-eat. The ability of some strains to persist, even for years, in food processing environments can increase the risk of food contamination. Persistence can results from Lm survival after disinfection, thanks to protective biofilm formation, disinfectants and stresses resistance mechanisms or from the repeated reintroduction from raw materials 2 3. The identification of recurring highly genetically related isolates (Whole Genome Sequencing, WGS and core genome MLST, cgMLST) is necessary to define a strain persistent in a plant 4. The aim of this study was to evaluate persistence and resistance to commercial sanitizers commonly used in food processing environments, in Lm strains isolated within the laboratory activity of IZSUM (Istituto Zooprofilattico Sperimentale Umbria e Marche).     A Real-Time PCR Screening Assay for Rapid Detection of Listeria Monocytogenes Outbreak Strains

    From January 2015 to March 2016, an outbreak of 23 human cases of listeriosis in the Marche region and one human case in the Umbria region of Italy was caused by Listeria monocytogenes strains showing a new pulsotype never described before in Italy. A total of 37 clinical strains isolated from patients exhibiting listeriosis symptoms and 1374 strains correlated to the outbreak were received by the Italian National Reference Laboratory for L. monocytogenes (It NRL Lm) of Istituto Zooprofilattico Sperimentale dell&rsquo;Abruzzo e del Molise (IZSAM) for outbreak investigation. A real-time PCR assay was purposely designed for a rapid screening of the strains related to the outbreak. PCR-positive strains were successively typed through molecular serogrouping, pulsed field gel electrophoresis (PFGE), and Next Generation Sequencing (NGS). Applying the described strategy, based on real-time PCR screening, we were able to considerably reduce time and costs during the outbreak investigation activities
