    Dairy buffalo life cycle assessment (LCA) affected by a management choice: the production of wheat crop

    Life cycle assessment (LCA) was performed in dairy buffalo farms representative of Southern Italian farming systems, similar due to several characteristics, with the exception of wheat production. This work evaluated the impacts derived from this management choice, comparing farms with wheat crop (WWC) or not (NWC). In agreement with the literature, economic allocation was chosen as a useful strategy to attribute equivalents to by-products, i.e., culled animals; the same criterion was also adopted to assign pollutants to wheat grain, limited to WWC farms. Environmental impacts in terms of Global Warming Potential (GWP, kg CO2 eq), Acidification Potential (AC, g SO2 eq), Eutrophication Potential (EU, g PO43- eq), Agricultural Land Occupation (ALO, m2 y) and Water Depletion (WD, m3) were estimated. The production of wheat crop significantly affected (p < 0.05) the Agricultural Land Occupation (ALO) category as WWC farms need adequate land. WWC farms could allow a significant reduction in eutrophication (EU) compared to NWC farms (p < 0.05)

    Fosmid library end sequencing reveals a rarely known genome structure of marine shrimp Penaeus monodon

    <p>Abstract</p> <p>Background</p> <p>The black tiger shrimp (<it>Penaeus monodon</it>) is one of the most important aquaculture species in the world, representing the crustacean lineage which possesses the greatest species diversity among marine invertebrates. Yet, we barely know anything about their genomic structure. To understand the organization and evolution of the <it>P. monodon </it>genome, a fosmid library consisting of 288,000 colonies and was constructed, equivalent to 5.3-fold coverage of the 2.17 Gb genome. Approximately 11.1 Mb of fosmid end sequences (FESs) from 20,926 non-redundant reads representing 0.45% of the <it>P. monodon </it>genome were obtained for repetitive and protein-coding sequence analyses.</p> <p>Results</p> <p>We found that microsatellite sequences were highly abundant in the <it>P. monodon </it>genome, comprising 8.3% of the total length. The density and the average length of microsatellites were evidently higher in comparison to those of other taxa. AT-rich microsatellite motifs, especially poly (AT) and poly (AAT), were the most abundant. High abundance of microsatellite sequences were also found in the transcribed regions. Furthermore, <it>via </it>self-BlastN analysis we identified 103 novel repetitive element families which were categorized into four groups, <it>i.e</it>., 33 WSSV-like repeats, 14 retrotransposons, 5 gene-like repeats, and 51 unannotated repeats. Overall, various types of repeats comprise 51.18% of the <it>P. monodon </it>genome in length. Approximately 7.4% of the FESs contained protein-coding sequences, and the Inhibitor of Apoptosis Protein (IAP) gene and the Innexin 3 gene homologues appear to be present in high abundance in the <it>P. monodon </it>genome.</p> <p>Conclusions</p> <p>The redundancy of various repeat types in the <it>P. monodon </it>genome illustrates its highly repetitive nature. In particular, long and dense microsatellite sequences as well as abundant WSSV-like sequences highlight the uniqueness of genome organization of penaeid shrimp from those of other taxa. These results provide substantial improvement to our current knowledge not only for shrimp but also for marine crustaceans of large genome size.</p

    Application of NIR spectroscopy and image analysis for the characterisation of grated Parmigiano-Reggiano cheese

    Grated Parmigiano–Reggiano cheese holds a valuable market segment and its quality strictly depends on the amount of rind, size, shape of cheese particles and original cheese properties. Textural properties of the rind and inner part of the cheese significantly affect size and shape of grated particles. Rind produces a higher amount of finer and less circular particles than the inner region. Rind content established by European regulation (maximum 18%) is a major issue and could be successfully predicted by multivariate models developed on near-infrared (NIR) spectra. Image analysis (IA) was a suitable method to estimate rind percentage that was found positively correlated to number of particles, total surface covered by particles and circularity. IA and NIR spectroscopy enabled characterisation of the distribution of the particle in dimensional classes and could be used to control the maximum limit of 25% of particles finer than 0.5 mm provided by European regulation

    Dairy farms and life cycle assessment (Lca): The allocation criterion useful to estimate undesirable products

    In this study, the life cycle assessment (LCA) principle was performed to estimate the environmental impact of three dairy farms that operate using different farming systems, namely, conventional (CON), organic (ORG), and high-quality (HQ) modes. In Italy, the typical style of high-quality (HQ) farming is commonly included in the conventional system but is more strictly regulated by the Decree of the Italian Ministry of Health N◦ 185/1991. Although the farms are not fully representative of each conduct, they showed intrinsic peculiarities, e.g., the cow-culling rate of each system. This rate requires a quantification as it may be related to loss of income. Allocation criteria were applied to attribute the quantities of pollutants to the co-products: wheat, involved in the congruence and number of cows culled, the latter being undesirable and therefore necessary to quantify. Analysis of variance (ANOVA) highlighted that the no-dairy products significantly mitigated (p &lt; 0.05) some of the impacts’ categories. The allocation of culled cows decreased the impacts of the CON and particularly those of the ORG farms when the mass mode was adopted. HQ showed values similar to the results without allocation. Overall, the objective of identifying a “marker” of undesirable products, estimated by the culling rate, was partially achieved

    Effect of fermentation-produced camel chymosin on quality of Crescenza cheese

    Quality properties of Crescenza cheese, a typical Italian soft cheese with a short shelf life, are negatively affected by proteolysis due to residual rennet activity, causing a bitter taste and excessive softening of its structure. Chemical composition, textural, rheological and sensory properties of Crescenza cheese manufactured with a fermentation-produced camel chymosin (FPCC) were compared with Crescenza cheese obtained with calf rennet during a 3-week shelf life period. Both Crescenza cheeses complied with the overall expected cheese quality. FPCC Crescenza cheese was characterised by a slightly higher moisture content and lower rate of hydrolysis of αS1-casein than Crescenza cheese obtained with calf rennet. However, FPCC cheese showed a slightly firmer body. The use of FPCC as clotting enzyme may be a valid alternative to calf rennet to prevent the defect of excessive cheese softening

    The impact of sodium chloride reduction on Grana-type cheese production and quality

    With the aim to reduce the Na content, hard cheeses manufactured using the same technology as for Grana cheese (Grana-type) were salted using three brines containing different amounts of KCl (K-brines) and compared with control cheeses, salted with marine NaCl. A lower weight loss was observed in cheeses salted with K-brines (K-cheeses), whereas the yield and dry matter did not differ significantly between K-cheeses and controls. After 3 months of ripening (T3), the distribution of the Na cations (Na) was centripetal, with a higher Na concentration in the outer (0-3 cm of depth) layer, whereas the K cations (K) seemed to diffuse into the cheese more rapidly and homogeneously. Starting from the 6th month (T6), the distribution of both Na and K was stabilized through the different cheese layers. The use of the brine with the highest concentration of potassium (53.8% K) enabled us to successfully halve the Na content compared to the controls whereas, with the other brines, the reduction of Na was below 30%. At the end of ripening (T9), all the cheeses were without defects and the partial substitution of Na with K did not impact on the chemical composition, microbiological characteristics and ripening process. The sensory evaluation did not show any difference between K-salted and control cheeses in discriminant analysis
