    Local and global spontaneous calcium events regulate neurite outgrowth and onset of GABAergic phenotype during neural precursor differentiation

    Neural stem cells can generate in vitro progenitors of the three main cell lineages found in the CNS. The signaling pathways underlying the acquisition of differentiated phenotypes in these cells are poorly understood. Here we tested the hypothesis that Ca2+ signaling controls differentiation of neural precursors. We found low-frequency global and local Ca2+ transients occurring predominantly during early stages of differentiation. Spontaneous Ca2+ signals in individual precursors were not synchronized with Ca2+ transients in surrounding cells. Experimentally induced changes in the frequency of local Ca2+signals and global Ca2+ rises correlated positively with neurite outgrowth and the onset of GABAergic neurotransmitter phenotype, respectively. NMDA receptor activity was critical for alterations in neuronal morphology but not for the timing of the acquisition of the neurotransmitter phenotype. Thus, spontaneous Ca2+ signals are an intrinsic property of differentiating neurosphere-derived precursors. Their frequency may specify neuronal morphology and acquisition of neurotransmitter phenotype

    Stronger Together: Tiny-housers’ Views of Community

    Over the last decade, tiny homes (generally described as homes smaller than 500 ft2) have grown in popularity thanks to recent representation in the media (Ford and Gomez‐Lanier 2017). An increasing number of people have begun to adopt this reimagined way of living, and some tiny-housers have sought to live near other tiny-housers, forming tiny house communities (Kilman 2016, Mangold et al. Forthcoming). Little systematic research focusing on the tiny-housers views of community exists. To provide initial insights, 30 interviews were conducted with people at various stages in their tiny house journeys. The 30 interviews were transcribed and analyzed in Nvivo 11. Preliminary findings suggest that many of these tiny-housers want to redefine the normative American concept of community by rejecting sub-urban frameworks and borrowing elements from both strong place and interest-based community models. They seek to accomplish this vision through one or more of the following: (i) increased community participation, (ii) shared spaces and resources, and (iii) development of significant relationships with neighbors rooted in mutual support and frequent interaction. While participants strive for this type of community, they also insist upon a clear recognition of personal space and boundaries. Participants also express an interest in having diverse communities, yet desire to maintain many common interests and goals. These community elements are not entirely unique to the tiny house movement. The small space, however, both encourages and facilitates this kind of community participation.     Self-care coping strategies in people with diabetes: a qualitative exploratory study

    <p>Abstract</p> <p>Background</p> <p>The management of diabetes self-care is largely the responsibility of the patient. With more emphasis on the prevention of complications, adherence to diabetes self-care regimens can be difficult. Diabetes self-care requires the patient to make many dietary and lifestyle changes. This study will explore patient perceptions of diabetes self-care, with particular reference to the burden of self-care and coping strategies among patients.</p> <p>Methods</p> <p>A maximum variation sample of 17 patients was selected from GP practices and diabetes clinics in Ireland to include patients with types 1 and 2 diabetes, various self-care regimens, and a range of diabetes complications. Data were collected by in-depth interviews; which were tape-recorded and transcribed. The transcripts were analysed using open and axial coding procedures to identify main categories, and were reviewed by an independent corroborator. Discussion of the results is made in the theoretical context of the health belief, health value, self-efficacy, and locus of control frameworks.</p> <p>Results</p> <p>Patients' perceptions of their self-care varied on a spectrum, displaying differences in self-care responsibilities such as competence with dietary planning, testing blood sugar and regular exercise. Three patient types could be distinguished, which were labeled: "proactive manager," a patient who independently monitors blood glucose and adjusts his/her self-care regime to maintain metabolic control; "passive follower," a patient who follows his/her prescribed self-care regime, but does not react autonomously to changes in metabolic control; and "nonconformist," a patient who does not follow most of his/her prescribed self-care regimen.</p> <p>Conclusion</p> <p>Patients have different diabetes self-care coping strategies which are influenced by their self-care health value and consequently may affect their diet and exercise choices, frequency of blood glucose monitoring, and compliance with prescribed medication regimens. Particular attention should be paid to the patient's self-care coping strategy, and self-care protocols should be tailored to complement the different patient types.</p

    Robot Mapping and Localisation for Feature Sparse Water Pipes Using Voids as Landmarks

    Robotic systems for water pipe inspection do not generally include navigation components for mapping the pipe network and locating damage. Such navigation systems would be highly advantageous for water companies because it would allow them to more effectively target maintenance and reduce costs. In water pipes, a major challenge for robot navigation is feature sparsity. In order to address this problem, a novel approach for robot navigation in water pipes is developed here, which uses a new type of landmark feature - voids outside the pipe wall, sensed by ultrasonic scanning. The method was successfully demonstrated in a laboratory environment and showed for the first time the potential of using voids for robot navigation in water pipes

    Enzymes produced by psychrophilic organisms have successfully overcome the low temperature challenge and evolved to maintain high catalytic rates in their permanently cold environments. As an initial step in our attempt to elucidate the cold-adaptation strategies used by these enzymes we report here the 1H, 15N and 13C assignments for the reduced form of a thiol-disulphide oxidoreductase from the Antarctic bacterium Pseudoalteromonas haloplanktis TAC125.The NMR spectrometers are part of The National NMR Network (REDE/1517/RMN/2005), supported by ‘‘Programa Operacional Ciência e Inovação (POCTI) 2010’’ and Fundação para a Ciência e a Tecnologia (FCT).     PipeSLAM: Simultaneous Localisation and Mapping in Feature Sparse Water Pipes using the Rao-Blackwellised Particle Filter

    Water, a valuable resource, is usually distributed through urban environments by buried pipes. These pipes are difficult to access for inspection, maintenance and repair. This makes in-pipe robots an appealing technology for inspecting water pipes and localising damage prior to repair from above ground. Accurate localisation of damage is of critical importance because of the costs associated with excavating roads, disrupting traffic and disrupting the water supply. The problem is that pipes tend to be relatively featureless making robot localisation a challenging problem. In this paper we propose a novel simultaneous localisation and mapping (SLAM) algorithm for metal water pipes. The approach we take is to excite pipe vibration with a hydrophone (sound induced vibration), which leads to a map of pipe vibration amplitude over space. We then develop a SLAM algorithm that makes use of this new type of map, where the estimation method is based on the Rao-Blackwellised particle filter (RBPF), termed PipeSLAM. The approach is also suited to SLAM in plastic water pipes using a similar type of map derived from ultrasonic sensing. We successfully demonstrate the feasibility of the approach using a combination of experimental and simulation data

    Major practicum as a learning site for exercise science professionals: A pilot study

    Exercise science is now an integral part of the allied health framework in Australia and graduates from accredited programmes are equipped with skills recognised as being important in the prevention and management of lifestyle-related diseases. This pilot study sought to determine the experiences of 11 final-year exercise science students in their major practicum and identify skills learned and developed while on placement. Analysis of the interview data established that the students worked with clients from a broad range of sociocultural and socioeconomic backgrounds, both within and between practicum sites; the students' experiences and their preparedness to engage with clients from different backgrounds varied as a result. Although the students generally reported being technically skilled for their major placement, many reported being underprepared to deal with people from different backgrounds. However, all participants held that their interpersonal skills greatly improved in response to their placement and several remarked that they developed their problem-solving skills through watching and assisting their supervisors work with clients. The present study confirms the practicum as a critical learning site for improving communication and problem-solving skills with exercise science and exercise physiology students
