    Reproductive strategies and natural history of the arboreal Neotropical vesper mouse, Nyctomys sumichrasti

    The vesper mouse, Nyctomys sumichrasti (Rodentia: Cricetidae), a poorly known, arboreal, nocturnal, rodent found in Central America, has been considered rare, and limited information is available about its biology, especially reproduction. We obtained vesper mice from several sites in Costa Rica and prepared them as scientific specimens with standard external measurements and reproductive data recorded. We expanded our dataset by including specimens from museum collections. Vesper mice have 1:1 sex ratios throughout their range. They reproduce year-round and litters may occur in rapid succession. Testis size is positively correlated with body mass, but we could not determine onset of sperm production. The onset of reproduction for females, based on mass, is variable but most females are parous by 60 g. Litter size is small with a mode of 2 and embryos are not evenly distributed across the uterine horns. Vesper mice exhibit striking morphological differences in terms of tooth-wear and pelage patterns across their range. Much work is needed to fully appreciate N. sumichrasti, its role in tropical habitats, and its reproductive biology. Resumen El ratón, Nyctomys sumichrasti (Rodentia: Cricetidae), es una especie de roedor arborícola y nocturna de América Central, que se considera raro y sobre el cual se tiene muy poca información acerca de su biología, particularmente acerca de su reproducción. Nosotros obtuvimos ratones en varias localidades en Costa Rica, los preparamos como especímenes científicos y tomamos datos morfológicos y de su reproducción. El conjunto de datos se amplió utilizando especímenes de museos. El ratón mantiene una proporción de sexos de 1:1 a lo largo de su distribución. Esta especie se reproduce durante todo el año y las camadas pueden tener lugar en una rápida sucesión. El tamaño de los testículos se correlacionó positivamente con la masa corporal, pero no se pudo determinar el comienzo de la producción de esperma. El inicio de reproducción en las hembras, basado en la masa corporal, es variable, pero para la mayoría de ellas comienza a los 60 g. El tamaño de la camada es pequeño, generalmente 2, y los embriones no se distribuyen uniformemente a través de los cuernos uterinos. El ratón muestra sorprendentes diferencias morfológicas en términos de patrones de desgaste de la dentadura y el pelaje a través de toda su distribución. Se necesita mucha más investigación para entender mejor ésta especie, su papel en hábitats tropicales y su biología reproductiva

    Genetic relationships of Caribbean lowland spiny pocket mice (Heteromys desmarestianus: Rodentia; Heteromyidae): evidence of a distinct mitochondrial lineage

    Genetic studies provide important insights into the evolutionary history and taxonomy of species, allowing us to identify lineages dif-ficult to distinguish morphologically. The relationships among species in the genus Heteromys have been in flux as new species have been described, and candidate species have been suggested in the H. desmarestianus group. One new potential species may be in Costa Rica’s Carib-bean lowlands. Herein, we test the phylogenetic relationships of individuals from Costa Rica’s Caribbean lowlands to individuals from through-out the species’ range using mitochondrial sequences from cytochrome-b (cytb). We captured 116 individuals from the lowlands, sequenced their cytb gene, and incorporated 74 GenBank sequences from throughout the species’ range to test if individuals from Costa Rica’s Caribbean lowlands potentially constitute an undescribed species. Our results document a distinct mitochondrial lineage in the Caribbean lowlands of Costa Rica. Our results from extensive sampling within the lowlands show a unique mitochondrial DNA lineage, which suggests the presence of an undescribed species. The Caribbean lowlands of Costa Rica may hold other cryptic diversity, and further phylogenetic studies should incorporate samples from this area, as it may have a unique evolutionary history. Los estudios genéticos proporcionan información importante sobre la historia evolutiva y la taxonomía de las especies, lo que nos permite identificar linajes difíciles de distinguir morfológicamente. Las relaciones filogenéticas entre las especies del género Heteromys han estado cambiando a medida que se han descrito nuevas especies y se han sugerido especies candidatas en el grupo H. desmarestianus. Una nueva especie potencial podría encontrarse en las tierras bajas del Caribe de Costa Rica. En este trabajo analizamos las relaciones filogenéticas entre individuos de las tierras bajas del Caribe de Costa Rica con individuos de todo el rango de la especie utilizando secuencias mitocondriales del citocromo-b (cytb). Capturamos 116 individuos de las tierras bajas, secuenciamos su gen cytb e incorporamos 74 secuencias GenBank de todo el área de distribución de la especie para probar si los individuos de las tierras bajas del Caribe de Costa Rica constituyen potencialmente una especie no descrita. Nuestros resultados indican la presencia de un linaje distinto basado en el ADN mitocondrial, que sugiere que los indivi-duos de las tierras bajas del Caribe de Costa Rica probablemente son una especie distinta. Las tierras bajas del Caribe de Costa Rica pueden tener una diversidad críptica significativa. Por ello sugerimos que estudios filogenéticos adicionales deberían incorporar muestras de esta área, ya que puede tener una historia evolutiva única

    Group dynamics, behavior, and current and historical abundance of peccaries in Costa Rica’s Caribbean lowlands

