Congenital isolated adrenocorticotropic hormone (ACTH) deficiency (CIAD) is a rare disorder which can result in 20% mortality in the neonatal period if misdiagnosed. A 2 years and 7 months old boy was hospitalized many times because of recurrent hypoglycemia. On initial physical examination, the patient showed special appearance and indications of fast growth (≥P97). Laboratory investigations revealed low levels of ACTH and cortisol in his plasma. Except thyroid-stimulating hormone, the anterior pituitary hormone concentrations were normal. Molecular data showed compound heterozygosity for two novel mutations in the TBX19 gene (encoding the transcription factor T-Box 19). Mutation c.205C>T was inherited from mother and the fragment deletion (from g.168,247,374 to g.168,278,264) was from father. Hydrocortisone replacement therapy was effective. We reported two novel TBX19 mutations, expanding the mutation spectrum of this disorder, in a CIAD patient who presented with special appearance, signs of fast growth, and thyroid-stimulating hormone derangement. In addition, for avoiding misdiagnosis, criterion for ACTH and cortisol detection of CIAD should be established

Visual-based target tracking is one of the critical methodologies for the control problem of multi-robot systems. In dynamic mobile environments, it is common to lose the tracking targets due to partial visual occlusion. Technologies based on deep learning (DL) provide a natural solution to this problem. DL-based methods require less human intervention and fine-tuning. The framework has flexibility to be retrained with customized data sets. It can handle massive amounts of available video data in the target tracking system. This paper discusses the challenges of robot tracking under partial occlusion and compares the system performance of recent DL models used for tracking, namely you-only-look-once (YOLO-v5), Faster region proposal network (R-CNN) and single shot multibox detector (SSD). A series of experiments are committed to helping solve specific industrial problems. Four data sets are that cover various occlusion statuses are generated. Performance metrics of F1 score, precision, recall, and training time are analyzed under different application scenarios and parameter settings. Based on the metrics mentioned above, a comparative metric P is devised to further compare the overall performance of the three DL models. The SSD model obtained the highest P score, which was 13.34 times that of the Faster RCNN model and was 3.39 times that of the YOLOv5 model with the designed testing data set 1. The SSD model obtained the highest P scores, which was 11.77 times that of the Faster RCNN model and was 2.43 times that of the YOLOv5 model with the designed testing data set 2. The analysis reveals different characteristics of the three DL models. Recommendations are made to help future researchers to select the most suitable DL model and apply it properly in a system design.</jats:p

Microfluidic fabrication of polymer-based microparticles for biomedical applications

Delivery vehicles that can encapsulate and release active ingredients of pre-determined volumes at the target site on-demand present a challenge in biomedical field. Due to their tunable physiochemical properties and degradation rate, polymeric particles are one of the most extensively employed delivery vehicles. Generally they are fabricated from emulsion templates. Conventional bulk emulsification technique provides little control over the characteristics of droplets generated. Thus the properties of the subsequent particles cannot be controlled. The advance of droplet microfluidics enables the generation and manipulation of designer single, double or higher-order emulsion droplets with customizable structure. These droplets are powerful and versatile templates for fabricating polymeric delivery vehicles with pre-determined properties. Due to the monodispersity of droplet templates by microfluidics, the relationship between size, size distribution, shape, architecture, elastic responses and release kinetics can be systematically studied. These understandings are of key importance for the design and fabrication of the next generation polymeric delivery vehicles with custom-made functions for specific applications. In the present work, we engineer the droplet templates generated from microfluidics to fabricate designer polymeric microparticles as delivery vehicles. We investigate and obtain the relationship between the particle size, size distribution, structure of microparticles and their release kinetics. Moreover, we also identify an innovative route to tune the particle shape that enables the investigation of the relationship between particle shape and release kinetics. We take advantage of the dewetting phenomena driving by interfacial tensions of different liquid phases to vary the droplet shape. We find that the phase-separation-induced shape variation of polymeric composite particles can be engineered by manipulating the kinetic barriers during droplet shape evolution. To predict the performance of our advanced polymer particles in practical applications, for instance, in narrow blood vessels in vivo, we also develop a novel capillary micromechanics technique to characterize the linear and non-linear elastic response of our polymer particles on single particle level. The knowledge of the mechanical properties enables the prediction as well as the design of the mechanical aspects of polymer particles in different applications. The ability to control and design the physical, chemical, mechanical properties of the delivery vehicles, and the understanding between these properties and the biological functionalities of delivery vehicles, such as the release kinetics, lead towards tailor-designed delivery vehicles with finely-designed functionalities for various biomedical applications. Our proposed electro-microfluidic platform potentially enables generation of submicron droplet templates with a narrow size distribution and nanoscaled delivery vehicles with well-controlled properties, leading to a next generation of intracellular delivery vehicles. Microfluidic-based technique has the potential to be scaled up by parallel operation. Therefore, we are well-equipped for the massive production of custom-made droplet templates of both micron-size and nanosized, and we can design the physiochemical properties and biological functionalities of the delivery vehicles. These abilities enable us to provide solutions for applications and fundamental topics where encapsulation, preservation and transportation of active ingredients are needed.published_or_final_versionMechanical EngineeringDoctoralDoctor of Philosoph

Recent advances in liver‐on‐chips: Design, fabrication, and applications

    No full text
Abstract The liver is a multifunctional organ and the metabolic center of the human body. Most drugs and toxins are metabolized in the liver, resulting in varying degrees of hepatotoxicity. The damage of liver will seriously affect human health, so it is very important to study the prevention and treatment of liver diseases. At present, there are many research studies in this field. However, most of them are based on animal models, which are limited by the time‐consuming processes and species difference between human and animals. In recent years, liver‐on‐chips have emerged and developed rapidly and are expected to replace animal models. Liver‐on‐chips refer to the use of a small number of liver cells on the chips to simulate the liver microenvironment and ultrastructure in vivo. They hold extensive applications in multiple fields by reproducing the unique physiological functions of the liver in vitro. In this review, we first introduced the physiology and pathology of liver and then described the cell system of liver‐on‐chips, the chip‐based liver models, and the applications of liver‐on‐chips in liver transplantation, drug screening, and metabolic evaluation. Finally, we discussed the currently encountered challenges and future trends in liver‐on‐chips

    No full text
With the rapid development of China's economy and the continuous increase in economic volume, pollution hasn’t been limited to cities. The rural “scattered and polluted” enterprises, as the main source of pollution in the rural environment, affect the rural ecological environment and restrict the green and healthy development of the rural economy. Since April 2017, the Tianjin Municipal Government has issued a series of documents on the governance of “scattered and polluted” enterprises in rural areas, which has adopted targeted countermeasures and achieved obvious results, but there are also some problems. How to scientifically and effectively govern the rural “scattered and polluted” enterprises needs continuous exploration, summary and improvement
