    Indoor-Breeding of Aedes albopictus in Northern Peninsular Malaysia and Its Potential Epidemiological Implications

    Background: The mosquito Ae. albopictus is usually adapted to the peri-domestic environment and typically breeds outdoors. However, we observed its larvae in most containers within homes in northern peninsular Malaysia. To anticipate the epidemiological implications of this indoor-breeding, we assessed some fitness traits affecting vectorial capacity during colonization process. Specifically, we examined whether Ae. albopictus exhibits increased survival, gonotrophic activity and fecundity due to the potential increase in blood feeding opportunities. Methodology/Principal Findings: In a series of experiments involving outdoors and indoors breeding populations, we found that Ae. albopictus lives longer in the indoor environment. We also observed increased nighttime biting activity and lifetime fecundity in indoor/domestic adapted females, although they were similar to recently colonized females in body size. Conclusion/Significance: Taken together these data suggest that accommodation of Ae. albopictus to indoor/domestic environment may increase its lifespan, blood feeding success, nuisance and thus vectorial capacity (both in terms of increased vector-host contacts and vector population density).     Insecticide susceptibility of the dengue vector Aedes aegypti (Diptera: culicidae) in Makkah City, Saudi Arabia

    Objective To examine the insecticide susceptibility of Aedes aegypti (Ae. aegypti) from various sites in Makkah City, Saudi Arabia. Methods This was examined based on WHO standard procedures. Results The larvae of Ae. aegypti were susceptible to all larvicides examined, but this susceptibility was more pronounced in wild populations, which tended to show tolerance to icon. Icon was the most effective larvicide with LC50 values of 0.007 ppm and 0.012 ppm for the laboratory and field strains, respectively. Ae. aegypti adults exposed to lambda-cyhalothrin showed a low mortality rate in comparison with those exposed to deltamethrin and cyfluthrin. Conclusions The results of the present study indicate differential susceptibility between field and laboratory larval populations. Wild larvae are less susceptible to insecticide treatments than their laboratory-bred counterparts. Taken together, these results suggest that tolerance and the tendency toward resistance to commonly used insecticides are present in Ae. aegypti populations throughout Makkah City, Saudi Arabia

    Olfactory behavior and response of household ants (Hymenoptera) to different types of coffee odor: A coffee-based bait development prospect

    Odor sensation is a sensory modality of considerable significance in the foraging behavior and interactional organization of ants. In the food bait technology, smell is the basis of attraction, which, in turn, is the line of bait use and a key parameter for judging efficacy. Yet, the currently available baits possess low attractiveness to many ant pests. Hence, strategies to produce ant bait with increased attractiveness are needed. Despite evidence that coffee has a diverse aroma complex that affects the behavior of honey bees and ants, its attraction to house-invading ants has yet to be investigated. In a series of Y-tube olfactometer bioassays, we examined the behavioral responses of Tapinoma indicum (TI), Monomorium pharaonis (MP) and Solenopsis geminata (SG) to various coffee-induced odor stimuli, comprised of extracts from Arabica, Robusta and Liberica. All coffee extracts showed an influence on the behavior of TI, MP and SG workers, with Arabica showed the most significant influence to the tested ants. The workers of TI, MP and SG were more attracted to the odor of 0.01% Arabica extract (ONE), in comparison with 0.05% Arabica extract (TWO) or 0.10% Arabica extract (THREE). Arabica extract mixed with sugar (S) elicited a significant attraction from workers of all three species in a balanced competition with either unsweetened Arabica extract or water. These results indicated that coffee, particularly Arabica, was attractive to the foragers of TI, MP and SG, thus, the use of coffee as a novel stimulus agent seems plausible in ant bait development. © 201

    Fluorescence can be used to trace the fate of exogenous micro-organisms inside the alimentary tract of mosquitoes

    There is a great deal of current research interest in utilising bacteria for the control of intractable arthropod-borne diseases such as dengue. Although there is accumulating evidence that bacterial infection is a promising control strategy, most studies on bacteria-insect interactions lacked useful markers for detecting pathogenesis. This provided the impetus to investigate bacterial infection in the dengue vector Aedes albopictus. The infection persistence patterns in key organs of the alimentary canal of females were examined using a GFP-expressing strain of Escherichia coli (Migula). Just after feeding with sugar meal containing the bacteria, the crop and midgut as well as parts of the Malpighian tubules showed fluorescence. From 1 h onwards, bacterial populations declined sharply in both the midgut and crop, with complete elimination in the former but persistence of bacteria at 7 h post-feeding in the latter. After 24 h, neither organ retained the fluorescent marker. However, culture of homogenates of these organs in Luria-Bertani medium revealed the presence of a bacterial population in the crop, but not in the midgut. These observations suggest a difference in the potential hysiological actions expressible by the two organs. In fact, both are storage sites for ingested fluids, but the midgut has greater physiological activity. Presumably, one of these activities contributed to eliminating GFPexpressing E. coli from the A. albopictus midgut after 24 h. The results of the present study using a fluorescent marker to detect infection may be useful for developing strategies to fully characterise the main steps involved in the bacterial infection process in insects

    The effects of simulated rainfall on immature population dynamics of Aedes albopictus and female oviposition