    The abundances and habitat preferences of peccaries in Neotropical forests are important to understand because these keystone species influence many aspects of the ecosystem. In the Caribbean lowlands of Costa Rica, we conducted walking surveys for ~2 years to study the behavior and population trends of collared peccaries (Pecari tajacu), and found that peccaries are abundant at La Selva Biological Station and overall, detection rates were relatively constant through time. A stable estimate of detection rates was achieved only after 7–9 months of surveying. We found no habitat preferences between primary and secondary forest, yet there were some differences in group dynamics—group radius was larger and sighting distance was greater in primary forest, whereas the number of singletons was higher in secondary forest. More peccaries were seen closer to the laboratory clearing than at greater distances, for a variety of probable reasons: habituation to humans, lower predation and hunting pressure, and various environmental and habitat factors. Peccary groups had spatially clumped distributions across the landscape and were more active diurnally than nocturnally. Collared peccary densities are relatively high at La Selva compared to other Neotropical sites, with the exception of Barro Colorado Island. We combined our data with a review of the historical literature to assess changes in the populations of peccaries in the Caribbean lowlands. We found that collared peccaries have likely increased in abundance at La Selva, seemingly a few years after the extirpation of white-lipped peccaries (Tayassu pecari), which were abundant in the area 40–50 years ago. An understanding of the group dynamics, behavior, and habitat preference of collared peccaries is essential for management decisions and conservation efforts. Additionally, assessment of population changes should be carefully considered in a historical context, with a particular focus on how the populations of the 2 peccary species have changed, and how these species might differentially affect their environment. Resumen--Entender la abundancia y la preferencia de hábitat de las 2 especies de sainos en bosques neotropicales es importante porque estas especies clave afectan muchos aspectos del ecosistema. En las tierras bajas del Caribe costarricense, llevamos a cabo muestreos a pie durante ~2 años para estudiar el comportamiento y tendencias poblacionales del saino (Pecari tajacu), y encontramos que son abundantes en la Estación Biológica La Selva y las tasas de detección fueron relativamente constantes a través del tiempo. Se obtuvo una tasa estable de detección después de 7–9 meses de muestreos. Las tasas de detección fueron similares en bosque primario y secundario, sin embargo, se encontraron algunas diferencias en la dinámica de grupo (el radio de distribución del grupo era más grande y distancia de observación fue mayor en bosques primarios, mientras que el número de individuos solitarios fue mayor en bosques secundarios). Más sainos fueron vistos alrededor de las zonas abiertas rodeando el laboratorio, debido a varias posibles razones: habituación a la presencia de seres humanos, menos presión por depredación o cacería y otros factores ambientales o de hábitat. Los sainos están distribuidos de forma aglomerados y son más activos de día que de noche. Las densidades de sainos son relativamente altas en comparación con otros sitios neotropicales, con excepción de la Isla de Barro Colorado. El saino probablemente ha aumentado en abundancia en La Selva, aparentemente unos años después de la extirpación del cariblanco (Tayassu pecari), que eran abundantes en el área hace unos 40–50 años. El conocimiento de la dinámica de grupos, comportamiento y preferencias de hábitat del saino es esencial para las decisiones de manejo y los esfuerzos de conservación. Además, la evaluación de los cambios poblacionales debe considerarse cuidadosamente en un contexto histórico, con especial atención a cómo han cambiado las poblaciones del saino y cariblanco, y cómo estas especies afectan su ambiente

    Continuous single cell imaging reveals sequential steps of plasmacytoid dendritic cell development from common dendritic cell progenitors

    Functionally distinct plasmacytoid and conventional dendritic cells (pDC and cDC) shape innate and adaptive immunity. They are derived from common dendritic cell progenitors (CDPs) in the murine bone marrow, which give rise to CD11c(+) MHCII- precursors with early commitment to DC subpopulations. In this study, we dissect pDC development from CDP into an ordered sequence of differentiation events by monitoring the expression of CD11c, MHC class II, Siglec H and CCR9 in CDP cultures by continuous single cell imaging and tracking. Analysis of CDP genealogies revealed a stepwise differentiation of CDPs into pDCs in a part of the CDP colonies. This developmental pathway involved an early CD11c(+) SiglecH(-) pre-DC stage and a Siglec H+ CCR9(low) precursor stage, which was followed rapidly by upregulation of CCR9 indicating final pDC differentiation. In the majority of the remaining CDP pedigrees however the Siglec H+ CCR9(low) precursor state was maintained for several generations. Thus, although a fraction of CDPs transits through precursor stages rapidly to give rise to a first wave of pDCs, the majority of CDP progeny differentiate more slowly and give rise to longer lived precursor cells which are poised to differentiate on demand

    Induction of tolerogenic lung CD4+ T cells by local treatment with a pSTAT-3 and pSTAT-5 inhibitor ameliorated experimental allergic asthma

    Signal transducer and activator of transcription (STAT)-3 inhibitors play an important role in regulating immune responses. Galiellalactone (GL) is a fungal secondary metabolite known to interfere with the binding of phosphorylated signal transducer and activator of transcription (pSTAT)-3 as well of pSTAT-6 dimers to their target DNA in vitro. Intra nasal delivery of 50 μg GL into the lung of naive Balb/c mice induced FoxP3 expression locally and IL-10 production and IL-12p40 in RNA expression in the airways in vivo. In a murine model of allergic asthma, GL significantly suppressed the cardinal features of asthma, such as airway hyperresponsiveness, eosinophilia and mucus production, after sensitization and subsequent challenge with ovalbumin (OVA). These changes resulted in induction of IL-12p70 and IL-10 production by lung CD11c+ dendritic cells (DCs) accompanied by an increase of IL-3 receptor α chain and indoleamine-2,3-dioxygenase expression in these cells. Furthermore, GL inhibited IL-4 production in T-bet-deficient CD4+ T cells and down-regulated the suppressor of cytokine signaling-3 (SOCS-3), also in the absence of STAT-3 in T cells, in the lung in a murine model of asthma. In addition, we found reduced amounts of pSTAT-5 in the lung of GL-treated mice that correlated with decreased release of IL-2 by lung OVA-specific CD4+ T cells after treatment with GL in vitro also in the absence of T-bet. Thus, GL treatment in vivo and in vitro emerges as a novel therapeutic approach for allergic asthma by modulating lung DC phenotype and function resulting in a protective response via CD4+FoxP3+ regulatory T cells locall