    Larvae of Aedes albopictus Skuse typically inhabit natural and artificial containers. Since these larval habitats are replenished by rainfall, Ae. albopictus may experience increased loss of immature stages in areas with high levels of rainfall. In this study, we investigated the effects of rainfall and container water level on habitat quality, population density, and oviposition activity of Ae. albopictus. In field and laboratory experiments, we found that rainfall resulted in the flushing of breeding habitats. Excess rain negatively impacted larval and pupal retention, especially in small habitats. When filled with water to overflowing, container habitats were significantly repellent to ovipositing females.     Background: Mosquito-borne viruses are transmitted to human hosts via blood-feeding behavior of female mosquitoes. Female mosquitoes seek a host to take blood meals (host-seeking behavior). In order to prevent virus infections, it is important to understand how they modulate host-seeking behavior. Dopamine (DA) in the central nervous system acts as a neuromediator that regulates a variety of behaviors in insects. In female mosquitoes, host-seeking behavior increases when DA levels in the head decline after emergence. However, it remains unclear whether DA directly modulates host-seeking behavior in female mosquitoes. The aim of this study was to examine whether changes in DA levels in the head affects host-seeking activity in the adult female mosquito Aedes albopictus (Ae. albopictus). Findings: We compared host-seeking behavior in one group of emerging female adults treated with L-β-3,4-dihydroxyphenylalanine (L-DOPA), the precursor of DA, (L-DOPA group), with that in an untreated control (control group) after confirming elevation of head DA in L-DOPA group by using high-performance liquid chromatography. The content of head DA in L-DOPA group significantly remained higher than that in controls on all days examined. The host-seeking activity in the control group showed a gradual increase over the 6-day experimental period. In contrast, there was no such increase in the host-seeking activity in the L-DOPA group. Therefore, the host-seeking activity of L-DOPA group was significantly lower than that of the controls between day 3 and 6 post-emergence. Conclusion: Our results indicate that elevation of DA level reduces host-seeking activity in adult female mosquito Ae. albopictus

    Morphological and molecular characterization of fungus isolated from tropical bed bugs in Northern Peninsular Malaysia, Cimex hemipterus (Hemiptera: Cimicidae)

    Objective: To investigate some morphological and molecular characteristics of fungal parasites isolated from wild tropical bed bug, Cimex hemipterus. Methods: A series of culture methods were used to obtain fungal isolates from field-collected bed bugs. Characteristics of the isolates such as colony appearance, mycelial texture and pigmen-tation were studied to explore their morphology. Isolates were also subjected to a PCRbased genotyping test. Results: There were noticeable differences in morphological characteristics among the four isolates. Conidial areas of one isolate were dark green, whereas those of the remaining colonies were olive-green, black or dark brown. Conidia of the dark green isolate were globose, while those of olive-green, black and dark brown isolates were globose to subglobose, globose to spherical and globose to subglobose/finely roughened, respectively. These morphological specificities and the molecular analyses showed that the fungal internal transcribed spacer ribosomal region and β-tubulin gene sequences of the isolates shared clade with Trichoderma and Aspergillus sequences. Conclusions: Overall, the new discovery of common pathogens in agricultural field developed in live bed bugs storage tank may initiate the use of biological agents in later years

    Differential Protein Contents in Two Members of the Whitefly Bemisia tabaci (Hemiptera: Aleurodidae) Complex: Reproductive and Invasive Implications

    Although the displacement of some native whitefly species by the B biotype of Bemisia tabaci has been noted in China, it is still unclear which physiological and molecular mechanisms predominate during such invasions. Here, we investigated proteome variability in both B biotype and the native ZHJ1 haplotype. ZHJ1 eggs showed larger numbers of specific proteins than B eggs. A set of proteins found in B eggs were present at higher levels in ZHJ1. Three ZHJ1 egg proteins were present at higher levels in B eggs. ZHJ1 males possessed four specific proteins, and at least five other male proteins were shared by the two whiteflies. Two proteins identified in ZHJ1 males were present at higher levels in B males. Three other shared male proteins were found at much higher levels in ZHJ1 than in the B biotype. Both ZHJ1 and B expressed three specific female proteins. Most of the female proteins identified in ZHJ1 were present at much higher levels in B. In conclusion, protein content indexed by electrophoretic profiling has likely played an important role during the displacement of natives by the B biotype

    Coffee and its waste repel gravid Aedes albopictus females and inhibit the development of their embryos

    Background Dengue is a prevalent arboviral disease and the development of insecticide resistance among its vectors impedes endeavors to control it. Coffee is drunk by millions of people daily worldwide, which is associated with the discarding of large amounts of waste. Coffee and its waste contain large amounts of chemicals many of which are highly toxic and none of which have a history of resistance in mosquitoes. Once in solution, coffee is brownish in colour, resembling leaf infusion, which is highly attractive to gravid mosquitoes. To anticipate the environmental issues related to the increasing popularity of coffee as a drink, and also to combat insecticide resistance, we explored the deterrence potentials of coffee leachates against the ovipositing and embryonic stages of the dengue vector, Aedes albopictus. Methods In a series of choice, no-choice, and embryo toxicity bioassays, we examined changes in the ovipositional behaviours and larval eclosion of Ae. albopictus in response to coffee extracts at different concentrations. Results Oviposition responses were extremely low when ovicups holding highly concentrated extract (HCE) of coffee were the only oviposition sites. Gravid females retained increased numbers of mature eggs until 5 days post-blood feeding. When provided an opportunity to oviposit in cups containing coffee extracts and with water, egg deposition occurred at lower rates in those containing coffee, and HCE cups were far less attractive to females than those containing water only. Females that successfully developed in a coffee environment preferentially oviposited in such cups when in competition with preferred oviposition sites (water cups), but this trait did not continue into the fourth generation. Larval eclosion occurred at lower rates among eggs that matured in a coffee environment, especially among those that were maintained on HCE-moistened substrates. Conclusions The observations of the present study indicate a pronounced vulnerability of Ae. albopictus to the presence of coffee in its habitats during the early phases of its life cycle.
